    • The performance of a constructed wetland for treatment of landfill leachate has been evaluated based upon data obtained during 4 years (2003-2006). It consists of a series of 10 ponds with a total capacity of 52.000 m3 covering 8 ha. Using univariate and multivariate statistics (PCA) the reduction pattern of a large number of chemical parameters including heavy metals has been investigated in 3 parts of the wetland with equal volume. Analyses show that many parameters are removed to the greatest extent in the first part of the system (e.g. many heavy metals, total suspended solids) or the second part of the system (N-NH4) while other parameters such as total nitrogen are more gradually reduced (10 ton/year removed).  Toxicity testing with 5 bioassays showed that toxicity was sometimes observed at the inlet but no toxicity was observed at the outlet for 4 of the test species. The data presented will be used for optimizing the treatment process as well as to improve the monitoring program.

    • One of the open issues in fingerprint verification is the lack of robustness against image-quality degradation. Poor-quality images result in spurious and missing features, thus degrading the performance of the overall system. Therefore, it is important for a fingerprint recognition system to estimate the quality and validity of the captured fingerprint images. In this work, we review existing approaches for fingerprint image-quality estimation, including the rationale behind the published measures and visual examples showing their behavior under different quality conditions. We have also tested a selection of fingerprint image-quality estimation algorithms. For the experiments, we employ the BioSec multimodal baseline corpus, which includes 19 200 fingerprint images from 200 individuals acquired in two sessions with three different sensors. The behavior of the selected quality measures is compared, showing high correlation between them in most cases. The effect of low-quality samples in the verification performance is also studied for a widely available minutiae-based fingerprint matching system.

    • We report on experiments for the fingerprint modality conducted during the First BioSecure Residential Workshop. Two reference systems for fingerprint verification have been tested together with two additional non-reference systems. These systems follow different approaches of fingerprint processing and are discussed in detail. Fusion experiments involving different combinations of the available systems are presented. The experimental results show that the best recognition strategy involves both minutiae-based and correlation-based measurements. Regarding the fusion experiments, the best relative improvement is obtained when fusing systems that are based on heterogeneous strategies for feature extraction and/or matching. The best combinations of two/three/four systems always include the best individual systems whereas the best verification performance is obtained when combining all the available systems.

    • Use of biomaterials in autoimmunity research is widely explored, as an adjuvant to induce antigen specific immune responses, for facilitating induction of experimental models to study disease pathogenesis and for designing novel therapeutic targets. Similarly, polymeric biomaterials are explored as a delivery vehicle for sustained and specific release of auto-antigens/drugs to treat autoimmune disorders. Although considered as biocompatible, implantation/injection of polymers like silica and metallic implants are associated with development of chronic inflammation and autoimmunity. Despite these compatibility concerns, biomaterials are still considered as favorable materials for several applications in the autoimmunity field.

    • An integrated cell cultivation and protein product separation process was developed using a new type of supermacroporous polyacrylamide gel, called cryogel (pAAm-cryogel) support matrix. Human fibrosarcoma HT1080 and human colon cancer HCT116 cell lines were used to secrete urokinase (an enzyme of immense therapeutic utility) into the culture medium. The secreted protein was isolated from the circulating medium using a chromatographic capture column. A pAAm cryogel support with covalently coupled gelatin (gelatin-pAAm cryogel) was used for the cultivation of anchorage dependent cells in the continuous cell culture mode in 5% carbon dioxide atmosphere. The cells were attached to the matrix within 4-6 h of inoculation and grew as a tissue sheet inside the cryogel matrix. Continuous urokinase secretion into the circulating medium was monitored as a parameter of growth and viability of cells inside the bioreactor. No morphological changes were observed in the cells eluted from the gelatin-cryogel support and re-cultured in T-flask. The gelatin-pAAm cryogel bioreactor was further connected to a pAAm cryogel column carrying Cu(II)-iminodiacetic acid (Cu(II)-IDA)-ligands (Cu(II)-IDA-pAAm cryogel), which had been optimized for the capture of urokinase from the conditioned medium of the cell lines. Thus an automated system was built, which integrated the features of a hollow fiber reactor with a chromatographic protein separation system. The urokinase was continuously captured by the Cu(II)-IDA-pAAm cryogel column and periodically recovered through elution cycles. The urokinase activity increased from 250 PU/mg in the culture fluid to 2,310 PU/mg after recovery from the capture column which gave about ninefold purification of the enzyme. Increased productivity was achieved by operating integrated bioreactor system continuously for 32 days under product inhibition free conditions during which no back-pressure or culture contamination was observed. A total 152,600 Plough units of urokinase activity was recovered from 500 mL culture medium using 38 capture columns over a period of 32 days. (c) 2006 Wiley Periodicals, Inc.

