  • A systematic analysis of commonly used antibodies in cancer diagnostics

    • AimsImmunohistochemistry plays a pivotal role in cancer differential diagnostics. To identify the primary tumour from a metastasis specimen remains a significant challenge, despite the availability of an increasing number of antibodies. The aim of the present study was to provide evidence-based data on the diagnostic power of antibodies used frequently for clinical differential diagnostics. Methods and resultsA tissue microarray cohort comprising 940 tumour samples, of which 502 were metastatic lesions, representing tumours from 18 different organs and four non-localized cancer types, was analysed using immunohistochemistry with 27 well-established antibodies used in clinical differential diagnostics. Few antibodies, e.g. prostate-specific antigen and thyroglobulin, showed a cancer type-related sensitivity and specificity of more than 95%. A majority of the antibodies showed a low degree of sensitivity and specificity for defined cancer types. Combinations of antibodies provided limited added value for differential diagnostics of cancer types. ConclusionsThe results from analysing 27 diagnostic antibodies on consecutive sections of 940 defined tumours provide a unique repository of data that can empower a more optimal use of clinical immunohistochemistry. Our results highlight the benefit of immunohistochemistry and the unmet need for novel markers to improve differential diagnostics of cancer.

  • Slide navigation patterns among pathologists with long experience of digital review

    • Aims: In order to develop efficient digital pathologyworkstations, we studied the navigation patterns ofpathologists diagnosing whole-slide images. To gain abetter understanding of these patterns, we built aconceptual model based on observations. We alsodetermined whether or not new navigation patternshave emerged among pathologists with extensive digitalexperience.Methods and results: Five pathologists were asked todiagnose a set of four cases while thinking out loud.The navigation within the digital slides was recordedand divided into re-occurring navigation actions. Thepathologists reused the same type of actions, but theiroccurrence differed. The most common action was aslow panning that followed an edge structure orcovered an area systematically, which accounted for30.2% of all actions and had a median duration of7.2 s. Of all the actions, 49% were carried out withinthe navigation overview and 38% of the actionscould not have been performed with a conventionalmicroscope.Conclusions: The new navigation possibilities in thedigital workstation were used to a large extent. Thedivision of actions into different concepts can be usedto find and prioritize between existing user interfacedesigns as well as to understand the different navigationstyles used by different pathologists.

    • Two species of arbovirus vector, Culex torrentium and Culex pipiens (Diptera: Culicidae), occur in several European countries, but difficulties in their accurate identification and discrimination have hampered both detailed and large-scale distribution and abundance studies. Using a molecular identification method, we identified to species 2559 larvae of Cx. pipiens/torrentium collected from 138 sites in 13 European countries ranging from Scandinavia to the Mediterranean coast. In addition, samples of 1712 males of Cx. pipiens/torrentium collected at several sites in the Czech Republic were identified to species based on the morphology of their hypopygia. We found that the two species occur together in large areas of Europe, and that Cx. torrentium dominates in northern Europe and Cx. pipiens dominates south of the Alps. The transition in dominance occurs in central Europe, where both species are roughly equally common. There was a strong correlation between the length of the growing season at different sites and occurrences of the two species. As the growing season increases, the proportion and detection of Cx. torrentium decrease, whereas those of Cx. pipiens increase. The present findings have important consequences for the interpretation of the results of studies on major enzootic and link-vectors of mosquito-borne bird-associated viruses (i.e. Sindbis, West Nile and Usutu viruses), especially in central Europe and Scandinavia.
