  • Rye Bran Alkylresorcinols Inhibit Growth of Penicillium expansum and Neofabraea perennans In Vitro and In Vivo on Different Apple Cultivars

    • Apples (Malus domestica Borkh.) are highly susceptible to fungi causing rots, especially in organic orchards where these rots cause major economic losses. Since the use of conventional fungicides is prohibited or restricted in many countries and in organic production, altogether, one alternative is to make use of bioactive compounds that can inhibit fungal growth on apples. Antifungal effects of alkylresorcinols (ARs), isolated from rye bran, were used in two different bioactive emulsions and tested in vitro as well as in vivo against two fungi, Penicillium expansum and Neofabraea perennans, which cause important storage diseases in apple. For both fungi, application of AR emulsions produced a 50 % reduction in mycelial growth and spore germination. The in vivo tests were carried out on wound-inoculated fruit of four apple cultivars, 'Aroma', 'Ingrid Marie', Frida' and 'Gloster'. Spraying the inoculated fruit with ARs decreased lesion area diameter caused by P. expansum with 17-52 % and lesion area diameter caused by N. perennans with 31-77 %, depending on the apple cultivar. For each fungus, the inhibitory effect of ARs was stronger in those apple cultivars that appear to have the highest level of disease tolerance, i.e. the smallest lesions on the control fruit. The level of sun exposure of the fruit had neither significant influence on the decay nor in the inhibitory effect obtained with the postharvest application of ARs.

    • In this proceeding, architecting novel magnetic Cross-Linked-Enzyme-Aggregates (CLEA)nanoparticles to effectively and controllably modify textile surfaces without damaging the fabric itself, will bediscussed. The efficiency is due to exponentially increased surface concentration of biocatalyst and to thechoice of aggregate size which controls the penetration depth and hence preventing the fabric disruption. Inaddition, due to the presence of magnetic nanoparticles, the continuous recovery and reusability makes thisdesign more economic and ecologic compared to the conventional chemical processes. The careful fabricationand functionalization of such a design with preliminary results will be presented in this contribution.

  • Isolation and structural identification of glycopolymers of Bifidobacterium bifidum BIM B-733D as putative players in pathogenesis of autoimmune thyroid diseases

    • Bifidobacterium bifidum 791 (commercially available as B. bifidum BIM B-733D) cell-surface biopolymers (BPs) interact selectively with human serum thyroid peroxidase (TPO) and thyroglobulin (Tg) autoantibodies (anti-TPO and anti-Tg, respectively). BPanti-TPO and BPanti-Tg were isolated from the soluble fraction of B. bifidum BIM B-733D by affinity chromatography with anti-TPO or anti-Tg, respectively. Homogeneity of affinity eluates (AE(anti-TPO) and AE(anti-Tg)) was tested by size exclusion chromatography. For each AE, the elution profiles generated on the basis of absorbance at 280 nm do not conform to ELISA data for functional activity characteristic of BPs. Moreover, high functional activity was detected in chromatographic fractions that had significantly different molecular weights and no absorbance at 280 nm, which suggests a non-protein (carbohydrate) nature of BPanti-TPO and BPanti-Tg. The semi-preparative size exclusion chromatography of AE(anti-TPO) and AE(anti-Tg) with detection by refractometer gave 5,000-7,000 Da fractions containing substances that interact selectively with either anti-TPO (BPanti-TPO) or anti-Tg (BPanti-Tg) according to ELISA data. Analysis by two-dimensional NMR spectroscopy including a H-1, C-13-heteronuclear single-quantum coherence experiment indicated that both substances are linear alpha-1,6-glucans. For the first time, an immunological similarity (molecular mimicry) of glycopolymers of B. bifidum BIM B-733D and human thyroid proteins, TPO and Tg, was shown. On the whole, our data point to a possible role of bifidobacteria in the pathogenesis of autoimmune thyroid diseases (ATD). The main requirements for triggering/acceleration or prevention/abrogation of ATD by bifidobacteria through molecular mimicry mechanism are hypothesised to be (1) genetic predisposition to ATD and (2) intestinal epithelium penetration by alpha-1,6-glucan.

