    • Multicentre patch test studies (MPTSs) can contribute useful information for diagnostic and preventive measures. The aim of the present paper is to propose how to perform high-quality MPTSs. To this end, factors of significance for the patch test result are discussed with regard to the standardization and calibration of high-quality MPTSs. The 16 factors discussed are scored 0, 1, 2, or 3, depending on the relative importance of a particular factor for the patch test result. The total score of an MPTS allows it to be ranked as having doubtful, acceptable, high or excellent quality. A total score of 30 is possible. Depending on the total score the MPTSs are grouped into those with a doubtful, acceptable, high, and excellent quality. In conclusion, high-quality MPTSs can be performed and are facilitated if a guideline and check list are followed when the study is being planned. The scoring enables the calculation of a total score, which can be used for quality ranking.

A brief summary of related papers published in the literature within the areas of skin irritant and allergy patch testing, microdialysis and skin tumour circulation is presented, as well as early applications within other fields such as diabetology, wound healing and microvascular research.

    • Cases of allergic contact dermatitis due to chromium caused by chromium‐tanned leather articles such as shoes and gloves are well‐known.1, 2 In the European Union, hexavalent chromium in new leather articles on the market is restricted to 3 mg/kg since 2015.3 A recent use‐test study showed that also trivalent chromium, the dominating form of chromium released from new leather articles,4 can elicit allergic reactions in chromium‐allergic individuals.5 For leather articles that are used for a prolonged period, data on the released chromium form are scarce, but one long‐term study indicates that the fraction of hexavalent chromium increases with duration of use.6

  • Imaging mass spectrometry for novel insights into contact allergy - a proof-of-concept study on nickel

    • Background. In spite of extensive regulation to limit exposure, nickel remains the main cause of contact allergy in the general population. More detailed knowledge on the skin uptake of haptens is required. So far, no method exists for the visualization of this clinically relevant hapten and its distribution in the skin. Objectives. To show, in terms of a proof of concept, that imaging mass spectrometry [time of flight secondary ion mass spectrometry (ToF-SIMS)] can be applied for investigation of the penetration and distribution of nickel in human skin. Method. Full-thickness human skin obtained from breast reduction surgery was exposed to nickel sulfate (5% in deionized water) for 24 h in Franz-type diffusion cells. Biopsies were obtained from nickel-treated samples and control (deionized water). The tissue was sliced, and analysed with ToF-SIMS, generating high-resolution images of ion distribution in the epidermis and upper dermis. Results. The skin layers could be discerned from the ToF-SIMS data, particularly on the basis of the collagen signal. Nickel ions were localized to the stratum corneum and upper epidermis. Conclusions. This is the first time that ToF-SIMS has been applied to trace the distribution of a hapten in human skin. Proof of principle was shown for nickel, and the technique can, in the future, be expanded for investigation of the skin distribution of clinically relevant sensitizers in general.