    • The increasing interest in Additive Manufacturing (AM) is due to its huge advantage in producing parts without any geometrical limitations. It is due to this reason, AM is extensively utilized in automotive, aerospace, medical and dental applications. Despite their popularity, AM is often associated with inferior surface quality which is one of the many reasons why it has failed to fully replace traditional methods. Hence, AM is always followed by a subsequent post-processing step to produce the end-product. To establish control over the surface quality it is first necessary to fully understand the surface behaviour concerning the factors affecting it. In this paper, the focus is mainly on having a better understanding of the surfaces by using scale-sensitive fractal analysis. In addition, the paper documents the influence of build inclination and post-processing in particular shot blasting on surface topography and utilizes a multi-scale approach to identify the most important scale and parameters for characterization. Results of this study reveal that shot blasting has a minimalistic effect on surface features at a large scale as it cannot remove the waviness completely. At smaller scales, blasting imparts additional features on the surface due to the impact of abrasive particles at high pressure. At the intermediate scales, the influence of shot blasting is highest as it successfully eliminates the surface features comprising of partially melted powder particles and stair-step effect.

  • Monoclonal antibody production using a new supermacroporous cryogel bioreactor

    • A supermacroporous cryogel bioreactor has been developed to culture hybridoma cells for long-term continuous production of monoclonal antibodies (mAb). Hybridoma clone M2139, secreting antibodies against J1 epitope (GERGAAGIAGPK; amino acids, 551-564) of collagen type II, are immobilized in the porous bed matrix of a cryogel column (10 mL bed volume). The cells got attached to the matrix within 48 h after inoculation and grew as a confluent sheet inside the cryogel matrix. Cells were in the lag phase for 15 days and secreted mAb into the circulation medium. Glucose consumption and lactic acid production were also monitored, and during the exponential phase (similar to 20 days), the hybridoma cell line consumed 0.75 mM day(-1) glucose, produced 2.48 mM day(-1) lactic acid, and produced 6.5 mu g mL(-1) day(-1) mAb during the exponential phase. The mAb concentration reached 130 mu g mL(-1) after continuous run of the cryogel column for 36 days. The yield of the mAb after purification was 67.5 mg L-1, which was three times greater than the mAb yield obtained from T-flask batch cultivation. Even after the exchange of medium reservoir, cells in the cryogel column were still active and had relatively stable mAb production for an extended period of time. The bioreactor was operated continuously for 55 days without any contamination. The results from ELISA as well as arthritis experiments demonstrate that the antibodies secreted by cells grown on the cryogel column did not differ from antibodies purified from the cells grown in commercial CL-1000 culture flasks. Thus, supermacroporous cryogels can be useful as a supporting material for productive hybridoma cell culture. Cells were found to be viable inside the porous matrix of the cryogel during the study period and secreted antibodies continuously. The antibodies thus obtained from the cryogel reactor were found to be functionally active in vivo, as demonstrated by their capacity to induce arthritis in mice.

    • There is great potential to apply energy recovery from municipal wastewater treatment plants (WWTP) to achieve better energy efficiency and greenhouse gas emissions abatement. However, in most of the wastewater treatment plants that produce biogas through sludge digestion, there is no thermal energy recovery or electricity generation, and the biogas is flared and wasted, particularly in developing countries. This paper discusses the recent technological advances related to biogas recovery from wastewater treatment besides identifying research gaps and local conditions, which affect implementation. Techniques for enhancement of biogas production such as co-digestion and microalgae systems for energy recovery were reviewed in terms of performance and context for application. This paper provides an assessment of how local conditions promote or hinder biogas recovery from WWTP in megacities of developing countries. The lack of government subsidies and a biogas market that is not well developed and regulated hinder biogas recovery implementation in the studied megacities. Infrastructure and electricity prices are also discussed. Strategies to promote energy recovery are recommended based on successful examples of energy self-sufficient WWTP. The findings may not only support planning on energy recovery but also guide technological development and research on this topic, aligned with the sustainable development goals.

  • Biomethane yield from different European Phragmites australis genotypes, compared with other herbaceous wetland species grown at different fertilization regimes

    • Phragmites australis, Typha latifolia, T. angustifolia and Arundo donax are tall wetland graminoids with the potential to replace fossil fuels under sustainable cultivation conditions. We investigated the biomethane (CH4) production of these four species, including four different genotypes of P. australis, which represent the high intraspecific diversity of European reed. All plants were grown under three different macronutrient supplies (no nutrients added, an equivalent of 75 kg N ha−1 year−1 added and an equivalent of 500 kg N ha−1 year−1 added). Biomethane production was measured in four independent batch digestion tests. Across all experiments, fertilization regime had little effect on CH4 yield, which was on average 222 ± 31 L kg−1 volatile solids (VS). The lowest yield was produced by T. angustifolia (140 L kgVS−1) receiving no nutrients, while the highest yield was produced by A. donax (305 L kgVS−1) in the highest nutrient treatment. The intraspecific diversity of P. australis did not affect biomethane production. All P. australis genotypes produced on average 226 ± 19 L CH4 kgVS−1, which, although high, was still lower than conventional biogas species. The biomass production of P. australis was less increased by fertilization than that of Typha sp. and A. donax, but all species had similar biomass without fertilization.
