  • Carbon dioxide reduction mechanism on Ru-based electrocatalysts [Ru(bpy)(2)(CO)(2)](2+) : insights from first-principles theory

    • Solar fuel production through the so-called artificial photosynthesis has attracted a great deal of attention to the development of a new world energy matrix that is renewable and environmentally friendly. This process is characterized by light absorption with enough photon energy to generate conduction electrons, which drive the carbon dioxide reduction to produce organic fuels. It is also common to couple Ru-complex electrocatalysts to form a more efficient and selective hybrid system for this application. In this work, we have undertaken a thorough investigation of the redox reaction mechanism of Ru-based electrocatalysts by means of density functional theory (DFT) methods under the experimental conditions that have been previously reported. More specifically, we have studied the electrochemistry and catalytic activity of the [Ru(bpy)(2)(CO)(2)](2+) coordination complex. Our theoretical assessment supports the following catalytic cycle: (i) [Ru(bpy)(2)(CO)(2)](2+) is transformed into [Ru(bpy)(2)(CO)](0) upon two-electron reduction and CO release; (ii) [Ru(bpy)(2)(CO)](0) is protonated to form the [Ru(bpy)(2)(CO)H](+) hydride complex; (iii) CO2 is activated by the hydride complex through an electrophilic addition to form the [Ru(bpy)(2)(CO)(OCHO)](+) intermediate; (iv) the resulting formic acid ligand is released in solution; and, finally, (v) the CO ligand is reattached to the complex to recover the initial [Ru(bpy)(2)(CO)(2)](2+) catalyst.

  • Cloning and Functional Studies of a Splice Variant of CYP26B1 Expressed in Vascular Cells

    • Background: All-trans retinoic acid (atRA) plays an essential role in the regulation of gene expression, cell growth and differentiation and is also important for normal cardiovascular development but may in turn be involved in cardiovascular diseases, i.e. atherosclerosis and restenosis. The cellular atRA levels are under strict control involving several cytochromes P450 isoforms (CYPs). CYP26 may be the most important regulator of atRA catabolism in vascular cells. The present study describes the molecular cloning, characterization and function of atRA-induced expression of a spliced variant of the CYP26B1 gene. Methodology/Principal Findings: The coding region of the spliced CYP26B1 lacking exon 2 was amplified from cDNA synthesized from atRA-treated human aortic smooth muscle cells and sequenced. Both the spliced variant and full length CYP26B1 was found to be expressed in cultured human endothelial and smooth muscle cells, and in normal and atherosclerotic vessel. atRA induced both variants of CYP26B1 in cultured vascular cells. Furthermore, the levels of spliced mRNA transcript were 4.5 times higher in the atherosclerotic lesion compared to normal arteries and the expression in the lesions was increased 20-fold upon atRA treatment. The spliced CYP26B1 still has the capability to degrade atRA, but at an initial rate one-third that of the corresponding full length enzyme. Transfection of COS-1 and THP-1 cells with the CYP26B1 spliced variant indicated either an increase or a decrease in the catabolism of atRA, probably depending on the expression of other atRA catabolizing enzymes in the cells. Conclusions/Significance: Vascular cells express the spliced variant of CYP26B1 lacking exon 2 and it is also increased in atherosclerotic lesions. The spliced variant displays a slower and reduced degradation of atRA as compared to the full-length enzyme. Further studies are needed, however, to clarify the substrate specificity and role of the CYP26B1 splice variant in health and disease.

    • The global loss of biodiversity continues at an alarming rate. Genomic approaches have been suggested as a promising tool for conservation practice as scaling up to genome-wide data can improve traditional conservation genetic inferences and provide qualitatively novel insights. However, the generation of genomic data and subsequent analyses and interpretations remain challenging and largely confined to academic research in ecology and evolution. This generates a gap between basic research and applicable solutions for conservation managers faced with multifaceted problems. Before the real-world conservation potential of genomic research can be realized, we suggest that current infrastructures need to be modified, methods must mature, analytical pipelines need to be developed, and successful case studies must be disseminated to practitioners.

