  • Origin of organic compounds in fluids from ultramafic-hosted hydrothermal vents of the Mid-Atlantic Ridge

    • Natural gas, primarily methane (CH4), is produced in substantial amounts in ultramafic-hosted hydrothermal systems. These systems could also generate oil (heavier hydrocarbons) and the first building blocks of life (prebiotic molecules). In the presence of iron bearing minerals, serpentinisation reactions generate H2. Subsequently, CH4 could be synthesised by Fischer-Tropsch Type (FTT) reaction (4H2 + CO2 → CH4 + 2H2O) which is an abiotic process. This has lead to the idea of abiotic formation of larger organic molecules. Both thermodynamics and laboratory work support this idea, yet field data have been lacking. This study focuses on determining the organic content of fluids from ultramafic-hosted hydrothermal systems from the Mid-Atlantic Ridge (MAR) and the origin of the compounds. Fluids were collected from the Lost City, Rainbow, Ashadze and Logatchev vent fields during the EXOMAR (2005), SERPENTINE (2007), MoMARDREAMnaut (2007) and MOMAR08-Leg2 (2008) cruises conducted by IFREMER, France. A SBSE-TD-GC-MS technique was developed and used to extract, concentrate, separate and identify compounds in the fluids. Hydrothermally derived compounds appeared to consist mainly of hydrocarbons and O-bearing molecules. In addition, some amino acids were detected in the fluids by ULPC-ESI-QToF-MS but their origin will need to be determined. The organic content of the Rainbow fluids did not show intra field variability unlike differences could be noted over the years. In order to address the question of the source of the molecules, compound specific carbon isotopic analyses were carried out and completed with a bacterial (Pyrococcus abyssi) hydrothermal degradation experiment. The δ13C data fall in the range of -40 to -30‰ (vs. V-PDB), but individual δ13C values preclude the identification of a biogenic or abiogenic origin of the compounds. The degradation experiment, however, suggests an abiogenic origin of a portion of saturated hydrocarbons whereas carboxylic acids would be biogenic, and aromatic compounds would be thermogenic.

  • Detection of trace organic compounds by Stir Bar Sorptive Extraction: application to the evolution of the organic composition of hydrothermal fluids from the Rainbow site (36°N, Mid-Atlantic Ridge) over 3 years

    • The question of the abiogenic oil has been discussed and hydrothermal reactions have been suggested to abiotically generate hydrocarbons. Besides, ultramafic-hosted hydrothermal systems have been regarded as favourable environment for the emergence of life. Therefore, it is of great importance to investigate the presence of organic compounds in hydrothermal fluids. However, because of the sampling difficulties, the complexity of the matrix, the numerous interferences and the assumed trace level concentrations; it was considered a real challenge. Here we show that Stir Bar Sorptive Extraction (SBSE) coupled to Thermal Desorption – Gas Chromatography – Mass Spectrometry (TD-GC-MS) can be successfully applied to analyses of volatile and semi-volatile organic compounds in hydrothermal fluids. The method exhibited excellent reproducibility and conditioned samples appeared to be really stable for up to 3 years. This innovative and versatile technique was used to study the fluids from the ultramafic-hosted hydrothermal field Rainbow located at 36°14’N on the Mid-Atlantic Ridge (MAR). Despite relative stability for mineral composition and gas content is observed, analyses of fluids samples from 2005, 2007 and 2008 revealed more distinct organic compositions. In addition, results suggest the similarity of the organic content of the fluids regardless of the sampling location within the field, which may indicates the presence of a single reaction zone and that no further chemical reactions occur while fluids are transported to the seafloor.

    • The role of muscarinic acetylcholine receptors (mAChRs) in regulating glucose uptake in L6 skeletal muscle cells was investigated. [H-3]-2-Deoxyglucose uptake was increased in differentiated L6 cells by insulin, acetylcholine, oxotremorine-M and carbachol. mAChR-mediated glucose uptake was inhibited by the AMPK inhibitor Compound C. Whole cell radioligand binding using [H-3]-N-methyl scopolamine chloride identified mAChRs in differentiated but not undifferentiated L6 cells and M-3 mAChR mRNA was detected only in differentiated cells. M-3 mAChRs are Gq-coupled, and cholinergic stimulation by the rnAChR agonists acetylcholine, oxotremorine-M and carbachol increased Ca2+ in differentiated but not undifferentiated L6 cells. This was due to muscarinic but not nicotinic activation as responses were antagonised by the muscarinic antagonist atropine but not the nicotinic antagonist tubocurarine. Western blotting showed that both carbachol and the AMPK activator AICAR increased phosphorylation of the AMPK alpha subunit at Thr172, with responses to carbachol blocked by Compound C and the CaMKK inhibitor STO609 but not by the PI3K inhibitor wortmannin. AICAR-stimulated AMPK phosphorylation was not sensitive to STO-609, confirming that this compound inhibits CaMKK but not the classical AMPK kinase LKB1. The TAK1 inhibitor (5Z)-7-oxozeaenol and the G(i) inhibitor pertussis toxin both failed to block AMPK phosphorylation in response to carbachol. Using CHO-K1 cells stably expressing each of the mAChR subtypes (M-1-M-4), it was determined that only the M-1 and M-3 mAChRs phosphorylate AMPK, confirming a G(q)-dependent mechanism. This study demonstrates that activation of M-3 mAChRs in L6 skeletal muscle cells stimulates glucose uptake via a CaMKK-AMPK-dependent mechanism, independent of the insulin-stimulated pathway.

