  • Affinity binding of inclusion bodies on supermacroporous monolithic cryogels using labeling with specific antibodies

    • A new chromatographic method based on affinity supermacroporous monolithic cryogels is developed for binding and analyzing inclusion bodies during fermentation. The work demonstrated that it is possible to bind specific IgG and IgY antibodies to the 15 and 17 amino acids at the terminus ends of a 33 kDa target protein aggregated as inclusion bodies. The antibody treated inclusion bodies from lysed fermentation broth can be specifically retained in protein A and pseudo-biospecific ligand sulfamethazine modified supermacroporous cryogels. The degree of binding of IgG and IgY treated inclusion bodies to the Protein A and sulfamethazine gels are investigated, as well as the influence of pH on the sulfamethazine ligand. Optimum binding of 78 and 72% was observed on both protein A and sulfamethazine modified cryogel columns, respectively, using IgG labeling of the inclusion bodies. The antibody treated inclusion bodies pass through unretained in the sulfamethazine supermacroporous gel at pH that does not favour the binding between the ligand on the gel and the antibodies on the surface of inclusion bodies. Also the unlabeled inclusion bodies went through the gel unretained, showing no non-specific binding or trapping within the gel. These findings may very well be the foundation for the building of a powerful analytical tool during fermentation of inclusion bodies as well as a convenient way to purify them from fermentation broth. These results also support our earlier findings [Kumar, A., Plieva, F.M., Galaev, I.Yu., Mattiasson, B.. 2003. Affinity fractionation of lymphocytes using a monolithic cyogel. J. Immunol. Methods 283, 185-194] with mammalian cells that were surface labeled with specific antibodies and recognized on protein A supermacroporous gels. A general binding and separation system can be established on antibody binding cryogel affinity matrices.

    • The growth of iron phthalocyanine (FePc) on InSb(001) c(8 x 2) at submonolayer coverage has been investigated with scanning tunneling microscopy (STM). FePc adsorbs flat centered on the In rows both at 70 K and at room temperature (RT). However, the shapes of the two-dimensional molecular islands are fundamentally different; while the RT growth results in chainlike structures along the [I 10] direction, as already observed for other Pc's adsorbed on the same surface, the islands are prolonged along [110], i.e., perpendicular to the substrate rows, at 70 K. These observations are explained on the basis of a recently observed new surface phase at low temperature, resulting in structural domains on the surface. The molecular growth front follows the propagating domain boundary that freezes at low temperature.

  • Institutional trust and alcohol consumption in Sweden: The Swedish National Public Health Survey 2006

    • Background: Trust as a measure of social capital has been documented to be associated with health. Mediating factors for this association are not well investigated. Harmful alcohol consumption is believed to be one of the mediating factors. We hypothesized that low social capital defined as low institutional trust is associated with harmful alcohol consumption. Methods: Data from the 2006 Swedish National Survey of Public Health were used for analyses. The total study population comprised a randomly selected representative sample of 26.305 men and 30.584 women aged 16-84 years. Harmful alcohol consumption was measured using a short version the Alcohol Use Disorders Identification Test (AUDIT), developed and recommended by the World Health Organisation. Low institutional trust was defined based on trust in ten main welfare institutions in Sweden. Results: Independent of age, country of birth and socioeconomic circumstances, low institutional trust was associated with increased likelihood of harmful alcohol consumption (OR (men) = 1.52, 95% CI 1.34-1.70) and (OR (women) = 1.50, 95% CI 1.35-1.66). This association was marginally altered after adjustment for interpersonal trust. Conclusion: Findings of the present study show that lack of trust in institutions is associated with increased likelihood of harmful alcohol consumption. We hope that findings in the present study will inspire similar studies in other contexts and contribute to more knowledge on the association between institutional trust and lifestyle patterns. This evidence may contribute to policies and strategies related to alcohol consumption.

  • Gender differences in daily smoking prevalence in different age strata: A population-based study in southern Sweden.

