  • Phenotypic and transcriptomic acclimation of the green microalga Raphidocelis subcapitata to high environmental levels of the herbicide diflufenican

    • Herbicide pollution poses a worldwide threat to plants and freshwater ecosystems. However, the understanding of how organisms develop tolerance to these chemicals and the associated trade-off expenses are largely unknown. This study aims to investigate the physiological and transcriptional mechanisms underlying the acclimation of the green microalgal model species Raphidocelis subcapitata (Selenastraceae) towards the herbicide diflufenican, and the fitness costs associated with tolerance development. Algae were exposed for 12 weeks (corresponding to 100 generations) to diflufenican at the two environmental concentrations 10 and 310 ng/L. The monitoring of growth, pigment composition, and photosynthetic performance throughout the experiment revealed an initial dose-dependent stress phase (week 1) with an EC50 of 397 ng/L, followed by a time-dependent recovery phase during weeks 2 to 4. After week 4, R. subcapitata was acclimated to diflufenican exposure with a similar growth rate, content of carotenoids, and photosynthetic performance as the unexposed control algae. This acclimation state of the algae was explored in terms of tolerance acquisition, changes in the fatty acids composition, diflufenican removal rate, cell size, and changes in mRNA gene expression profile, revealing potential fitness costs associated with acclimation, such as up-regulation of genes related to cell division, structure, morphology, and reduction of cell size. Overall, this study demonstrates that R. subcapitata can quickly acclimate to environmental but toxic levels of diflufenican; however, the acclimation is associated with trade-off expenses that result in smaller cell size.

  • The delivery of organic contaminants to the Arctic food web : why sea ice matters

    • For decades sea ice has been perceived as a physical barrier for the loading of contaminants to the Arctic Ocean. We show that sea ice, in fact, facilitates the delivery of organic contaminants to the Arctic marine food web through processes that: 1) are independent of contaminant physical–chemical properties (e.g. 2–3-fold increase in exposure to brine-associated biota), and 2) depend on physical–chemical properties and, therefore, differentiate between contaminants (e.g. atmospheric loading of contaminants to melt ponds over the summer, and their subsequent leakage to the ocean). We estimate the concentrations of legacy organochlorine pesticides (OCPs) and current-use pesticides (CUPs) in melt pond water in the Beaufort Sea, Canadian High Arctic, in 2008, at near-gas exchange equilibrium based on Henry's law constants (HLCs), air concentrations and exchange dynamics. CUPs currently present the highest risk of increased exposures through melt pond loading and drainage due to the high ratio of melt pond water to seawater concentration (Melt pond Enrichment Factor, MEF), which ranges from 2 for dacthal to 10 for endosulfan I. Melt pond contaminant enrichment can be perceived as a hypothetical ‘pump’ delivering contaminants from the atmosphere to the ocean under ice-covered conditions, with 2–10% of CUPs annually entering the Beaufort Sea via this input route compared to the standing stock in the Polar Mixed Layer of the ocean. The abovementioned processes are strongly favored in first-year ice compared to multi-year ice and, therefore, the dynamic balance between contaminant inventories and contaminant deposition to the surface ocean is being widely affected by the large-scale icescape transition taking place in the Arctic.

  • Boreal forest riparian zones regulate stream sulfate and dissolved organic carbon

