  • Microbial communities and Food safety aspects of crickets (Acheta domestica) reared under controlled conditions

    • In an approach combining microbiological culture methods with high-throughput sequencing, this study investigated the microbial communities (bacteria, moulds and yeasts) in Swedish-produced edible crickets (Acheta domesticus) reared in a controlled environment. The effects of different feeds on microbial loads and populations in crickets were also studied. The crickets used were third-generation offspring from wild-caught individuals from Sweden, which are adapted to grow in a laboratory environment. The efficiency of rinsing to decrease microbial load was evaluated not obtaining a significant decrease of plating counts for total aerobic counts (TAC) and Enterobacteriaceae. Crickets were divided into three batches and fed different diets (control feed, early-cut red clover hay (ECH), late-cut fresh red clover (LCF)) for 62 days. Bacterial numbers (TAC and Enterobacteriaceae) on whole raw crickets ranged between 7 and 8 log cfu/g. Pre-rinsing in water did not reduce these levels (P=0.19). All batches tested negative for the food-borne bacteria Salmonella, Listeria monocytogenes, Bacillus cereus and Clostridium perfringens. The mean mould count for crickets fed control feed was 2.8 log cfu/g, while the values for crickets fed ECH and LCF were 4.2 and 4.5 log cfu/g, respectively. The dominant bacterial communities were Proteobacteria, Bacteroidetes and Firmicutes, with Firmicutes and Proteobacteria dominating in crickets fed control feed, Firmicutes dominating in crickets fed LCF and Proteobacteria dominating in crickets fed ECH. Aspergillus flavus, a fungus that is capable of producing mycotoxins, was detected in control feed and ECH reared crickets. More work is needed to identify specific food-borne pathogens in edible crickets and establish possible bacterial quality reference values, as an important step in developing microbial quality and safety parameters to ensure consumer safety.

  • Short-Term Interaction between Dogs and Their Owners : Effects on Oxytocin, Cortisol, Insulin and Heart Rate-An Exploratory Study

    • The aim of this exploratory study was to determine heart rate and the levels of oxytocin, cortisol, and insulin in dogs and their owners in response to a short-term interaction. In addition, the dogs' behavior was studied. The owners' responses were compared with those obtained from a control group. Ten female volunteers and their own male Labrador dogs participated in an experiment during which the owner stroked, petted, and talked with her dog during the first 3 minutes. Blood samples were collected from both dog and owner before (0) and at 1, 3, 5, 15, 30, and 60 minutes after the start of the interaction. Blood samples were analyzed by EIA. Heart rate was monitored telemetrically. The data were analyzed using linear mixed models and paired t-tests. The dogs' oxytocin levels were significantly increased 3 minutes after the start of the interaction (p = 0.027). Cortisol levels were significantly increased after 15 and 30 minutes (p = 0.004 and p = 0.022, respectively), and heart rate was significantly decreased after 55 minutes (p = 0.008). The dogs displayed normal behaviors during the experiment. The owners' oxylocin levels peaked between 1 and 5 minutes after interaction (p = 0.026). No such effect was seen in the controls. Cortisol levels displayed a significant decrease at 15 or 30 minutes in both owners and controls, and insulin levels did so at 60 minutes (p = 0.030, p = 0.002 and p = 0.002, p < 0.0001, respectively). Heart rate decreased significantly in the owners at 55 and 60 minutes (p = 0.0008) but not in the controls. In conclusion, short-term sensory interaction between dogs and their owners influences hormonal levels and heart rate. However, further studies need to be performed in order to better understand the effects of interaction between dogs and their owners.

