  • Optimizing the yield and physico-chemical properties of pine cone cellulose nanocrystals by different hydrolysis time

    • Cellulose nanocrystals (CNCs) were isolated for the first time from pine cones (PC) by alkali and bleaching treatments and subsequent sulfuric acid hydrolysis (64%) at 45 degrees C. The influence of the hydrolytic reaction time (30, 45, and 90 min) on the yield, chemical composition and structure, and thermal stability of CNCs was evaluated. The removal of non-cellulosic constituents during the alkaline and bleaching treatment resulted in high pure cellulosic fibres. The isolation of CNCs from these cellulosic fibres at different reaction times was verified by the nano-dimensions of the individual crystals (< 3 and < 335 nm of average diameter and length, respectively). The highest yield (15%) and the optimum CNCs properties in terms of aspect ratio, thermal stability and crystallinity were obtained for an extraction time of 45 min. PC appeared to be a new promising source of cellulose fibres and CNCs with potential to be applied as reinforcement in composites and for food-packaging.

    • In this work, different loads (3, 5 and 7 wt%) of pine cone cellulose nanocrystals (CNCs) were added to films ofpoly(3-hydroxybutyrate)/poly(ε-caprolactone) (PHB/PCL) blends with a composition of 75 wt% PHB and 25 wt% PCL (PHB75/PCL25). The films were obtained after solvent casting followed by melt compounding in anextruder and finally subjected to a thermocompression process. The influence of different CNCs loadings on themechanical, thermal, optical, wettability and disintegration in controlled compost properties of the PHB75/PCL25blend was discussed. Field emission scanning electron microscopy (FESEM) revealed the best dispersion of CNCson the polymeric matrix was at a load of 3 wt%. Over this loading, CNCs aggregates were formed enhancing thefilms fragilization due to stress concentration phenomena. However, the addition of CNCs improved the opticalproperties of the PHB75/PCL25films by increasing their transparency and accelerated the film disintegration incontrolled soil conditions. In general, the blend with 3 wt% CNCs offers the best balanced properties in terms ofmechanical, thermal, optical and wettability

    • Sustainable biocomposites based on thermoplastic starch copolymers (Mater-Bi KE03B1) and biofibres (cotton, hemp and kenaf) were prepared and characterised in terms of their thermo-mechanical and morphological properties. Biocomposites exhibit improved thermal stability and mechanical properties in comparison with the Mater-Bi KE. Biofibres act as suitable thermal stabilizers for the Mater-Bi KE, by increasing the maximum decomposition temperature and the Ea associated to the thermal decomposition process. Biofibre addition into the Mater-Bi KE results in higher storage modulus and in a reduction of the free-volume-parameter associated to the Mater-Bi KE glass transition. The influence of different biofibres on the thermo-mechanical properties of the biocomposites has been discussed. Hemp and kenaf enhance the thermal stability and reduce the free volume-parameter of Mater-Bi KE more significantly than cotton fibres, although the latter exhibits the highest mechanical performance. These differences may be explained by the improved interaction of lignocellulosic fibres with the Mater-Bi KE, due to the presence of hemicellulose and lignin in their formulation. © 2010 Elsevier Ltd.     • The spectral functions of highly correlated systems Gd and NiO have been calculated within the GW approximation. In the case of Gd, satellites are found at the positions of the experimental main peaks. However, some quasiparticle weight remains at the local density eigenvalue but its weight is unusually small (0.3). In NiO, satellite structure is observed at around -10 eV when starting from the local density Hamiltonian. It is found, however, that the satellite intensity is reduced towards self-consistency and almost disappears. © 1996 The American Physical Society.

    • A Green’s function formalism for calculating spin-wave excitations is developed for practical calculations and tested for real solids. The mapping to the Heisenberg Hamiltonian commonly used in spin-wave calculations is avoided, making the formalism suitable for both localized and itinerant magnetic systems. To test the formalism, we have calculated the spin-wave spectra and dispersions of ferromagnetic Fe and Ni. The results prove to be in very good agreement with experiment and some novel features are predicted.