  • Conspecific boldness and predator species determine predation-risk consequences of prey personality

    • Abstract: Individual variation in the behavior of prey can influence predation risk in complex ways. We ran individual roach (Rutilus rutilus), a common freshwater fish, through a standard refuge emergence protocol to characterize their boldness, a key animal personality trait. We then paired a bold and a shy roach and exposed the pair to one of two predator species that have contrasting hunting modes to ascertain how personality traits shaped their survival during predator encounters. When a paired bold and shy prey fish interacted with a perch predator (active foraging mode), bold and shy prey were consumed in almost equal numbers. However, pike predators (ambush foraging mode) selectively consumed more shy prey, and prey body size and boldness score both contributed significantly to which prey fish was eaten. Our findings support the idea that multiple predators with different foraging modes, and hence differential selection on prey personality, could contribute to maintaining variation in personality in prey populations. Furthermore, for social species, including shoaling fish, the ultimate consequences of an individual’s personality may depend upon the personality of its nearby conspecifics. Significance statement: Animals of the same species often look similar, but individuals show differences in their behavior that can have important consequences, for instance when these individuals interact with predators. The common roach is a freshwater fish that shows inter-individual variation in its propensity to take risks, a key personality trait often termed boldness. Variation in boldness may affect the outcome when roach interact with predators, i.e., if they get eaten or survive. However, we found the impact of roachs’ personality type depends on what species of predatory fish they face. When we put a shy and a bold roach together with predatory perch, the roachs’ personality did not significantly affect which individual was eaten. But when the predator was a pike, the predators selectively ate more shy roach, and the likelihood an individual would be eaten depended on their body size.

  • Map-based prediction of organic carbon in headwater streams improved by downstream observations from the river outlet

    • In spite of the great abundance and ecological importance of headwater streams, managers are usually limited by a lack of information about water chemistry in these headwaters. In this study we test whether river outlet chemistry can be used as an additional source of information to improve the prediction of the chemistry of upstream headwaters (size < 2 km(2)), relative to models based on map information alone. We use the concentration of total organic carbon (TOC), an important stream ecosystem parameter, as the target for our study. Between 2000 and 2008, we carried out 17 synoptic surveys in 9 mesoscale catchments (size 32-235 km(2)). Over 900 water samples were collected in total, primarily from headwater streams but also including each catchment's river outlet during every survey. First we used partial least square regression (PLS) to model the distribution (median, interquartile range (IQR)) of headwater stream TOC for a given catchment, based on a large number of candidate variables including sub-catchment characteristics from GIS, and measured river chemistry at the catchment outlet. The best candidate variables from the PLS models were then used in hierarchical linear mixed models (MM) to model TOC in individual headwater streams. Three predictor variables were consistently selected for the MM calibration sets: (1) proportion of forested wetlands in the sub-catchment (positively correlated with headwater stream TOC), (2) proportion of lake surface cover in the sub-catchment (negatively correlated with headwater stream TOC), and (3) river outlet TOC (positively correlated with headwater stream TOC). Including river outlet TOC improved predictions, with 5-15% lower prediction errors than when using map information alone. Thus, data on water chemistry measured at river outlets offer information which can complement GIS-based modelling of headwater stream chemistry.

  • Consequences of mixing assumptions for time-variable travel time distributions

    • The current generation of catchment travel time distribution (TTD) research, integrating nearly three decades of work since publication of Water's Journey from Rain to Stream, seeks to represent the full distribution in catchment travel times and its temporal variability. Here, we compare conceptualizations of increasing complexity with regards to mixing of water storages and evaluate how these assumptions influence time-variable TTD estimates for two catchments with contrasting climates: the Gardsjon catchment in Sweden and the Marshall Gulch catchment in Arizona, USA. Our results highlight that, as long as catchment TTDs cannot be measured directly but need to be inferred from input-output signals of catchments, the inferred catchment TTDs depend strongly on the underlying assumptions of mixing within a catchment. Furthermore, we found that the conceptualization of the evapotranspiration flux strongly influences the inferred travel times of stream discharge. For the wet and forested Gardsjon catchment in Sweden, we inferred that evapotranspiration most likely resembles a completely mixed sample of the water stored in the catchment; however, for the drier Marshall Gulch catchment in Arizona, evapotranspiration predominantly contained the younger water stored in the catchment. For the Marshall Gulch catchment, this higher probability for young water in evapotranspiration resulted in older water in the stream compared to travel times inferred with assumptions of complete mixing. New observations that focus on the TTD of the evapotranspiration flux and the actual travel time of water through a catchment are necessary to improve identification of mixing and consequently travel times of stream water.   • From cohorts to molecules: Adverse impacts of endocrine disrupting mixtures

