  • Chronic development of collagen-induced arthritis is associated with arthritogenic antibodies against specific epitopes on type II collagen

    • Antibodies against type II collagen (CII) are important in the development of collagen-induced arthritis (CIA) and possibly also in rheumatoid arthritis. We have determined the fine specificity and arthritogenicity of the antibody response to CII in chronic relapsing variants of CIA. Immunization with rat CII in B10.Q or B10.Q(BALB/cxB10.Q)F2 mice induces a chronic relapsing CIA. The antibody response to CII was determined by using triple-helical peptides of the major B cell epitopes. Each individual mouse had a unique epitope-specific response and this epitope predominance shifted distinctly during the course of the disease. In the B10.Q mice the antibodies specific for C1 and U1, and in the B10.Q(BALB/cxB10.Q)F2 mice the antibodies specific for C1, U1 and J1, correlated with the development of chronic arthritis. Injection of monoclonal antibodies against these epitopes induced relapses in chronic arthritic mice. The development of chronic relapsing arthritis, initially induced by CII immunization, is associated with an arthritogenic antibody response to certain CII epitopes.

  • Predator-induced spine length and exocuticle thickness in Leucorrhinia dubia (Insecta : Odonata): a simple physiological trade-off?

    • 1. Morphological defence structures evolve against predators but are costly to the individual, and are induced only when required. A well-studied example is the development of longer abdominal spines in dragonfly larvae in the presence of fish. Numerous attempts to discover trade-offs between spine size and behaviour, development time or body size have, however, produced little evidence. 2. We considered a physiological trade-off. Spines consist of cuticle and using material to build longer structures may result in less material remaining elsewhere. We therefore measured exocuticle thickness at nine locations on Leucorrhinia dubia larvae from habitats with and without fish. 3. Our results show a significant effect of the interaction between fish presence and spine length on head and fore leg exocuticle thickness. Relative thickness increased with relative length of lateral spine 9 in the absence of fish, whereas no such relationship existed with fish. Hence, synthesis and secretion of cuticle material occur as a trade-off when larvae react to fish presence. 4. We assume the mechanism to be a selective synthesis of material with different responses in different parts of the larval body. These findings offer a new angle to the fish/spine trade off debate.

  • The usefulness of a multidisciplinary educational programme after breast cancer surgery : A prospective and comparative study

    • The aim of the study was to compare and evaluate a multidisciplinary educational programme with traditional follow-up visits to a physician after breast cancer surgery in terms of well-being, aspects of self-care and coping ability 1 year after diagnosis. A reduction in the intensity of follow-up after breast cancer surgery is recommended. New follow-up models are being debated and could be of interest. The study design was non-randomised and comparative. Ninety-six consecutively selected women with newly diagnosed breast cancer, classified as stage I or stage II, participated in either a multidisciplinary educational programme (n = 5 0), or traditional follow-up by a physician (n = 4 6). Three questionnaires were used: Functional Assessment of Cancer Therapy-General (FACT-G), a study specific questionnaire regarding self-care aspects (SCA) and Sense of Coherence (SOC). With the exception of physical well-being at baseline there was no significant difference between the groups. The women in the multidisciplinary educational programme increased their physical and functional well-being (P < 0.0 1). The women in traditional follow-up by a physician increased their functional well-being while social/family well-being (P < 0.0 1) decreased over time. There was a statistically significant difference in SOC (P < 0.0 0 1) in the traditional follow-up by a physician between baseline (mean=74.4, SD=12.4) and the 1-year follow up (mean=67.7, SD=11.4). Thus, women in the traditional follow-up by a physician scored lower in the area of SOC 1 year after diagnosis. A multidisciplinary educational programme may be an alternative to traditional follow-up by a physician after breast cancer surgery, but more research is needed about the financial benefits and effectiveness of such a programme.