  • Non-Invasive Acoustical sensing of Drug-Induced Effects on the Contractile Machinery of Human Cardiomyocyte Clusters

    • There is an urgent need for improved models for cardiotoxicity testing. Here we propose acoustic sensing applied to beating human cardiomyocyte clusters for non-invasive, surrogate measuring of the QT interval and other characteristics of the contractile machinery. In experiments with the acoustic method quartz crystal microbalance with dissipation monitoring (QCM-D), the shape of the recorded signals was very similar to the extracellular field potential detected in electrochemical experiments, and the expected changes of the QT interval in response to addition of conventional drugs (E-4031 or nifedipine) were observed. Additionally, changes in the dissipation signal upon addition of cytochalasin D were in good agreement with the known, corresponding shortening of the contraction-relaxation time. These findings suggest that QCM-D has great potential as a tool for cardiotoxicological screening, where effects of compounds on the cardiomyocyte contractile machinery can be detected independently of whether the extracellular field potential is altered or not.

    • The energy-resolved momentum densities of thin polycrystalline aluminium films have been measured using electron momentum spectroscopy (EMS), for both the valence band and the outer core levels. The spectrometer used for these measurements has energy and momentum resolutions of around 1.0 eV and 0.15 atomic units, respectively. These measurements should, in principle, describe the electronic structure of the film very quantitatively, i.e. the dispersion and the intensity can be compared directly with theoretical spectral momentum densities for both the valence band and the outer core levels. Multiple scattering is found to hamper the interpretation somewhat. The core-level intensity distribution was studied with the main purpose of setting upper bounds on these multiple-scattering effects. Using this information we wish to obtain a full understanding of the valence band spectra using different theoretical models of the spectral function. These theoretical models differ significantly and only the cumulant expansion calculation that takes the crystal lattice into account seems to describe the data reasonably well.

    • Inflammatory bowel disease is characterized by a number of immunological alterations, not the least in the T-cell compartment. Numerous animal models of colitis have revealed aberrant thymocyte dynamics associated with skewed thymocyte development. The recent advancements in quantitative methods have proposed critical kinetic alterations in the thymocyte development during the progression of colitis. This review focuses on the aberrant thymocyte dynamics in Gαi2-deficient mice as this mouse model provides most quantitative data of the thymocyte development associated with colitis. Herein, we discuss several dynamic changes during the progression of colitis and propose a hypothesis for the underlying causes for the skewed proportions of the thymocyte populations seen in the Gαi2-deficient mice and in other mouse models of colitis.

    • Plastocyanin (Pc) is a copper-containing protein, which functions as an electron carrier between the cytochrome b6f and photosystem 1 (PS1) complexes in the photosynthetic electron transfer (ET) chain. The ET is mediated by His87 situated in the hydrophobic surface in the north region of Pc. Also situated in this region is Leu12, which mutated to other amino acids severely disturbs the ET from cytochrome f and to PS1, indicating the importance of the hydrophobic surface. The crystal structure of the Pc double mutant G8D/L12E has been determined to 2.0 Å resolution, with a crystallographic R-factor of 18.3% (Rfree=23.2%). A comparison with the wild-type structure reveals that structural differences are limited to the sites of the mutations. In particular, there is a small but significant change in the hydrophobic surface close to His87. Evidently, this leads to a mismatch in the reactive complex with the redox partners. For PS1 this results in a 20 times weaker binding and an eightfold slower ET as determined by kinetic measurements. The mutations that have been introduced do not affect the optical absorption spectrum. However, there is a small change in the EPR spectrum, which can be related to changes in the copper coordination geometry.

  • From forest residues to hydrophobic nanocomposites with high oxygen-barrier properties

    • A biorefinery of forest resources should be able to convert all components of trees, including the bark and other types of forest residues, into value-added products. Here, non-cellulosic polysaccharides (NCPs) isolated from Norway spruce bark and cellulose nanocrystals (CNCs) isolated from the logging residues of Norway spruce were mixed to prepare nanocomposites with competitive thermo-mechanical properties. Polyepoxy acid (PEA) derived from a monomer of suberin in birch bark was used as a coating on the nanocomposites to develop functional materials entirely based on forest resources. All of the PEA-coated nanocomposites were hydrophobic. At 50% and 80% relative humidity, they showed high oxygen-barrier properties that were comparable to or even better than those of some renewable materials such as xylan-, galactoglucomannan- and nanofibrillated cellulose-based films and synthetic materials such as polyvinylidene chloride and polyamide.
