  • Strong impact of micropollutants on prokaryotic communities at the horizontal but not vertical scales in a subtropical reservoir, China

    • Micropollutants have become of great concern, because of their disrupting effects on the structure and function of microbial communities. However, little is known about the relative importance of trace micropollutants on the aquatic prokaryotic communities as compared to the traditional physico-chemical characteristics, especially at different spatial dimensions. Here, we investigated free-living (FL) and particle-associated (PA) prokaryotic communities in a subtropical water reservoir, China, across seasons at horizontal (surface water) and vertical (depth-profile) scales by using 16S rRNA gene amplicon sequencing. Our results showed that the shared variances of physico-chemicals and micropollutants explained majority of the spatial variations in prokaryotic communities, suggesting a strong joint effect of the two abiotic categories on reservoir prokaryotic communities. Micropollutants appeared to exert strong independent influence on the core sub-communities (i.e., abundant and wide-spread taxa) than on the satellite (i.e., less abundant and narrow-range taxa) counterparts. The pure effect of micropollutants on both core and satellite sub-communities from FL and PA fractions was similar to 1.5 folds greater than that of physico-chemical factors at the horizontal scale, whereas an opposite effect was observed at the vertical scale. Moreover, eight micropollutants including anti-fungal agents, antibiotics, bisphenol analogues, stimulant and UV-filter were identified as the major disrupting compounds with strong associations with core taxa of typical freshwater prokaryotes. Altogether, we concluded that the ecological disrupting effects of micropollutants on prokaryotic communities may vary along horizontal and vertical dimensions in freshwater ecosystems.

    • Anthropogenic inputs into the environment may serve as sources of antimicrobial resistant bacteria and alter the ecology and population dynamics of synanthropic wild animals by providing supplemental forage. In this study, we used a combination of phenotypic and genomic approaches to characterize antimicrobial resistant indicator bacteria, animal telemetry to describe host movement patterns, and a novel modeling approach to combine information from these diverse data streams to investigate the acquisition and long-distance dispersal of antimicrobial resistant bacteria by landfill-foraging gulls. Our results provide evidence that gulls acquire antimicrobial resistant bacteria from anthropogenic sources, which they may subsequently disperse across and between continents via migratory movements. Furthermore, we introduce a flexible modeling framework to estimate the relative dispersal risk of antimicrobial resistant bacteria in western North America and adjacent areas within East Asia, which may be adapted to provide information on the risk of dissemination of other organisms and pathogens maintained by wildlife through space and time.   • First detection of SARS-CoV-2 genetic material in the vicinity of COVID-19 isolation Centre in Bangladesh : Variation along the sewer network

    • We made the first and successful attempt to detect SARS-CoV-2 genetic material in the vicinity wastewaters of an isolation centre i.e. Shaheed Bhulu Stadium, situated at Noakhali, Southeastern Bangladesh. Owing to the fact that isolation centre, in general, always contained a constant number of 200 COVID-19 patients, the prime objective of the study was to check if several drains carrying RNA of coronavirus are actually getting diluted or accumulated along with the sewage network. Our finding suggested that while the temporal variation of the genetic load decreased in small drains over the span of 50 days, the main sewer exhibited accumulation of SARS-CoV-2 RNA. Other interesting finding displays that probably distance of sampling location in meters is not likely to have a significant impact on the detected gene concentration, although the quantity of the RNA extracted in the downstream of the drain was higher. These findings are of immense value from the perspective of wastewater surveillance of COVID-19, as they largely imply that we do not need to monitor every wastewater system, and probably major drains monitoring may illustrate the city health. Perhaps, we are reporting the accumulation of SARS-CoV-2 genetic material along with the sewer network i.e. from primary to tertiary drains. The study sought further data collection in this line to simulate conditions prevailed in most of the developing countries and to shed further light on decay/accumulation processes of the genetic load of the SARS-COV-2.

    • Climate change is the variability of the climate system that includes the atmosphere, the biogeochemical cycles (Carbon cycle, Nitrogen cycle and Hydrological cycle), the land surface, ice and the biotic and abiotic components of the planet earth. Significant impact of climate change is seen in the form of rise in temperature called as global warming. Carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) are the primary greenhouse gases (GHGs) mainly responsible for the global warming and climate change. These GHGs have drawn lot of attention due to their significant role in the global warming potential Intergovernmental Panel on Climate Change (IPCC) suggested to stop global warming at 1.5oC above preindustrial levels as warming beyond this level might lead to heat extremes, alter insect and plant phenology (Phenological shifts) and more occurrence of vector borne diseases. Climate change is the topic of interest in all fields of life starting from social science and going to the applied science. Global climate cycles and world food production systems are under threat due to the recent climate extreme events. These events include heat waves and change in the rainfall patterns. Thus, risk reduction intervention in the form of mitigation and adaptation is required to minimize the impacts of climate change. Mitigation option includes understanding the present and future components of the climate system and interaction among them through coupled modeling systemic. Global Circulation Model (GCM), finally, global issue of climate change could be addressed by taking worldwide cooperation and action and adopting sustainable measures like use of alternative energy sources.   • Phenotyping for drought resistance in bread wheat using physiological and biochemical traits