  • The effect of rider weight and additional weight in Icelandic horses in tölt: part I. Physiological responses

    • This study examined the effect of increasing BW ratio (BWR) between rider and horse, in the BWR range common for Icelandic horses (20% to 35%), on heart rate (HR), plasma lactate concentration (Lac), BWR at Lac 4 mmol/l (W-4), breathing frequency (BF), rectal temperature (RT) and hematocrit (Hct) in Icelandic horses. In total, eight experienced school-horses were used in an incremental exercise test performed outdoors on an oval riding track and one rider rode all horses. The exercise test consisted of five phases (each 642 m) in tolt, a four-beat symmetrical gait, at a speed of 5.4 +/- 0.1 m/s (mean +/- SD), where BWR between rider (including saddle) and horse started at 20% (BWR20), was increased to 25% (BWR25), 30% (BWR30), and 35% (BWR35) and finally decreased to 20% (BWR20b). Between phases, the horses were stopped (similar to 5.5 min) to add lead weights to specially adjusted saddle bags and a vest on the rider. Heart rate was measured during warm-up, the exercise test and after 5, 15 and 30 min of recovery and blood samples were taken and BF recorded at rest, and at end of each of these aforementioned occasions. Rectal temperature was measured at rest, at end of the exercise test and after a 30-min recovery period. Body size and body condition score (BCS) were registered and a clinical examination performed on the day before the test and for 2 days after. Heart rate and BF increased linearly (P < 0.05) and Lac exponentially (P < 0.05) with increasing BWR. The W-4 was 22.7 +/- 4.3% (individual range 17.0% to 27.5%). There was a positive correlation between back BCS and W-4 (r = 0.75; P = 0.032), but no other correlations between body measurements and W-4 were found. Hematocrit was not affected by BWR (P>0.05), but negative correlations (P < 0.05) existed between body size measurements and Hct. While HR, Hct and BF recovered to values at rest within 30 min, Lac and RT did not. All horses had no clinical remarks on palpation and at walk 1 and 2 days after the test. In conclusion, increasing BWR from 20% to 35% resulted in increased HR, Lac, RT and BF responses in the test group of experienced adult Icelandic riding horses. The horses mainly worked aerobically until BWR reached 22.7%, but considerable individual differences (17.0% to 27.5%) existed that were not linked to horse size, but to back BCS.

    • This study investigated the effects of rider weight in the BW ratio (BWR) range common for Icelandic horses (20% to 35%), on stride parameters in tolt in Icelandic horses. The kinematics of eight experienced Icelandic school horses were measured during an incremental exercise test using a high-speed camera (300 frames/s). Each horse performed five phases (642 m each) in tolt at a BWR between rider (including saddle) and horse starting at 20% (BWR20) and increasing to 25% (BWR25), 30% (BWR30), 35% (BWR35) and finally 20% (BWR20b) was repeated. One professional rider rode all horses and weight (lead) was added to saddle and rider as needed. For each phase, eight strides at speed of 5.5 m/s were analyzed for stride duration, stride frequency, stride length, duty factor (DF), lateral advanced placement, lateral advanced liftoff, unipedal support (UPS), bipedal support (BPS) and height of front leg action. Stride length became shorter (Y = 2.73-0.004x; P < 0.01) and more frequent (Y = 2.56+0.002x; P < 0.001) with added weight. Duty factor and BPS increased with increased BWR (P < 0.001), whereas UPS decreased (P < 0.001). Lateral advanced timing of limb placement and liftoff and height of front leg action were not affected by BWR (P > 0.05). In conclusion, increased BWR decreased stride length and increased DF proportionally to the same extent in all limbs, whereas BPS increased at the expense of decreased UPS. These changes can be expected to decrease tolt quality when subjectively evaluated according to the breeding goals for the Icelandic horse. However, beat, symmetry and height of front leg lifting were not affected by BWR.

