    • With the ability to resist biodegradation and exert therapeutic effects at low concentrations, emerging contaminants have become environmental stressors for wildlife. One such contaminant is the anxiolytic oxazepam, a psychoactive pharmaceutical which is frequently detected in surface waters globally. Despite the growing interest in understanding how wildlife responds to such contaminants, the synergistic fitness effects of pharmaceuticals and increased variability in temperature remain unclear. Here, by using a multi-stressor approach, we investigated the effects of 7-d oxazepam exposure (6.5 μg/L) on anxiety-related behaviours in juvenile European perch (Perca fluviatilis). The multi-stressor approach was achieved by exposing perch to oxazepam at either low (10°C) or high (18°C) temperature, with or without a predation cue, generating 8 treatments. Our exposures resulted in a successful uptake of the drug from the water, i.e. oxazepam was measured at muscle tissue concentrations around 50 ± 17 ng/g (mean ± SD). We found significant effects on boldness induced by the studied drug: 92.8% of the fish in the 'oxazepam and predation and high temperature' treatment entered the white background (representing a novel area where exposure to presumed risks are higher) within the first 5 min, compared to 79.3% of the 'control and predation and high temperature' fish. We also found a significant effect on temperature on the total time freezing (i.e. staying motionless). Specifically, fish in the low temperature treatments (oxazepam, predation and control) froze for longer than fish in the high temperatures, respectively. Our study is the first to show altered anxiety-related behaviours in a native juvenile fish resulting from oxazepam, predation and high temperature. As adaptation to a range of biotic and abiotic pressures is essential to living organisms, our study highlights the need to focus on multiple stressors to improve understanding of how organisms not only survive, but adapt to human-induced environmental change.

  • Identification of bases required for P2 Integrase core-binding and recombination

    • Temperate coliphage P2 integrates its genome into the host chromosome upon lysogenization via a site-specific recombination event mediated by an integrase belonging to the complex family of tyrosine recombinases. The host integration site attB (BOB´) is localized in the end of the cyaR gene and share 27 nucleotides with the core of attP (COC´). In the present study we determine the minimal attB site using an in vivo recombination assay and 10 nt on the left side (B) are found to be nonessential for recombination. We show that the integrase has higher affinity for the right side (B´) compared to B and that artificial B´OB´ and an attP site with a matching core (C´OC´) are efficient substrates for recombination in vitro. We have analyzed single nucleotides in attB and find that sequence homology within a non-centrally located quadruplet in the hypotetical overlap region is essential for efficient recombination in vivo.

  • "Cheaper and better" : an economic analysis of changing first line treatment for cutaneous leishmaniasis in Bolivia

    • Introduction: Cutaneous leishmaniasis (CL) is endemic in Bolivia, mostly affecting poor people in rainforest areas. The current first-line treatment consists of systemic pentavalent antimonials (SPA) for 20 days and is paid for by the Ministry of Health (MoH). Long periods of drug shortages, a lack of conditions to deliver treatment safely, treatment interruption are challenges to implementation. Intralesional pentavalent antimonials (ILPA) are an alternative to SPA. This study aims to compare the cost of ILPA and SPA, and to estimate the health and economic impacts of changing the first-line treatment for CL in an endemic area of Bolivia.Methods: The cost per patient treated was estimated for SPA and ILPA from the perspectives of the MoH and society. The quantity and unit costs of medications, staff time, transportation and loss of production were obtained through a health facility survey (N=12), official documents and key informants. A one-way sensitivity analysis was conducted on key parameters to evaluate the robustness of the results. The annual number of patients treated and the budget impact of switching to ILPA as the first-line treatment were estimated under different scenarios of increasing treatment utilization using previous estimates of the extent of underreporting. Costs were reported in 2016 international dollars (1 INT$ = 3.10 BOB).Results: Treating CL using ILPA was associated with a cost saving of $248 per patient treated from the MoH perspective, and $688 per patient treated from the societal perspective. ILPA was cost-saving even under a hypothetical increase of 80% in the number of cases treated. Switching first-line treatment would allow two-and-a-half times the current number of patients to be treated, while maintaining the current budget.Conclusions: The results of this study support a shift to ILPA as the first-line treatment for CL in Bolivia and possibly in other South American countries.