    • Drought is one of the most prominent limiting factors that negatively affect crop productivity by manipulating its physiological pathway. One hundred twenty diverse bread wheat genotypes were used in a pot experiment to explore the relationship among their fifteen physio-biochemical traits (PBT) by using multivariate analysis, heatmapping and stress tolerance index (STI) for grain yield as a marker trait to identify high yielding genotype with maximum stress tolerance capability. Increased proline and sugar accumulation were observed from control to moisture deficient environments by 159% and 122%, respectively. Moreover, leaf membrane stability index (LMSI), leaf relative water content (LRWC), relative dry weight (RDW), chlorophyll content, leaf surface area (LSA), Leaf succulence (LS), canopy temperature depression (CTD), relative excised leaf water loss (RELWL) and leaf osmotic potential (LOP) showed significantly decreasing trend in drought stress treatment as compared to well-watered plants by −21%, −21%, −34%, −22%, −38%, −37%, −46%, −18% and −35% respectively. Additionally, principal component analysis and genotype by trait biplot analysis showed that initial 7 principal components (PC1 to PC7) represented 77.27% and 79.02% of total cumulative variation under control and drought stress respectively. Genotypic-Phenotypic correlation revealed that most of the attributes were higher in case of genotypic correlation component (rg) as compared to the phenotypic correlation component (rp) indicating more genetic association between traits. The darker and lighter colour scale produced by heatmap exhibited contrasting nature of genotypes, as positive side with higher values represented drought resistance while values on the negative side with lower values showed susceptible performance of genotypes. Our results concluded that the studied PBT associated with STI for grain yield are the main factors which may contribute in improved productivity of wheat crop and if these traits show appropriate performance under stress condition the crop will show the more productive returns under changing climate.

  • Optimization of state-of-the-art fuzzy-metaheuristic ANFIS-based machine learning models for flood susceptibility prediction mapping in the Middle Ganga Plain, India

    • This study is an attempt to quantitatively test and compare novel advanced-machine learning algorithms in terms of their performance in achieving the goal of predicting flood susceptible areas in a low altitudinal range, sub-tropical floodplain environmental setting, like that prevailing in the Middle Ganga Plain (MGP), India. This part of the Ganga floodplain region, which under the influence of undergoing active tectonic regime related subsidence, is the hotbed of annual flood disaster. This makes the region one of the best natural laboratories to test the flood susceptibility models for establishing a universalization of such models in low relief highly flood prone areas. Based on highly sophisticated flood inventory archived for this region, and 12 flood conditioning factors viz. annual rainfall, soil type, stream density, distance from stream, distance from road, Topographic Wetness Index (TWI), altitude, slope aspect, slope, curvature, land use/land cover, and geomorphology, an advanced novel hybrid model Adaptive Neuro Fuzzy Inference System (ANFIS), and three metaheuristic models-based ensembles with ANFIS namely ANFIS-GA (Genetic Algorithm), ANFIS-DE (Differential Evolution), and ANFIS-PSO (Particle Swarm Optimization), have been applied for zonation of the flood susceptible areas. The flood inventory dataset, prepared by collected flood samples, were apportioned into 70:30 classes to prepare training and validation datasets. One independent validation method, the Area-Under Receiver Operating Characteristic (AUROC) Curve, and other 11 cut-off-dependent model evaluation metrices have helped to conclude that the ANIFS-GA has outperformed other three models with highest success rate AUC = 0.922 and prediction rate AUC = 0.924. The accuracy was also found to be highest for ANFIS-GA during training (0.886) & validation (0.883). Better performance of ANIFS-GA than the individual models as well as some ensemble models suggests and warrants further study in this topoclimatic environment using other classes of susceptibility models. This will further help establishing a benchmark model with capability of highest accuracy and sensitivity performance in the similar topographic and climatic setting taking assumption of the quality of input parameters as constant.

    • Even though production and open use of polychlorinated biphenyls (PCBs) have been phased out in Western industrialised countries since the 1980s, PCBs were still present in waste collected from different waste handling facilities in Norway in 2013. Sums of seven indicator-PCBs (I-PCB7:PCB-28, -52, -101, -118, -138, -153 and -180) were highest in plastic waste (3700 +/- 1800 mu g/kg, n=15), waste electrical and electronic equipment (WEEE) (1300 +/- 400 mu g/kg, n=12) and fine vehicle fluff (1800 = 1400 mu g/kg, n=4) and lowest in glass waste, combustibles, bottom ash and fly ash (0.3 to 65 mu g/kg). Concentrations in leachate water varied from 1.7 to 2900 ng/L, with higher concentrations found at vehicle and WEEE handling facilities. Particles in leachate water exhibited similar PCB sorption properties as solid waste collected on site, with waste-water partitioning coefficients ranging from 10(5)to 10(7) .I-PCB7 in air samples collected at the sites were mostly in the gas phase (100-24000 pg/m(3)), compared to those associated with particles (9-1900 pg/m(3)). In contrast, brominated flame retardants (BFRs) in the same samples were predominantly found associated with particles (e.g. sum of 10 brominated diethyl ethers, Sigma BDE10, associated with particles 77-194,000 pg/m(3)) compared to the gas phase (Sigma DE10 6-473 pg/m(3)). Measured gas-phase I-PCB7 concentrations are less than predicted, assuming waste-air partitioning in equilibrium with predominant waste on site. However, the gas-particle partitioning behavior of PCBs and BFRs could be predicted using an established partitioning model for ambient aerosols. PCB emissions from Norwegian waste handling facilities occurred primarily in the form of atmospheric vapor or leachate particles.