  • Growth, training response and health in Standardbred yearlings fed a forage-only diet

    • The aim of this study was to, from a holistic perspective, describe the effects of a forage-only feeding system and a conventional training program on young Standardbred horses and compare data with similar observations from the literature. Sixteen Standardbred colts fed a forage-only diet for 4 months from breaking (August to December) and with the goal to vigorously trot 5 to 7 km at a speed of 5.6 m/s (3 min/km) were studied. The horses were fed grass haylage (56 to 61% dry matter (DM), 2.80 to 3.02 Mcal DE/kg DM and 130 to 152 g CP/kg DM) ad libitum, 1 kg of a lucerne product and minerals. The amount of training and number of training sessions were documented daily, and feed intake and body development were measured once every month. Heart rate (HR) was measured during and after a standardized exercise test in October and December. In December, a postexercise venous blood sample was collected and analyzed for plasma lactate concentration. Muscle biopsies (m. gluteus medius) were taken and analyzed for glycogen and fiber composition. Health was assessed in October and November by an independent veterinarian using a standardized health scoring protocol. BW and height at withers increased from 402 to 453 kg (root mean square error (RMSE) 6) and from 148.7 to 154.1 cm (RMSE 0.7), respectively, and the body condition score was 4.9 (RMSE 0.2) at the end of the study. Muscle glycogen content was 532 mmol/kg dry weight (s.d. 56). There was a significant decrease in postexercise HR (81 v. 73 bpm, RMSE 8), and the individual amount of training was negatively correlated with HR during and after exercise. Health scores were high and similar at both assessments (8.4 and 8.4 (RMSE 1.0) out of 10; P > 0.05), and the number of lost training days per month due to health problems was <0.9, with the exception of November (5.3 days). It is concluded that yearlings in training fed high-energy forage ad libitum can reach a conventional training goal and grow at least as well as earlier observations on yearlings of other light breeds.

    • The aim of the present study was to investigate feed allowances, body weight (BW), haematocrit, haemoglobin, plasma ionised calcium (iCa), sodium, potassium, pH and glucose concentration and faecal K/Na ratio in local growing pigs, sows and piglets kept by small-holder farmers in Laos. Starting hypotheses were that (1) local pigs are under fed, (2) BW is higher in pigs receiving supplementary feed and (3) the blood profile of pigs subjected to very poor nutrition is outside the normal range. On 54 pig-keeping smallholdings in Borikhamxay province, Lao PDR, daily feed allowances were weighed and BW recorded for 27 lactating sows, 54 piglets and 27 growing pigs. Blood samples were collected from the vena jugularis in all pigs. Feed supplementation did not affect BW, but plasma iCa concentration was outside the normal range for all pigs. There was a tendency for lower faecal K/Na ratio in Na-supplemented sows. The results confirm that local pigs in small-scale production systems in Laos suffer from poor nutrition. The most important challenge for farmers appears to be provision of more feed, particularly feed with a high Ca content.

  • A forage-only diet alters the metabolic response of horses in training

    • Most athletic horses are fed a high-starch diet despite the risk of health problems. Replacing starch concentrate with high-energy forage would alleviate these health problems, but could result in a shift in major substrates for muscle energy supply from glucose to short-chain fatty acids (SCFA) due to more hindgut fermentation of fibre. Dietary fat inclusion has previously been shown to promote aerobic energy supply during exercise, but the contribution of SCFA to exercise metabolism has received little attention. This study compared metabolic response with exercise and lactate threshold (V-La4) in horses fed a forage-only diet (F) and a more traditional high-starch, low-energy forage diet (forage-concentrate diet - FC). The hypothesis was that diet F would increase plasma acetate concentration and increase V-La4 compared with diet FC. Six Standardbred geldings in race training were used in a 29-day change-over experiment. Plasma acetate, non-esterified fatty acids (NEFA), lactate, glucose and insulin concentrations and venous pH were measured in samples collected before, during and after a treadmill exercise test (ET, day 25) and muscle glycogen concentrations before and after ET. Plasma acetate concentration was higher before and after exercise in horses on diet F compared with diet FC, and there was a tendency (P = 50.09) for increased V-La4 on diet F. Venous pH and plasma glucose concentrations during exercise were higher in horses on diet F than diet FC, as was plasma NEFA on the day after ET. Plasma insulin and muscle glycogen concentrations were lower for diet F, but glycogen utilisation was similar for the two diets. The results show that a high-energy, forage-only diet alters the metabolic response to exercise and, with the exception of lowered glycogen stores, appears to have positive rather than negative effects on performance traits.