    • In boreal forest catchments, solute transfer to streams is controlled by hydrological and biogeochemical processes occurring in the riparian zone (RZ). However, RZs are spatially heterogeneous and information about solute chemistry is typically limited. This is problematic when making inferences about stream chemistry. Hypothetically, the strength of links between riparian and stream chemistry is time-scale dependent. Using a ten-year (2003-2012) dataset from a northern Swedish catchment, we evaluated the suitability of RZ data to infer stream dynamics at different time scales. We focus on the role of the RZ versus upslope soils in controlling sulfate (SO42-) and dissolved organic carbon (DOC). A priori, declines in acid deposition and redox-mediated SO42- pulses control sulfur (S) fluxes and pool dynamics, which in turn affect dissolved organic carbon (DOC). We found that the catchment is currently a net source of S, presumably due to release of the S pool accumulated during the acidification period. In both, RZ and stream, SO42- concentrations are declining over time, whereas DOC is increasing. No temporal trends in SO42- and DOC were observed in upslope mineral soils. SO42- explained the variation of DOC in stream and RZ, but not in upslope mineral soil. Moreover, as SO42- decreased with time, temporal variability of DOC increased. These observations indicate that: (1) SO42- is still an important driver of DOC trends in boreal catchments and (2) RZ processes control stream SO42- and subsequently DOC independently of upslope soils. These phenomena are likely occurring in many regions recovering from acidification. Because water flows through a heterogeneous mosaic of RZs before entering the stream, upscaling information from limited RZ data to the catchment level is problematic at short-time scales. However, for long-term trends and annual dynamics, the same data can provide reasonable representations of riparian processes and support meaningful inferences about stream chemistry.     • On-site sewage treatment facilities (OSSFs), which are used to reduce nutrient emissions in rural areas, were screened for anthropogenic compounds with two-dimensional gas chromatography–mass spectrometry (GC × GC–MS). The detected compounds were prioritized based on their persistence, bioaccumulation, ecotoxicity, removal efficiency, and concentrations. This comprehensive prioritization strategy, which was used for the first time on OSSF samples, ranked galaxolide, α-tocopheryl acetate, octocrylene, 2,4,7,9-tetramethyl-5-decyn-4,7-diol, several chlorinated organophosphorus flame retardants and linear alkyl benzenes as the most relevant compounds being emitted from OSSFs. Twenty-six target analytes were then selected for further removal efficiency analysis, including compounds from the priority list along with substances from the same chemical classes, and a few reference compounds. We found significantly better removal of two polar contaminants 2,4,7,9-tetramethyl-5-decyn-4,7-diol (p = 0.0003) and tris(2-butoxyethyl) phosphate (p = 0.005) in soil beds, a common type of OSSF in Sweden, compared with conventional sewage treatment plants. We also report median removal efficiencies in OSSFs for compounds not studied in this context before, viz. α-tocopheryl acetate (96%), benzophenone (83%), 2-(methylthio)benzothiazole (64%), 2,4,7,9-tetramethyl-5-decyn-4,7-diol (33%), and a range of organophosphorus flame retardants (19% to 98%). The environmental load of the top prioritized compounds in soil bed effluents were in the thousands of nanogram per liter range, viz. 2,4,7,9-tetramethyl-5-decyn-4,7-diol (3000 ng L− 1), galaxolide (1400 ng L− 1), octocrylene (1200 ng L− 1), and α-tocopheryl acetate (660 ng L− 1).

  • In situ characterization of benthic fluxes and denitrification efficiency in a newly re-established mussel farm

    • Mussel farming has been proposed as a mechanism to mitigate eutrophication in coastal waters. However, localizing the intensive filtration of organic matter by mussels can cause a concomitant enrichment of organic matter in sediments below farms, which may influence biogeochemical processes and fates of nutrients in the system. In the context of mitigating eutrophication, it is important to quantify sedimentary changes induced at early life stages of mussel farms. Accordingly, this study investigated how a newly re-established mussel farm affected sedimentation rates, sediment characteristics, sediment-water solute fluxes and nitrate (NO3−) reduction rates (measured in situ) during the first year of production. Sedimentation rates were enhanced at the farm relative to a reference station, and both organic and inorganic carbon accumulated in the sediment with time. Increased organic matter input likely drove the slightly elevated sedimentary effluxes of ammonium (NH4+) and dissolved inorganic phosphorus (DIP) in the farm. Denitrification was the main NO3− reduction process, however, there was a relative increase in the remobilization of bioavailable nitrogen underneath the farm as dissimilatory nitrate reduction to ammonium (DNRA) rates were enhanced by >200% and the denitrification efficiency was 49% lower compared to the reference station. The sedimentary methane (CH4) release tended to be higher at the farm, but fluxes were not significantly different from reference conditions. Low sedimentary pigment concentrations indicated a reduced presence of benthic microalgae at the farm, which likely influenced sediment-water solute fluxes. Over the production cycle, the release of dissolved inorganic nitrogen (DIN) and DIP underneath the farm were respectively 426% and 510% relative to reference conditions. Impacts of the mussel farm were thus measurable already during the first year of establishment. These immediate changes to the sediment biogeochemistry, as well as long-term effects, should be considered when estimating the environmental impact of mussel aquaculture.