    • Metallic trace elements (MTEs) soil pollution has become a worldwide concern, particularly regarding its impact on earthworms. Earthworms, which constitute the dominant taxon of soil macrofauna in temperate regions and are crucial ecosystem engineers, are in direct contact with MTEs. The impacts of MTE exposure on earthworms, however, vary by species, with some able to cope with high levels of contamination. We combined different approaches to study the effects of MTEs at different levels of biological organisation of an earthworm community, in a contaminated urban wasteland. Our work is based on field collection of soil and earthworm samples, with a total of 891 adult earthworms from 8 species collected, over 87 quadrats across the study plot. We found that MTE concentrations are highly structured at the plot scale and that some elements, such as Pb, Zn, and Cu, are highly correlated. Comparing species assemblage to MTE concentrations, we found that the juvenile and adult abundances, and community composition, were significantly affected by pollution. Along the pollution gradient, as species richness decreased, Lumbricus castaneus became more dominant. We thus investigated the physiological response of this species to a set of specific elements (Pb, Zn, Cu, and Cd) and studied the impacts of MTE concentrations at the plot scale on its population genetic. These analyses revealed that L. castaneus is able to bioaccumulate high quantities of Cd and Zn, but not of Cu and Pb. The population genetic analysis, based on the genotyping of 175 individuals using 8 microsatellite markers, provided no evidence of the role of the heterogeneity in MTE concentrations as a barrier to gene flow. The multidisciplinary approach we used enabled us to reveal the comparatively high tolerance of L. castaneus to MTE concentrations, suggesting that this is a promising model to study the molecular bases of MTE tolerance.

    • Elevated Arsenic (As) and Fluoride (F) concentrations in groundwater have been studied in the shallow aquifers of northeastern of La Pampa province, in the Chaco-Pampean plain, Argentina. The source of As and co-contaminants is mainly geogenic, from the weathering of volcanic ash and loess (rhyolitic glass) that erupted from the Andean volcanic range. In this study we have assessed the groundwater quality in two semi-arid areas of La Pampa. We have also identified the spatial distribution of As and co-contaminants in groundwater and determined the major factors controlling the mobilization of As in the shallow aquifers. The groundwater samples were circum-neutral to alkaline (7.4 to 92), oxidizing (Eh similar to 0.24 V) and characterized by high salinity (EC = 456-11,400 mu S/cm) and Na+-HCO3- water types in recharge areas. Carbonate concretions ("tosca") were abundant in the upper layers of the shallow aquifer. The concentration of total As (5.6 to 535 mu g/L) and F (0.5 to 14.2 mg/L) were heterogeneous and exceeded the recommended WHO Guidelines and the Argentine Standards for drinking water. The predominant As species were arsenate As(V) oxyanions, determined by thermodynamic calculations. Arsenic was positively correlated with bicarbonate (HCO3-), fluoride (F), boron (B) and vanadium (V), but negatively correlated with iron (Fe), aluminium (Al), and manganese (Mn), which were present in low concentrations. The highest amount of As in sediments was from the surface of the dry lake. The mechanisms for As mobilization are associated with multiple factors: geochemical reactions, hydrogeological characteristics of the local aquifer and climatic factors. Desorption of As(V) at high pH, and ion competition for adsorption sites are considered the principal mechanisms for As mobilization in the shallow aquifers. In addition, the long-term consumption of the groundwater could pose a threat for the health of the local community and low cost remediation techniques are required to improve the drinking water quality.

    • Information on how key environmental conditions such as natural dissolved organic matter (DOM) and water pH alter the possible risks posed by pharmaceuticals (PPCPs) is still scarce. In our previous study, the presence of nat-ural DOM at high pH reduced the toxicity of a mix of waterborne PPCPs to algae. DOM-complexation and pH ef-fect on speciation of the more hydrophobic and neutral compounds of the mix was suggested to be driving this behaviour. However, the study design did not allow the verification of this hypothesis. Here, the DOM-PPCPs in-teraction at different pH was investigated for 6 PPCPs through equilibrium dialysis, under the same conditions of DOM and pH as our previous study. Association with DOM was confirmed for the more hydrophobic PPCPs at high pH. The results suggest the binding was driven by i) the presence of carboxylic groups of PPCPs, ii) high pH shifting the structural configuration of DOM, making it more suited to bind some of the PPCPs. A non-linear change of binding capacity with increasing DOM concentration was also observed among the tested PPCPs. (c) 2021 The Author(s). Published by Elsevier B.V.