    • Costs and limits are assumed to be the major constraints on the evolution of phenotypic plasticity. However, despite their expected importance, they have been surprisingly hard to find in natural populations. It has therefore been argued that natural selection might have removed high-cost genotypes in all populations. However, if costs of plasticity are linked to the degree of plasticity expressed, then high costs of plasticity would only be present in populations where increased plasticity is under selection. We tested this hypothesis by investigating costs and limits of adaptive phenotypic plasticity in development time in a common garden study of island populations of the common frog Rana temporaria, which have varying levels of development time and phenotypic plasticity. Costs of plasticity were only found in populations with high-plastic genotypes, whereas the populations with the most canalized genotypes instead had a cost of canalization. Moreover, individuals displaying the most extreme phenotypes also were the most plastic ones, which mean we found no limits of plasticity. This suggests that costs of plasticity increase with increased level of plasticity in the populations, and therefore costs of plasticity might be more commonly found in high-plastic populations.

    • The developmental threshold is the minimum size or condition that a developing organism must have reached in order for a life-history transition to occur. Although developmental thresholds have been observed for many organisms, inter-population variation among natural populations has not been examined. Since isolated populations can be subjected to strong divergent selection, population divergence in developmental thresholds can be predicted if environmental conditions favour fast or slow developmental time in different populations. Amphibian metamorphosis is a well-studied life-history transition, and using a common garden approach we compared the development time and the developmental threshold of metamorphosis in four island populations of the common frog Rana temporaria: two populations originating from islands with only temporary breeding pools and two from islands with permanent pools. As predicted, tadpoles from time-constrained temporary pools had a genetically shorter development time than those from permanent pools. Furthermore, the variation in development time among females from temporary pools was low, consistent with the action of selection on rapid development in this environment. However, there were no clear differences in the developmental thresholds between the populations, indicating that the main response to life in a temporary pool is to shorten the development time.

    • Although theoretical models have identified environmental heterogeneity as a prerequisite for the evolution of adaptive plasticity, this relationship has not yet been demonstrated experimentally. Because of pool desiccation risk, adaptation of development rate is important for many amphibians. In a simulated pool-drying experiment, we compared the development time and phenotypic plasticity in development time of populations of the common frog Rana temporaria, originating from 14 neighbouring islands off the coast of northern Sweden. Drying regime of pools used by frogs for breeding differed within and among the islands. We found that the degree of phenotypic plasticity in development time was positively correlated with the spatial variation in the pool-drying regimes present on each island. In addition, local adaptation in development time to the mean drying rate of the pools on each island was found. Hence, our study demonstrates the connection between environmental heterogeneity and developmental plasticity at the island population level, and also highlights the importance of the interplay between local specialization and phenotypic plasticity depending on the local selection pressures.

  • Scaling of skeletal muscle shortening velocity in mammals representing a 100,000-fold difference in body size

    • To fully understand the effect of scaling on skeletal muscle shortening velocity (V 0), it is important to know which phenotypic characteristics drive the changes between species. The purpose of the current investigation was to compare the effects of body mass and femur length, as an estimate of total limb length, on V 0 in species that cover a 100,000-fold range of body masses. Using the slack test procedure, V 0 was determined for fibers expressing types I and IIa myosin heavy chain (MyHC) isoforms in the mouse, rat, dog, human, horse, and rhinoceros under identical experimental conditions. A significant scaling effect on V 0 was detected when compared to body mass (type I fibers, r=0.95, p<0.01; type IIa fibers, r=0.83, p<0.05). However, the horse's V 0 for both fiber types was faster than the human's, despite having a 5-fold greater body mass than the human. When V 0 was scaled vs limb length, the strength of the relationships improved in fibers expressing both types I and IIa MyHC (r=0.98, p<0.001, and r=0.89, p<0.05, respectively) and scaled with the expected relationship, with the species with the shorter femur, the horse, having the faster V 0. A similar effect can be seen with stride frequency scaling more closely with limb length than body mass. These results suggest that limb length, not body mass, is a more relevant factor driving the scaling effect on skeletal muscle shortening velocity.
