    • OBJECTIVE: MEIS1, a HOX cofactor, collaborates with multiple HOX and NUP98-HOX fusion proteins to accelerate the onset of acute myeloid leukemia (AML) through largely unknown molecular mechanisms. MATERIALS AND METHODS: To further resolve these mechanisms, we conducted a structure-function analysis of MEIS1 and gene-expression profiling, in the context of NUP98-HOXD13 (ND13) leukemogenesis. RESULTS: We show, in a murine bone marrow transplantation model, that the PBX-interaction domain, the homeodomain, and the C-terminal domain of MEIS1, are all required for leukemogenic collaboration with ND13. In contrast, the N-terminal domain of MEIS1 is dispensable for collaboration with ND13, but is required for Flt3 upregulation, indicating additional roles for MEIS1 in induction of leukemia independent of alterations in Flt3 expression. Gene-expression profiling of a cloned ND13 preleukemic cell line transduced with wild-type or Meis1 mutant forms revealed deregulation of multiple genes, including a set not previously implicated as MEIS1 targets. Chromatin immunoprecipitation revealed the in vivo occupancy of MEIS1 on regulatory sequences of Trib2, Flt3, Dlk1, Ccl3, Ccl4, Pf4, and Rgs1. Furthermore, engineered overexpression of Trib2 complements ND13 to induce AML while Ccl3 potentiates the repopulating ability of ND13. CONCLUSION: This study shows that Meis1-induced leukemogenesis with ND13 can occur in the absence of Flt3 upregulation and reveals the existence of other pathways activated by MEIS1 to promote leukemia.

  • Enhancing the Sustainability of Electronic Access to ELPUB Proceedings : Means for Long-term Dissemination

    • ELPUB can look back on a track record of a steadily growing number of conference papers. From a long-term perspective, access to this body of knowledge is of great interest to the community. Beyond this, extended preoccupation with the collected scientific work in the area of digital publishing has to be mentioned. Naturally, the authors are particularly focussed on the individual paper itself and possible connections with related efforts. Typically, conferences amplify and enhance opportunities of “getting- together”. A well-stocked repository may, however, serve in this respect as a fruitful complementary addition. In this contribution, the implementation of persistent identifiers on the existing ELPUB.scix.net-base is elaborated in detail. Furthermore, the authors present the result of efforts related to the harvesting of ELPUB-metadata and to the creation of a citation index. The paper concludes with an outlook on future plans.

    • The purpose of this work is the investigation of some constitutive and configurational aspects of phenomenological model formulations for a class of materials with history-dependent gradient microstructure. The assumption that the behavior of a material point is affected by history-dependent processes in a finite neighbor of this point yields an extended continuum characterized by non-simple material behavior and by additional degrees-of-freedom. This includes both standard micromorphic materials as well as inelastic gradient materials as special cases. As in the case of simple materials, the corresponding constitutive relations are subject to restrictions imposed by material frame-indifference and material symmetry. In the latter case, both direct and differential restrictions are obtained in the case of assuming that the free energy density is an isotropic function of its arguments. In addtion, the concept of material isomorphism is shown to extend to inelastic gradient continua, resulting in a gradient generalization of the well-known elastoplastic multiplicative decomposition of the deformation gradient. Finally, we examine the consequences of gradient extension for the formulation of configurational field and balance relations, and in particular for the Eshelby stress. This is carried out with the help of an incremental stress potential formulation as based on a continuum thermodynamic approach to the coupled field problem involved.

    • Social costs are often important in promoting the honesty of sexually selected traits. What happens, then, when social costs are relaxed? In species that breed in shallow coastal waters, increases in the frequency and severity of phytoplankton blooms may undermine the value of visual signals by reducing visibility and, in so doing, lead to dishonest signaling by relaxing the social consequences of high signaling effort for poor-quality individuals. Here, we experimentally test the effects of algally induced water turbidity on the role of male-male competition in facilitating reliable sexual displays in three-spined sticklebacks. We found that males in poor condition reduced their courtship effort in the presence of competition in turbid water. This reduction, however, was to a much lesser extent than that observed in clear water. Thus, courtship under conditions of algal turbidity did not reflect male condition as honestly as courtship in clear water. Algal turbidity also influenced breeding coloration, with males in poor condition reducing their area of red nuptial coloration in turbid conditions. Our findings suggest that anthropogenic disturbance to the signaling environment can potentially reduce the evolutionary potential of sexual selection by diminishing the efficacy of visual displays and weakening socially enforced signals of male quality.
