    • Following systemic injection, hematoporphyrin derivative (HPD) is known to be selectively retained in malignant tissue. This property can be used in a two-fold way: a) for localizing tumours by observing the characteristic dual-peaked laser-induced fluorescence from HPD, and b) for photodynamic therapy (HPD-PDT) using a localized HPD-assisted singlet oxygen release induced by irradiation of 630 nm laser light. This rapidly developing field has recently been reviewed by DOUGHERTY.

  • Microcalorimetry as a tool for the detection of complement-dependent cytotoxicity

    • Microcalorimetry using a 4-channel static ampoule microcalorimeter of thermopile type has been evaluated as a tool for the detection of complement-dependent cytotoxicity against the surface antigens of living cells. Cytotoxic reactions mediated by a rabbit antiserum against human white blood cells and by 2 different monoclonal antibodies recognizing a melanoma-associated antigen on a human melanoma cell line were studied. The cytotoxic reactions were registered as a decrease of the heat production rate when the cells were exposed to antibodies in the presence of active complement as compared to the heat production rate of the cells exposed to the same antibodies in the presence of inactive complement. This investigation shows that microcalorimetry can be used as a highly sensitive method for the detection of complement-dependent immune reactions, detecting antibody dilutions higher than 10-5. It also indicates that microcalorimetry may become a particularly important technique in the analysis of the kinetics of cytotoxic immune reactions in vitro.

Contrast enhancement techniques have been developed which improve the ability to localize a tumor when using fluorescence from the tumor-seeking substance hematoporphyrin derivative (HPD). A multi-color fluorescence imaging technique has been tested. A discussion of clinical applications is included.

  • C-band heteromorphism in breast cancer patients

    • The pattern of heteromorphism in the C-band-positive constitutive heterochromatin of human chromosomes #1, #9, and #16 was studied in peripheral lymphocytes of 54 breast cancer patients and 78 control individuals. The parameters of the heterochromatic regions analyzed were relative size, symmetry-asymmetry within homologous pairs, and prevalence of inversions. Significant differences between the two groups were found in C-band size of chromosomes #1, #9, and #16 and in incidence of inversions on chromosomes #1 and #9. Significant differences were noted between premenopausal and postmenopausal cancer patients in regard to inversions on chromosome #9 and between familial and sporadic patients in regard to C-band size on chromosome #16.

  • Immune functions in melanoma patients during treatment with interferon [HuIFN-alpha (Le)] alone or in combination with cimetidine

    • While leukocyte interferon was found therapeutically ineffective in a series of 20 patients with metastatic malignant melanoma, subsequent combination treatment with interferon and cimetidine induced 5 complete and 1 partial tumour remissions. Prior to interferon therapy initiation, regressor patients demonstrated a significantly greater ability to mediate antibody-dependent cellular cytotoxicity than progressor patients and also tended to have higher natural killer-cell activity. These differences were more pronounced following in vitro exposure of effector cells to interferon alone or in combination with cimetidine. During therapy the differences decreased to statistically nonsignificant levels. The number of immunoglobulin producing cells and lymphocyte proliferative responses to Con A were found to increase in both patient groups after interferon therapy initiation; but this augmentation vanished gradually upon combined treatment with cimetidine. A gradual decrease of the number of T lymphocytes and granulocytes was also recorded. None of the demonstrated alterations in the activities of circulating lymphocytes appears to be a relevant correlate to the efficacy of combined therapy compared to interferon alone.

  • Cytogenetic studies in non‐Hodgkin lymphomas‐Results from fineneedle aspiration samples

    • 118 samples obtained by means of fine‐needle aspiration from 86 cases of non‐Hodgkin, non‐Burkitt lymphomas have been investigated cytogenetically with chromosome banding technique. If>5 times 106 cells were obtained at aspiration the success rate with the technique used was almost 70%, while if the number of cells was lower it was only about 15%. Technical aspects, and detailed cytogenetic findings in 48 successful cases, are reported. There was no difference in success rate due to tumor stage, or sampling at diagnosis or in relapse. It is concluded that the fine‐needle aspiration technique has certain advantages in cancer cytogenetics; it is easy to handle, requires no surgical intervention, causes little distress to the patient, and the success rate is acceptable.

    • The activites of ornithine decarboxylase (ODC) and ODC inhibitory protein (ODC-antizyme) were studied in Ehrlich ascites tumor cells, separated according to their position in the cell cycle by centrifugal elutriation. Release and/or synthesis of ODC-antizyme was induced by putrescine treatment. Each mouse received an intraperitoneal injection of 25 moles of putrescine at 0, 1, 2, and 3 hr after tumor transplantation. Tumor cells obtained from putrescine-treated and control mice at 4 hr after transplantation were separated into fractions representing all phases of the cell cycle. The cell cycle distribution of the tumor cells in each fraction was determined by flow cytometry. In control tumor cells the ODC activity exhibited two maxima; inlate-G1/early-S and in late-S/G2. A marked decrease in ODC activity was observed in mid-S phase. This decrease coincided with maximum ODC-antizyme activity (revealed by putrescine treatment), suggesting that ODC-antizyme is involved in the regulation of ODC activity during the cell cycle.
