    • The starting point for the study is the idea that many practices rely on specific methods that regularly evade formalized accounts. Some techniques are considered too trivial or idiosyncratic even when providing descriptions of the work intended as instructions. This chapter analyzes such a case taken from endovascular surgery: a form of image-guided intervention that relies on angiography and fluoroscopy imaging modalities. In this practice, surgeons must repeatedly move between different images while still remembering certain visualized features that are now lost from view. Some procedures to overcome this challenge have been outlined in the medical literature. We describe an additional method, which is de facto used in practice while still residing outside of the formally recognized methods of that practice. The technique relies on the creative use of a computer cursor as a visual aid for marking locations in the images. This workaround is built on local practices and technologies and is now an integral part of how the work gets done. It operates as a form of instructed action: a series of activities prospectively oriented to creating a shared phenomenal field by and for the collaborating surgeons.

    • Magnetic resonance imaging (MRI) is considered being state-of-the-art technology for body composition analysis. Compared to other indirect techniques such as scales, calipers, bioimpedance, and dual-energy X-ray absorptiometry (DXA), MRI offers direct and precise measurements of the volumes of different tissue compartments and also enables quantification of diffuse fat infiltration in organs. Here, we describe a protocol for acquiring of fat–water-separated MRI data and the image postprocessing required for the quantification of several body composition biomarkers relevant for metabolic research. This protocol has successfully been used in several clinical studies and also in the large UK Biobank population study.

    • Cardiac magnetic resonance spectroscopy (MRS) opens a window to the metabolism of the heart. Various intermediates of metabolic pathways can be observed and followed over time. Most applications of cardiac MRS have been performed with the 31P nuclei, which reflect the metabolites from the high-energy phosphate metabolism. Other nuclei, such as 1H or 13C, have also been investigated but less intensively, most likely because of either large background signals (e.g., water) or inherent low sensitivity of the method. MRS can be used for the examination of tissue extracts, isolated organs, whole animals in vivo, as well as healthy human subjects and patients. Although the primary motivation is to gain an understanding of metabolism using animal models, a potential for diagnostic applications in humans certainly exists.

    • GPU’s have recently emerged as a significantly more powerful computing plat-form, capable of several orders of magnitude faster computations compared toCPU based approaches. However, they require significant changes in the algorithmic design compared to traditional programming paradigms. In this chapter we specifically introduce the reader to an overview of GPGPU development tools and the potential algorithmic pitfalls and bottlenecks when developing medical imaging algorithms for the GPU. We present a few general methodologies and building blocks for implementing fast image processing on GPUs. More specifically they include: methods for performing fast image convolutions and filtering;line detection, and bandwidth and memory considerations when processing volumetric datasets. Finally we conclude with a discourse on numerical precision as well as on mixing single floating-point versus double floating-point code.

    • In imaging, penetration depth comes at the expense of lateral resolution, which restricts the scope of 3D in-vivo imaging of small animals at micrometer resolution. Bioimaging will need to expand beyond correlative light and electron microscopy (CLEM) approaches to combine insights about in-vivo dynamics in a physiologically relevant 3D environment with ex-vivo information at micrometer resolution (or beyond) within the spatial, structural and biochemical contexts. Our report demonstrates the immense potential for biomedical discovery and diagnosis made available by bridging preclinical in-vivo imaging with ex-vivo biological microscopy to zoom in from the whole organism to individual structures and by adding localized spectroscopic information to structural and functional information. We showcase the use of two novel imaging pipelines to zoom into mural lesions (occlusions/hyperplasia and micro-calcifications) in murine vasculature in a truly correlative manner, that is using exactly the same animal for all integrated imaging modalities. This correlated multimodality imaging (CMI) approach includes well-established technologies such as Positron Emission Tomography (microPET), Autoradiography, Magnetic Resonance Imaging (microMRI) and Computed Tomography (microCT), and imaging approaches that are more novel in the biomedical setting, such as X-Ray Fluorescence Spectroscopy (microXRF) and High Resolution Episcopic Microscopy (HREM). Although the current pipelines are focused on mural lesions, they would also be beneficial in preclinical and clinical investigations of vascular diseases in general.

    • Hyperthermia therapy involves mild heating (40-44°C) that has been shown to be a potent enhancer of radiation and chemotherapy. Microwave (MW) and radiofrequency (RF) hyperthermia devices provide a variety of heating approaches that can treat most cancers regardless of their size and depth. This review introduces the physics of MW/RF hyperthermia, the current state-of-the-art systems for both local and regional heating, and recent advancements in hyperthermia treatment planning and guidance using real-time magnetic resonance imaging. The clinical evidence from clinical trials involving RF/MW hyperthermia as an adjuvant for radiation and chemotherapy is reviewed to show that adequate heating can be obtained with state-of-the-art systems. These studies strongly support the use of adjuvant hyperthermia as it significantly improves curative and palliative clinical outcomes. We anticipate that recent technology advances will improve the treatment delivery and reduce its complexity, as pivotal components for expanding the use of hyperthermia at medical and radiation oncology centers and enhancing cancer therapy.