    • © 2019 The Authors Eutrophication is a serious threat to aquatic ecosystems globally with pronounced negative effects in the Baltic and other semi-enclosed estuaries and regional seas, where algal growth associated with excess nutrients causes widespread oxygen free “dead zones” and other threats to sustainability. Decades of policy initiatives to reduce external (land-based and atmospheric) nutrient loads have so far failed to control Baltic Sea eutrophication, which is compounded by significant internal release of legacy phosphorus (P) and biological nitrogen (N) fixation. Farming and harvesting of the native mussel species (Mytilus edulis/trossulus) is a promising internal measure for eutrophication control in the brackish Baltic Sea. Mussels from the more saline outer Baltic had higher N and P content than those from either the inner or central Baltic. Despite their relatively low nutrient content, harvesting farmed mussels from the central Baltic can be a cost-effective complement to land-based measures needed to reach eutrophication status targets and is an important contributor to circularity. Cost effectiveness of nutrient removal is more dependent on farm type than mussel nutrient content, suggesting the need for additional development of farm technology. Furthermore, current regulations are not sufficiently conducive to implementation of internal measures, and may constitute a bottleneck for reaching eutrophication status targets in the Baltic Sea and elsewhere.

  • Baltic Sea sediment records : Unlikely near-future declines in PCBs and HCB

    • We present a comprehensive study on PCBs and HCB in dated sediment cores from the Baltic Sea covering the 20th century, and compare their spatiotemporal trends with those of PCDD/Fs from the same areas. PCB concentrations in coastal impacted sediment followed the temporal trend of estimated global emissions of PCBs and thus responded quickly to changes in global industrial use, whereas concentrations in offshore sediment needed 10-20 years longer to respond. Differences in spatiotemporal trends of PCDD/Fs and PCBs were smaller than expected based on documented differences in key sources and source areas. Sediment concentrations of HCB varied little over time and space, but concentrations are increasing in recent years. The steep PCB concentration reduction over time observed for the late 20th century levelled off during the last 20 years, and levels of PCBs appear to be at or near a steady-state condition. Capsule: PCB concentrations in Baltic Sea sediments appear to be at or near steady-state, and no significant concentration decreases are to be expected in the near future.

    • High levels of PCDD/Fs and PCBs in Baltic Sea biota have been a matter of great concern during the last decades. We measured the freely dissolved concentrations of PCDD/Fs and PCBs in sediment pore water and bottom water in eight areas along the Swedish coast of the Gulf of Bothnia, by using state-of-the-art passive samplers. Chemical activity ratios (calculated from freely dissolved concentrations in pore water and bottom water based on chemical activity ratios) for PCDD/Fs were higher than 1 at all stations (PCDD/Fs average 27; stdev 22). High activity ratios suggest that the sediments have a potential to act as a source of dissolved PCDD/Fs to the water column. Activity ratios for PCBs varied between 0.3 and 17 (average 2; stdev 4). The concentrations of PCDD/Fs and PCBs in bottom water were significantly correlated with concentrations in sediment pore water (p < 0.00001 to p = 0.03) as well as with concentrations in juvenile perch caught in the same areas (p < 0.00001 to p = 0.02). To our knowledge, this is the first study demonstrating a correlation between in-situ measured freely dissolved PCDD/F concentrations and lipid-normalized contents in stationary fish. Our results confirm that freely dissolved concentrations should be used as chemical predictors of bioaccumulation. The results from this study imply that continued efforts to reduce levels of PCDD/Fs and PCBs in coastal sediments will have positive effects on concentrations of these contaminants in lower trophic levels of Baltic Sea ecosystems.