  • New insight into the contributions of thermogenic processes and biogenic sources to the generation of organic compounds in hydrothermal fluids

    • Experiments on hydrothermal degradation of Pyrococcus abyssi biomass were conducted at elevated pressure (40 MPa) over a 200–450 °C temperature range in sapphire reaction cells. Few organic compounds could be detected in the 200 °C experiment. This lack was attributed to an incomplete degradation of P. abyssi cells. On the contrary, a wide range of soluble organic molecules were generated at temperatures ≥350 °C including toluene, styrene, C8–C16 alkyl-benzenes, naphthalene, C11–C16 alkyl-naphthalenes, even carbon number C12–C18 polycyclic aromatic hydrocarbons, C15–C18 alkyl-phenanthrenes and C8:0–C16:0 n-carboxylic acids. The effect of time on the final organic composition of the degraded P. abyssi solutions at 350 °C was also investigated. For that purpose the biomass was exposed for 10, 20, 60, 90, 270 and 720 min at 350 °C. We observed a similar effect of temperature and time on the chemical diversity obtained. In addition, temperature and time increased the degree of alkylation of alkyl-benzenes. This study offers additional evidence that a portion of the aliphatic hydrocarbons present in the fluids from the Rainbow ultramafic-hosted hydrothermal field may be abiogenic whereas a portion of the aromatic hydrocarbons and n-carboxylic acids may have a biogenic origin. We suggest that aromatic hydrocarbons and linear fatty acids at the Rainbow site may be derived directly from thermogenic alteration of material from the sub-seafloor biosphere. Yet we infer that the formation and dissolution of carboxylic acids in hydrothermal fluids may be controlled by other processes than in our experiments.

  • THE POTENTIAL OF ABIOTIC ORGANIC SYNTHESIS IN ALKALINE ENVIRONMENTS OF SUBDUCTION ZONES

    • Reduced carbon and nitrogen precursor compounds may be formed under off-axis hydrothermal conditions in oceanic lithosphere in the presence of native Fe and Ni and are adsorbed on authigenic layer silicates and zeolites. The native metals as well as the molecular hydrogen reducing CO2 to CO/CH4 and NO3-/NO2- to NH3/NH4+ are a result of serpentinization of mafic rocks. Once the reduced carbon and nitrogen compounds have been formed the synthesis of hydrogen cyanide is, in principle, possible [1]. HCN is central to most of the reaction pathways leading to abiotic formation of simple organic compounds containing nitrogen, such as amino acids, purines and pyrimidines. Oceanic plates are conveyor belts of reduced carbon and nitrogen compounds from the off-axis hydrothermal environments to the subduction zones, where compaction, dehydration, desiccation and diagenetic reactions affect the organic precursors. CO/CH4 and NH3/NH4+ in fluids distilled out of layer silicates and zeolites in the subducting plate at an early stage of subduction will react upon heating and form the HCN, which is then available for further organic reactions. Since the released fluids will rise through hydrated ultramafic mantle rocks of the lower part of the overriding plate, they will experience a strong pH increase. The reason for this increase is that the serpentinization of ferromagnesian minerals in ultramafic rocks in contact with the water leads to the formation of the secondary magnesium hydroxide mineral brucite, which causes high pH. The high pH may promote abiotic formation of carbohydrates like pentose sugars, particularly ribose, as well as nucleosides and even nucleotides, since pyrophosphate is stable under alkaline conditions at low water-rocks ratios [2]. Convergent margins in the initial phase of subduction must, therefore, be considered the most potent sites for prebiotic organic reactions on terrestrial planets, provided that some type of plate tectonics occurs.

  • Detection of trace concentrations of amino acid in aqueous solutions by Ion-pairing reversed-phase UPLC-ESI-QToF-MS: application to hydrothermal fluids