  • Preparation of molecularly imprinted polymers in supercritical carbon dioxide

    • Molecularly imprinted polymer nanoparticles were prepared in supercritical carbon dioxide using a noncovalent imprinting approach. In the present work, propranolol was used as a model template, methacrylic acid as a functional monomer, and divinylbenzene as a crosslinker. Under a high dilution condition, the heterogeneous polymerization resulted in discrete crosslinked polymer nanoparticles. Compared with the nonimprinted polymers, the imprinted nanoparticles displayed much higher propranolol uptake in a low polarity organic solvent. The use of a single enantiomer (S)-propranolol as the template clearly demonstrated that the imprinted binding sites are chiral-selective, with a cross-reactivity towards (R)-propranolol of less than 5%. The overall binding performance of the imprinted nanoparticles was comparable to imprinted polymers prepared in conventional organic solvents. (c) 2006 Wiley Periodicals, Inc.

    • A combination of supercritical carbon dioxide (scCO2) impregnation of pyrrole and sonochemical transformation of permanganate (KMnO4) was used to impart conductive and catalytic properties to silk fibers. The results indicated that the conductivity (from polypyrrole –PPy) and catalytic activities (from manganese dioxide –MnO2) were independent and complementary within the processing parameters used. The enhanced conductivity was attributed to scCO2 preferentially distributing the pyrrole monomers along with the silk internal fibrillar structure and hence, yielding a more linear PPy. The oxidative properties of the PPy-MnO2-silk hybrid showed an enzyme-like behavior for the degradation of hydrogen peroxide (H2O2) with a Km of about 13 mM and specific activity of 1470 ± 75 μmol/min/g. Finally, we demonstrated that the PPy-MnO2-silk hybrid could be used as soft working electrodes for the simultaneous degradation and detection of H2O2.

    • The use of supercritical CO, (SC-CO2) for extracting alkylresorcinols from rye bran was investigated. For pure SC-CO2, 35 MPa and 55 degrees C were used. SC-CO2 with 10% ethanol or methanol as co-solvents was tested at four pressure levels (8, 15, 30 and 35 MPa) and two temperatures (40 and 55 degrees C). Experiments were conducted at a constant SC-CO2 flow of 5 g/min for 2 h. For comparative purposes acetone at ambient temperature and pressure was also used for extraction. The best extractions were achieved when SC-CO2 was used with co-solvents. At 55 degrees C and 15-30 MPa, the use Of SC-CO2 with co-solvents has yielded from 8 to 80% (w/w) more extracted product than acetone. Yield of extracts was proportional to pressure (when above 30 MPa) and temperature. The results suggest that cleaner bran extracts can be obtained by using pure SC-CO2 followed by the addition of co-solvents. There is no clear advantage of using either methanol or ethanol.     • This chapter focuses on the use of different methods for isolation of alkylresorcinols. Alkylresorcinols are members of a lipid group called non-isoprenoid phenolic lipids. Different aspects of the extraction by classical methods and supercritical CO2 are discussed. Supercritical CO2 extraction of alkylresorcinols from rye bran is discussed for the first time. As compared to the classical extraction methods, supercritical CO2 gives higher yields and it allows the separation of the crude extrac into long- and short-chain alkylresorcinol homologues.

    • The continuous growth of world population and its concentration in the urban areas require food supplies that are continuous, sufficient and of good quality. To resolve this problem techniques have been developed for increasing food quantity and quality. The techniques are applied throughout the food chain from production, conservation and during distribution to the consumers (from "the field to the fork"). During handling of food, chemicals are often deliberately added to achieve improved processing and better quality. This is one of the main reasons food undergoes different kinds of contamination. This overview focuses on the application of supercritical fluids as media for handling food materials during processing with the perspective of reducing chemical contamination of food. Examples of developmental applications of this technique and on research work in process are presented. Emphasis is given to extraction and biotransformation techniques.

    • Tailored supercritical carbon dioxide (scCO(2)) extraction of alkyhesorcinols (ARs) from rye bran resulting in pre-purification of ARs already during extraction process. The pre-purification was achieved by splitting the extraction process in 2 steps.. In both steps, the CO2 pressure was set at 25 MPa with a flow rate of 10 g/min. The first step was carried out at 70 degrees C. 0.06% of ethanolic co-solvent for 2 h, followed by the second step at 45,C using 10% of ethanolic co-solvent for 4 h, resulting in a pre-purified ARs extract. From the pre-purified scCO(2) extract, chromatographed on C8 column, were obtained pure ARs homologues of C17, C19 and C21 (68% of total) eluting separately in a linear gradient of ethanol. Additionally, a rapid method for the isolation of high purified pools of ARs homologues using a disposable solid phase extraction SPE-C18 column and a step gradient of ethanol was developed.
