PIV MEASUREMENTS OF TURBULENT FLOW IN A CHANNEL WITH SOLID OR PERFORATED RIBS

- Particle image velocimetry measurements are performed in a channel with periodic ribs on one wall. We investigate the flow around two different rib configurations: solid and perforated ribs with a slit. The ribs obstruct the channel by 20% of its height and are arranged 10 rib heights apart. For the perforated ribs, the slit height is 20% of the rib height, and the open-area ratio is 16%. We discuss the flow in terms of mean velocity, streamlines, vorticity, turbulence intensity, and Reynolds shear stress. We find that the recirculation bubbles after the perforated ribs are significantly smaller than those after the solid ribs. The reattachment length after perforated ribs is smaller by about 45% compared with the solid ribs. In addition, the Reynolds shear stresses around the perforated ribs are significantly smaller than in the solid rib case, leading to a reduction of the pressure loss in the perforated rib case.

- This paper presents an integrated approach for developing a web-based system with enhanced adaptability, including distributed process planning, real-time monitoring and remote machining. The objective is to develop a new methodology and relevant processing algorithms for enhancing adaptability in digital manufacturing. This approach is enabled by a Wise-ShopFloor (Web-based integrated sensor-driven e-ShopFloor) framework targeting distributed yet collaborative manufacturing environments. Utilising the latest Java technologies (Java 3D and Java Servlet) for system implementation, it allows end-users to plan and control distant manufacturing operations based on runtime information from shop floors. Details on the principle of the Wise-ShopFloor framework, system architecture, and a prototype system are reported in this paper. An example of distributed process planning for remote machining is chosen as a case study to demonstrate the effectiveness of this approach toward web-based digital manufacturing.

- Highly turbulent environment of dynamic job-shop operations affects shop-floor layout as well as manufacturing operations. Due to the dynamic nature of layout changes, essential requirements such as adaptability and responsiveness to the changes need to be considered in addition to the cost issues of material handling and machine relocation when reconfiguring a shop floor’s layout. Here, based on the source of uncertainty, the shop-floor layout problem is split into two sub-problems and dealt with by two modules: re-layout and find-route. Genetic algorithm is used where changes cause the entire shop re-layout, while function blocks are utilized to find the best sequence of robots for the new conditions within the existing layout. This chapter reports the latest development to the authors’ previous work.

Resistance exercise enhances the molecular signaling of mitochondrial biogenesis induced by endurance exercise in human skeletal muscle.

- Combining endurance and strength training (concurrent training) may change the adaptation compared with single mode training. However, the site of interaction and the mechanisms are unclear. We have investigated the hypothesis that molecular signaling of mitochondrial biogenesis after endurance exercise is impaired by resistance exercise. Ten healthy subjects performed either only endurance exercise (E: 1h cycling at ~65% of VO(2max)) or endurance exercise followed by resistance exercise (ER: 1h cycling + 6 sets of leg press at 70-80% of 1 repetition maximum) in a randomized cross-over design. Muscle biopsies were obtained before and after exercise (1 and 3h Post cycling). The mRNA of genes related to mitochondrial biogenesis (PGC-1α, PRC) and substrate regulation (PDK4) increased after both E and ER, but the mRNA levels were about 2-fold higher after ER (P<0.01). Phosphorylation of proteins involved in the signaling cascade of protein synthesis (mTOR, S6K1 and eEF2) was altered after ER but not after E. Moreover, ER induced a larger increase in mRNA of genes associated with positive mTOR signaling (cMyc and Rheb). Phosphorylation of AMPK, ACC and Akt increased similarly at 1h Post (P<0.01) after both types of exercise. Contrary to our hypothesis, the results demonstrate that resistance exercise, performed after endurance exercise, amplifies the adaptive signaling response of mitochondrial biogenesis compared with single-mode endurance exercise. The mechanism may relate to a crosstalk between signaling pathways mediated by mTOR. The results suggest that concurrent training may be beneficial for the adaptation of muscle oxidative capacity.

- The recent research by a group of European scientists shows that, each starling bird adjusts its flight in the light of topological interaction, that according to a fixed number of its nearest neighbors, rather than all agents within a fixed Euclidean distance. A theoretical study to understand such topological interaction is given in this paper, and a sufficient synchronization condition imposing only on the initial states is developed. Furthermore, the number of neighbors needed for the synchronization is discussed, which shows that for the sake of connectivity the number of neighbors should increase as the density of group increasing. - SUCLA2-related mitochondria! DNA (mtDNA) depletion syndrome is a result of mutations in the beta subunit of the ADP-dependent isoform of the Krebs cycle succinyl-CoA synthase (SCS). The mechanism of tissue specificity and mtDNA depletion is elusive but complementation by the GDP-dependent isoform encoded by SUCLG2, and the association with mitochondrial nucleoside diphosphate kinase (NDPK), is a plausible link. We have investigated this relationship by studying SUCLA2 deficient fibroblasts derived from patients and detected normal mtDNA content and normal NDPK activity. However, knockdown of SUCLG2 by shRNA in both patient and control fibroblasts resulted in a significant decrease in mtDNA amount, decreased NDPK and cytochrome c oxidase activities, and a marked growth impairment. This suggests that, SUCLG2, to a higher degree than SUCLA2, is crucial for mtDNA maintenance and that mitochondrial NDPK is involved. Although results pertain to a cell culture system, the findings might explain the pathomechanism and tissue specificity in mtDNA depletion caused by defective SUCLA2. (C) 2011 Elsevier B.V.