    • Objectives: To investigate gender differences in daily smoking prevalence in different age groups in southern Sweden. Methods: The 2004 public-health survey in Skåne is a cross-sectional study. A total of 27,757 persons aged 18-80 years answered a postal questionnaire, which represents 59% of the random sample. A logistic regression model was used to investigate the associations between gender and daily smoking according to age. The multivariate analysis was performed to investigate the importance of possible confounders (country of origin, education, snus use, alcohol consumption, leisure-time physical activity, and BMI) on the gender differences in daily smoking in different age groups. Results: 14.9% of the men and 18.1% of the women were daily smokers. Middle-aged respondents were daily smokers to a significantly higher extent than young and old respondents. The prevalence of daily smoking also varied according to other demographic, socioeconomic, health related behaviour, and BMI characteristics. The crude odds ratios of daily smoking were 1.79 (1.42-2.26) among women compared to men in the 18-24 years age group, and 0.95 (0.80-1.12) in the 65-80 years age group. These odds ratios changed to 2.00 (1.49-2.67) and 0.95 (0.76-1.18), respectively, when all confounders were included. CONCLUSIONS: For the first time in Sweden women have a higher prevalence of daily smoking than men. The odds ratios of daily smoking are highest among women compared to men in the youngest age group of 18-24 years and the odds ratios decrease with increasing age. The findings point to a serious public health problem. Strategic interventions targeting young women's tobacco smoking are needed.

    • This article presents a survey of techniques for ranking results in search engines, with emphasis on link-based ranking methods and the PageRank algorithm. The problem of selecting, in relation to a user search query, the most relevant documents from an unstructured source such as the WWW is discussed in detail. The need for extending classical information retrieval techniques such as boolean searching and vector space models with link-based ranking methods is demonstrated. The PageRank algorithm is introduced, and its numerical and spectral properties are discussed. The article concludes with an alternative means of computing PageRank, along with some example applications of this new method.

    • The aim of this paper is to introduce n-ary Hom-algebra structures generalizing the n-ary algebras of Lie type including n-ary Nambu algebras, n-ary Nambu-Lie algebras and n-ary Lie algebras, and n-ary algebras of associative type including n-ary totally associative and n-ary partially associative algebras. We provide examples of the new structures and present some properties and construction theorems. We describe the general method allowing one to obtain an n-ary Hom-algebra structure starting from an n-ary algebra and an n-ary algebra endomorphism. Several examples are derived using this process. Also we initiate investigation of classification problems for algebraic structures introduced in the article and describe all ternary three-dimensional Hom-Nambu-Lie structures with diagonal homomorphism.

    • For some forty years the eye-tracking technology has facilitated the study of eye movement patterns for sighted people during reading and other visual activities. Today – a newly developed automatic finger tracking system makes it possible to reconstruct blind people’s tactile reading in real time and to automatically analyze finger movements during Braille text reading and tactile picture recognition. In this case study, the very first automatic finger tracking system is presented together with results indicating how Braille readers can increase awareness of their own reading styles. This opens up for future Braille education to become more evidence-based and, at the same time, for a new research field: contrastive studies of language in its auditory, visual and tactile manifestations.

    • For dynamical systems defined by a covering map of a compact Hausdorff space and the corresponding transfer operator, the associated crossed product C *-algebras C(X)⋊ α,ℒℕintroduced by Exel and Vershik are considered. An important property for homeomorphism dynamical systems is topological freeness. It can be extended in a natural way to in general non-invertible dynamical systems generated by covering maps. In this article, it is shown that the following four properties are equivalent: the dynamical system generated by a covering map is topologically free; the canonical embedding of C(X) into C(X)⋊ α,ℒℕis a maximal abelian C *-subalgebra of C(X)⋊ α,ℒN; any nontrivial two sided ideal of C(X)⋊ α,ℒℕhas non-zero intersection with the embedded copy of C(X); a certain natural representation of C(X)⋊ α,ℒℕis faithful. This result is a generalization to non-invertible dynamics of the corresponding results for crossed product C *-algebras of homeomorphism dynamical systems.

    • In this paper we extend the eliminant construction of Burchnall and Chaundy for commuting differential operators in the Heisenberg algebra to the q-deformed Heisenberg algebra and show that it again provides annihilating curves for commuting elements, provided q satisfies a natural condition. As a side result we obtain estimates on the dimensions of the eigenspaces of elements of this algebra in its faithful module of Laurent series.

    • A cultivation strategy combining the advantages of temperature-limited fed-batch and probing feeding control is presented. The technique was evaluated in fed-batch cultivations with E. coli BL21(DE3) producing xylanase in a 3 liter bioreactor. A 20% increase in cell mass was achieved and the usual decrease in specific enzyme activity normally observed during the late production phase was diminished with the new technique. The method was further tested by growing E. coli W3110 in a larger bioreactor (50 l). It is a suitable cultivation technique when the O2 transfer capacity of the reactor is reached and it is desired to continue to produce the recombinant protein.
