    • Inom den regenerativa medicinen strävar man efter att ersätta skadat eller sjukligt biologiskt mänskligt material (celler, organ, kroppsdelar) med nya biologiska komponenter. Området aktualiserar en rad etiska frågeställningar vad gäller (1) produktionen av ersättningsmaterialet (t.ex. embryonala stamceller eller införskaffande av transplantationsvävnad från donatorer), (2) risker i samband med försök på människa (genmodifierat material, material från djur), samt (3) gränserna för hur långt man bör gå i denna slags försök att förlänga människans livsspann. Föredraget ger en kort översikt över dessa frågeställningar, ståndpunkter och argument i debatten kring dem.

    • Five cases of glycerol kinase deficiency are presented with clinical, biochemical, and genetic results. Two had the glycerol kinase deficiency as part of an Xp21 contiguous gene deletion syndrome-complex form-and three had an isolated form of the enzyme deficiency. In these we found two splice site mutations (IVS1+4A>G, IVS9-1G>T) and one insertion (1393_1394insG). In patients with the complex form, a deletion of the DAX1, GK genes and the distal part of the DMD gene was found. A computerized study was performed to predict the effects of the splice site mutations. It showed that the IVS9-1G>T mutation substantially altered and removed the wild-type site and enhanced a cryptic site seven nucleotides downstream, and that the IVS1+4A>G diminished the strength of the wild-type donor site from strong to leaky. To verify these predictions, we developed an RT-PCR system with gene-specific primers that exclusively amplifies the Xp21 glycerol kinase gene transcript. Identification of individuals at risk is motivated by a need to avoid delay in a correct diagnosis. For reliable identification of heterozygotes for isolated glycerol kinase deficiency, knowledge of the specific mutation in the proband is required. This is easily obtained with the RT-PCR analyses developed in this study.

    • Sets of four parameters (m, Eo, ~o and •) of the cellular track structure model of Katz have been fitted to recently published dataconcerning human melanoma (AA) and mammalian (V79) cells exposed to a variety of lighter ions and to mixed ion-Co60 and ionionirradiation. We discuss the predictive capability of this model and propose standards in reporting cellular radiobiology data forapplication in modelling heavy ion beam radiotherapy.

  • Optimization of single skin surfaces for head injury prevention - a comparison of optima calculated for global versus local injury thresholds

    • This paper describes optimizations of material properties for a bonnet-like plate using finite element calculations and the Euro-NCAP pedestrian head impact test. Four different head models were used for the impact simulations, a Euro-NCAP dummy head, a Hybrid III dummy head and two biomechanical head models exhibiting different mechanical properties for the brain tissue. The objective function was to minimize the displacement of the bonnet plate while satisfying constraints on the head injury criterion (HIC), the resultant contact force and, for the human head models, the strain in the brain tissue. An investigation was also conducted of the kinematics of the head models during impact, evaluating the energy distribution and the apparent mass. The analysis gave at hand that optimization of the plate with respect to impact with the Euro-NCAP and Hybrid III head models reached substantially, different results compared to impact with the biomechanical head models. For the latter case, the stiffness of the brain tissue influenced which constraints were active in the final solution. The investigation of the kinematics at impact showed that a substantial portion of energy was confined within the brain during impact for the biomechanical head models. The apparent mass at impact coincided with the actual mass for the rigid dummy heads while for the human head models it was roughly the mass of the skull only.

  • Protein adsorption on model surfaces with controlled nanotopography and chemistry

    • To evaluate the influence of substratum surface characteristics on protein adsorption processes, we have investigated the adsorption (adsorbed amount, supramolecular organization) of collagen on model substrata exhibiting controlled topography and surface chemistry. Substrata were prepared in two steps: (i) gold deposition onto silicon wafers (smooth substrata) and onto a support with nanoscale protrusions created by colloidal lithography (rough substrata); (ii) functionalization with CH3 (hydrophobic) and OH (hydrophilic) groups, using alkanethiol self-assembly. Atomic force microscopy (AFM) images were recorded under water, prior to and after collagen adsorption, and the images were analyzed quantitatively using two independent approaches. On smooth substrata, collagen formed a similar to6 nm thick, homogeneous layer with low roughness on hydrophilic surfaces, and a similar to20 nm thick layer exhibiting elongated aggregated structures on hydrophobic surfaces. Film thickness measurements (AFM) together with X-ray photoelectron spectroscopy (XPS) revealed larger adsorbed amounts on hydrophobic surfaces compared to hydrophilic ones. On rough substrata, the adsorbed amounts were similar to those found on smooth substrata; however, the collagen molecules no longer formed aggregated structures on the hydrophobic surfaces. It is concluded that while the adsorbed amount is only affected by the surface chemistry, the supramolecular organization of the adsorbed layer is controlled both by surface chemistry and topography. The approach presented here will have great value in biophysics for investigating bioadsorption and bioadhesion processes on substrata of defined surface properties.
