- We demonstrate a technique to improve structural data obtained from Optical Projection Tomography (OPT) using Image Fusion (IF) and contrast normalization. This enables the visualization of molecular expression patterns in biological specimens with highly variable contrast values. In the approach, termed IF-OPT, different exposures are fused by assigning weighted contrasts to each. When applied to projection images from mouse organs and digital phantoms our results demonstrate the capability of IF-OPT to reveal high and low signal intensity details in challenging specimens. We further provide measurements to highlight the benefits of the new algorithm in comparison to other similar methods.

A low-absorption x-ray energy filter for small-scale applications

- We present an experimental and theoretical evaluation of an x-ray energy filter based on the chromatic properties of a prism-array lens (PAL). It is intended for small-scale applications such as medical imaging. The PAL approximates a Fresnel lens and allows for high efficiency compared to filters based on ordinary refractive lenses, however at the cost of a lower energy resolution. Geometrical optics was found to provide a good approximation for the performance of a flawless lens, but a field-propagation model was used for quantitative predictions. The model predicted a 0.29 ΔE/E energy resolution and an intensity gain of 6.5 for a silicon PAL at 23.5 keV. Measurements with an x-ray tube showed good agreement with the model in energy resolution and peak energy, but a blurred focal line contributed to a 29% gain reduction. We believe the blurring to be caused mainly by lens imperfections, in particular at the periphery of the lens.

ICL-based TDLAS sensor for real-time breath gas analysis of carbon monoxide isotopes

- We present a compact sensor for carbon monoxide (CO) in air and exhaled breath based on a room temperature interband cascade laser (ICL) operating at 4.69 µm, a low-volume circular multipass cell and wavelength modulation absorption spectroscopy. A fringe-limited (1σ) sensitivity of 6.5 × 10−8 cm−1Hz-1/2 and a detection limit of 9 ± 5 ppbv at 0.07 s acquisition time are achieved, which constitutes a 25-fold improvement compared to direct absorption spectroscopy. Integration over 10 s increases the precision to 0.6 ppbv. The setup also allows measuring the stable isotope 13CO in breath. We demonstrate quantification of indoor air CO and real-time detection of CO expirograms from healthy non-smokers and a healthy smoker before and after smoking. Isotope ratio analysis indicates depletion of 13CO in breath compared to natural abundance.

- For low-order adaptive optics systems, a method that is able to correct for system aberrations in the final focal plane is presented. The paper presents a novel figure of merit, corresponding to the integrated non-normalized tip-tilt-free optical transfer function. The inherent singular value decomposition modal content of the interaction matrix is used to optimize this figure of merit. The method has proven to be stable and robust, providing a simple mean to facilitate diffraction limited imaging in an experimental setup for ophthalmic applications. Fast-tracking of single emitters in large volumes with nanometer precision

- Multifocal plane microscopy allows for capturing images at different focal planes simultaneously. Using a proprietary prism which splits the emitted light into paths of different lengths, images at 8 different focal depths were obtained, covering a volume of 50x50x4 mu m(3). The position of single emitters was retrieved using a phasor-based approach across the different imaging planes, with better than 10 nm precision in the axial direction. We validated the accuracy of this approach by tracking fluorescent beads in 3D to calculate water viscosity. The fast acquisition rate (>100 fps) also enabled us to follow the capturing of 0.2 mu m fluorescent beads into an optical trap. Dual-conjugate adaptive optics for wide-field high-resolution retinal imaging.

- We present analysis and preliminary laboratory testing of a real-time dual-conjugate adaptive optics (DCAO) instrument for ophthalmology that will enable wide-field high resolution imaging of the retina in vivo. The setup comprises five retinal guide stars (GS) and two deformable mirrors (DM), one conjugate to the pupil and one conjugate to a plane close to the retina. The DCAO instrument has a closed-loop wavefront sensing wavelength of 834 nm and an imaging wavelength of 575 nm. It incorporates an array of collimator lenses to spatially filter the light from all guide stars using one adjustable iris, and images the Hartmann patterns of multiple reference sources on a single detector. Zemax simulations were performed at 834 nm and 575 nm with the Navarro 99 and the Liou- Brennan eye models. Two correction alternatives were evaluated; conventional single conjugate AO (SCAO, using one GS and a pupil DM) and DCAO (using multiple GS and two DM). Zemax simulations at 575 nm based on the Navarro 99 eye model show that the diameter of the corrected field of view for diffraction-limited imaging (Strehl >or= 0.8) increases from 1.5 deg with SCAO to 6.5 deg using DCAO. The increase for the less stringent condition of a wavefront error of 1 rad or less (Strehl >or= 0.37) is from 3 deg with SCAO to approximately 7.4 deg using DCAO. Corresponding results for the Liou-Brennan eye model are 3.1 deg (SCAO) and 8.2 deg (DCAO) for Strehl >or= 0.8, and 4.8 deg (SCAO) and 9.6 deg (DCAO) for Strehl >or= 0.37. Potential gain in corrected field of view with DCAO is confirmed both by laboratory experiments on a model eye and by preliminary in vivo imaging of a human eye.

Non-binary phase gratings for x-ray imaging with a compact Talbot interferometer

- X-ray imaging using a Talbot-Lau interferometer, consisting of three binary gratings, is a well-established approach to acquire x-ray phase-contrast and dark-field images with a polychromatic source. However, challenges in the production of high aspect ratio gratings limit the construction of a compact setup for high x-ray energies. In this study we consider the use of phase gratings with triangular-shaped structures in an x-ray interferometer and show that such gratings can yield high visibilities for significantly shorter propagation distances than conventional gratings with binary structures. The findings are supported by simulation and experimental results for both cases of a monochromatic and a polychromatic source.