    • Amino acids are the fundamental building blocks of proteins that are required for the initiation of living organism, therefore they are key compounds in the origin of life quest. The abiotic production of amino acids within ultramafic-hosted hydrothermal systems was suggested a few decades ago and is strongly supported by thermodynamic data and experimental work. However, field data were clearly lacking and thus investigation of amino acids in fluids from ultramafic-hosted hydrothermal systems was of major importance. Even though amino acids analysis have been carried out routinely for several decades in various field of application, none of the currently available methods appeared suitable for our purpose because of the complexity of the hydrothermal fluids matrix (salts, minerals, gases) and the extremely low concentrations of amino acids that were expected. We took up the challenge and here we describe a method to detect underivatized amino acids down to the sub-ppb level in aqueous matrix by ion-pairing reverse-phase Ultra-high Performance Liquid Chromatography - Electrospray Ionisation - Quadrupole Time of Flight - Mass Spectrometry (UPLC-ESI-QTOF-MS). Characterisation and separation of 10 chosen proteinogenic amino acids was achieved and excellent linearity in the response was obtained for all amino acids with correlation coefficient > 0.9921. This analytical method was successfully applied to natural hydrothermal fluid samples from ultramafic-hosted vents of the Mid-Atlantic Ridge region. Tryptophan and Phenylalanine were clearly evidenced in the fluids and concentrations of Leucine reached 15-55 ppt. We suggest that other amino acids are present in hydrothermal fluids but occur for the major part as metal and/or clay complexes and could not be detected using current approach. Finally, concentrations of amino acids measured in the deep seawater reference sample were in good agreement with generally accepted background level in the deep ocean.

    • Hydrocarbons gases (C1-C4) as well as larger organic compounds were reported elsewhere in fluids from the Rainbow and the Lost City ultramafic-hosted hydrothermal fields. Whereas hydrocarbon gases are likely abiogenic (Fischer-Tropsch Type reaction), the origin of larger molecules remains unclear. Our ability to differentiate between biotic and abiotic sources of organic compounds in deep-sea hydrothermal systems may give clues to the understanding of organic geochemistry on the early Earth. Here, we report a series of experiments of hydrothermal degradation (40 MPa / 200, 350 and 450°C) of the piezophile archaea Pyrococcus abyssi biomass, which provides supporting lines of evidence of the abiogenic origin of saturated hydrocarbons as well as of the possible biogenic origin of alkylated aromatic hydrocarbons, Polycyclic Aromatic Hydrocarbons (PAH) and C12:0-C16:0 fatty acids in fluids from ultramafic-hosted hydrothermal systems.

    • Along with summarizing the published literature and our own data some new results on properties of the trace metal biogeochemistry in the deep-sea hydrothermal ecosystems at the Mid-Atlantic Ridge (MAR) and East Pacific Rise (EPR) are shown. Differences in mean concentrations of big group of trace metals (Fe, Mn, Zn, Cu, Ni, Cr, Co, As, Pb, Cd, Ag, Hg) between the biotope water of the low- and high-temperature hydrothermal vent fields were firstly revealed. The same trace metals were studied in different groups of organisms within different temperature zones at one and the same vent field (9°50′N EPR), as well as in fauna inhabiting geochemically different vent sites. Distribution patterns of Fe, Mn, Zn, Cu, Cd, Pb, Ag, Ni, Cr, Co, As, Se, Sb, and Hg in different taxa gave an evidence of the influence of environmental and biological parameters on their bioaccumulation in organisms. Among the animals a particular “champion” with respect to the trace metal content was found to be a polychaeta Alvinella pompejana that inhabits the hottest places of the vent sulfide chimneys of the 9°50′N field, EPR. New data on the trace metal distribution between soft tissues and carbonate shell let us estimate a role of biomineralization in the accumulation of metals in the Bathimodiolus mussels. Contrasting geochemical behavior was revealed for Cu that is enriched in soft tissues of mussels and depleted in shells, on the one hand, and Mn that is accumulated almost totally in mussel shells, on the other hand.Deep-sea hydrothermal biological communities demonstrate a strong concentration function, and bioconcentration factors (BCF) of trace metals estimated for Bathimodiolus mussels collected at the four hydrothermal fields vary within the limits of n102–n105 and are similar to that of the littoral mussels. Due to this and to the high values of biomasses per square meter, the hydrothermal fauna may be considered as a newly discovered biological filter of the oceans.

    • Carbon, and molecules made from it, have already been observed in the early Universe. During cosmic time, many galaxies undergo intense periods of star formation, during which heavy elements like carbon, oxygen, nitrogen, silicon and iron are produced. Also, many complex molecules, from carbon monoxide to polycyclic aromatic hydrocarbons, are detected in these systems, like they are for our own Galaxy. Interstellar molecular clouds and circumstellar envelopes are factories of complex molecular synthesis. A surprisingly high number of molecules that are used in contemporary biochemistry on the Earth are found in the interstellar medium, planetary atmospheres and surfaces, comets, asteroids and meteorites and interplanetary dust particles. Large quantities of extra-terrestrial material were delivered via comets and asteroids to young planetary surfaces during the heavy bombardment phase. Monitoring the formation and evolution of organic matter in space is crucial in order to determine the prebiotic reservoirs available to the early Earth. It is equally important to reveal abiotic routes to prebiotic molecules in the Earth environments. Materials from both carbon sources (extra-terrestrial and endogenous) may have contributed to biochemical pathways on the Earth leading to life’s origin. The research avenues discussed also guide us to extend our knowledge to other habitable worlds.
