  • Metagenomic Sequencing of Marine Periphyton: Taxonomic and Functional Insights into Biofilm Communities

    • Periphyton communities are complex phototrophic, multispecies biofilms that develop on surfaces in aquatic environments. These communities harbor a large diversity of organisms comprising viruses, bacteria, algae, fungi, protozoans and metazoans. However, thus far the total biodiversity of periphyton has not been described. In this study, we use metagenomics to characterize periphyton communities from the marine environment of the Swedish west coast. Although we found approximately ten times more eukaryotic rRNA marker gene sequences compared to prokaryotic, the whole metagenome-based similarity searches showed that bacteria constitute the most abundant phyla in these biofilms. We show that marine periphyton encompass a range of heterotrophic and phototrophic organisms. Heterotrophic bacteria, including the majority of proteobacterial clades and Bacteroidetes, and eukaryotic macro-invertebrates were found to dominate periphyton. The phototrophic groups comprise Cyanobacteria and the alpha-proteobacterial genus Roseobacter, followed by different micro- and macro-algae. We also assess the metabolic pathways that predispose these communities to an attached lifestyle. Functional indicators of the biofilm form of life in periphyton involve genes coding for enzymes that catalyze the production and degradation of extracellular polymeric substances, mainly in the form of complex sugars such as starch and glycogen-like meshes together with chitin. Genes for 278 different transporter proteins were detected in the metagenome, constituting the most abundant protein complexes. Finally, genes encoding enzymes that participate in anaerobic pathways, such as denitrification and methanogenesis, were detected suggesting the presence of anaerobic or low-oxygen micro-zones within the biofilms.

    • The development of cracks and changes in appearance have been investigated on radial and tangential sections of pine (Pinus sylvestris L.) and spruce (Picea abies Karst.) after exposure outdoors for 61 months. The degradation of the sections has also been studied at the micro-level.The annual ring orientation was the most important factor affecting crack development on weathering. After 61 months of outdoor exposure, the tangential sections of spruce had 1.7–2.2 times greater mean total crack length per area unit than the corresponding radial sections. In pine, the total crack length per area unit on the tangential sections was 2.2–2.6 times greater than that on the radial sections. Tangential and radial sections show the same colour change as a result of weathering.Tangential sections have more and deeper cracks than radial surfaces. The cracks on the tangential sections occur frequently in both earlywood and latewood. On radial sections, cracks occur primarily at the annual ring borders, but to a certain extent also in the earlywood.Decomposition of the cell wall takes place in both radial and tangential cell walls and cracks tend to follow the fibril orientation in the S2-layer of the cell wall. The radial cell wall of the earlywood has a large number of pits which are degraded at an early stage.

  • X-Band Interferometric SAR Observations of Baltic Fast Ice

    • Detailed mapping of fast-ice deformation can be used to characterize the rheological behavior of fast ice and subsequently improve sea ice modeling. This study uses interferometric synthetic aperture radar to map fast-ice deformation with unprecedented spatial resolution (meter range) and sensitivity (cm-mm range). Two interferometric acquisitions, each with a temporal baseline of 24 h, were performed by the X-band SAR satellite constellation Cosmo-SkyMed over the northeast Bay of Bothnia in the middle of the 2012 ice season. The first interferogram shows deformation of the fast ice due to force from impinging drift ice, and the normal strain within the fast ice is measured. Complementary intensity correlation measurements reveal a slow movement of the drift ice toward the fast ice. The second interferogram exhibits a low fringe rate over the fast ice with fringes being aligned along the coastline. Deformation appears to be stronger around leads, skerries, and grounded ice ridges. It is also observed that the coherence images provide information that is complementary to the information in the backscatter images.

    • Beskriver historien över 1000 motoriserade fiskebåtar med träskrov. Förutom fartygsnamn och hemmahamn ges dokumenterade uppgifter om ägarna under fartygens skiftande historia, motorbyten och vad som blev fartygens öde. Fartygen har blivit kondemnerade, minsprängts, förlist, bunnit etc. En stor del av fartygen fanns fortfarande kvar i drift år 2000 - ett flertal som fritidsfartyg och några som ännu var fiskefartyg.

    • The links between market gardens and cities were symbiotic until the mid-1900s. In my thesis about market gardens in Sweden 1900-1950, four market gardens in Stockholm County are investigated, trying to find out how different resources and strategies contributed to the businesses competitiveness. Two of the market gardens belonged to mansions with all the staff employed. The other two were established as family businesses by entrepreneurs without employees. Garden products are sensitive to bumps, heat, low temperatures, etc. Due to this, the access to a nearby market was necessary at that time. The development of the trade with garden products was linked to a fast-growing population in the cities, the growing demand for garden products, and the development of the transport sector. From the growing city, gardeners bought horse manure, garbage, or anything that produced heat to the hotbeds when it decomposed. Most of the market gardens were small family businesses situated on small plots. In the 1930s and 1940s in 75% of the businesses a maximum of three people were engaged. The production was both resource-demanding and labour-intensive. The use of hotbeds and greenhouses extended the growing season and made it possible to grow all year around. The gardenerRSQUOs dependence on merchandises increased like an avalanche to beat the growing competition on the market. The import, often of high quality products, was growing very fast and affected the interest for garden products. It forced the domestic horticulture industry to increase the quality and effectiveness of the production.

    • The concept of productivity is often used to determine how well resources are used in an operation, and it is usually determined as the ratio of what is consumed in the production. Laminated veneer products are considered complicated products often taking complex shapes, using a raw material with high variation, and requiring machining processes that create scrap material that needs to be handled. Therefore, maintaining high productivity in industries producing such products may become challenging. This study reports on productivity measurements in a production cell consisting of an adhesive, pressing and a processing station. The study seeks to increase the understanding of production-related problems in this industry. This research has been based on productivity measurement as well as on interactive discussions between researchers and workers. Measurement of cycle times indicated bottlenecks in the processing cell. The discussion led via cycle times, processing residues and chatter marks to an examination of the foundation and rigidity of the CNC-machine in the processing cell. The study indicated that the performance of the CNC machine did not correspond to expectations. The machine was too weak to handle the required output in an efficient manner. Thus, there is a need to determine the performance expected before a machine or machine group is purchased. An update of the existing purchasing literature and its dissemination will support the crystallization of the purchasing process as a way forward to support the industry.

    • The growing environmental awareness is leading to an increased interest in the use of bio-based adhesives and proteins such as vital gluten. The purpose of this study was to investigate the influence of the wood-particle moisture content, water application and press time on the internal bond strength, thickness expansion and thickness swelling of particleboards glued with vital gluten. Green and dried wood particles with similar moisture contents were achieved through drying or water addition and were blended with vital gluten powder and pressed for 1 to 3 minutes. The results show that not only the pressing time and moisture content, but also the way of achieving the moisture content has a strong impact on the performance of the boards. At comparable moisture content, never-dried (green) particles with high moisture content in combination with a dry adhesive application produced boards which performed better than boards made of dry particles with water addition to simulate liquid adhesive application.