    • Convergent evidence associates exposure to endocrine disrupting chemicals (EDCs) with major human diseases, even at regulation-compliant concentrations. This might be because humans are exposed to EDC mixtures, whereas chemical regulation is based on a risk assessment of individual compounds. Here, we developed a mixture-centered risk assessment strategy that integrates epidemiological and experimental evidence. We identified that exposure to an EDC mixture in early pregnancy is associated with language delay in offspring. At human-relevant concentrations, this mixture disrupted hormone-regulated and disease-relevant regulatory networks in human brain organoids and in the model organisms Xenopus leavis and Danio rerio, as well as behavioral responses. Reinterrogating epidemiological data, we found that up to 54% of the children had prenatal exposures above experimentally derived levels of concern, reaching, for the upper decile compared with the lowest decile of exposure, a 3.3 times higher risk of language delay. © 2022 American Association for the Advancement of Science.     • The reactions of coronene dehydrogenation and fusion upon heat treatment in the temperature range of 500-700 degrees C were studied using XRD, TEM, Raman, IR, and NEXAFS spectroscopy. The formation of a coronene dimer (dicoronylene) was observed at temperatures 530-550 degrees C; dicoronylene can easily be separated using sublimation with a temperature gradient. An insoluble and not sublimable black precipitate was found to form at higher temperatures. Analysis of the data shows that dimerization of coronene is followed at 550-600 degrees C by oligomerization into larger molecules. Above 600 degrees C amorphization of the material and formation of graphitic nanoparticles was observed. Coronene fusion by annealing is proposed as a road to synthesis of larger polycyclic aromatic hydrocarbons and nanographenes.

    • This joint experimental-theoretical spectroscopy study of the fullerene derivative PC60BM ([6,6]-phenyl-C60-butyric acid methyl ester) aims to improve the understanding of the effect of photooxidation on its electronic structure. We have studied spin-coated thin films of PC60BM by X-ray Photoelectron Spectroscopy (XPS), Near-edge X-ray Absorption Fine Structure (NEXAFS) spectroscopy, and Fourier Transform Infrared Spectroscopy (FTIR), before and after intentional exposure to simulated sunlight in air for different lengths of time. The π* resonance in the C1s NEXAFS spectrum was found to be a very sensitive probe for the early changes to the fullerene cage, while FTIR spectra, in combination with O1s NEXAFS spectra, enabled the identification of the oxidation products. The changes observed in the spectra obtained by these complementary methods were compared with the corresponding Density Functional Theory (DFT) calculated single-molecule spectra of a large set of in silico generated oxidation products of PC60BM where oxygen atoms were attached to the C60 cage. This comparison confirms that photooxidation of PC60BM disrupts the conjugation of the fullerene cage by a transition from sp2 to sp3-hybridized carbon and causes the formation of several oxidation products, earlier proposed for C60. The agreement between experimental and calculated IR spectra suggests moreover the presence of dicarbonyl and anhydride structures on the fullerene cage, in combination with cage opening at the adsorption site. By including PC60BM with physisorbed O2 molecules on the cage in our theoretical description in order to model oxygen diffused through the film, the experimental O1s XPS and O1s NEXAFS spectra could be reproduced.

    • Hydrated electrons are important in radiation chemistry and charge transfer reactions, with applications that include chemical damage of DNA, catalysis, and signaling. Conventionally, hydrated electrons are produced by pulsed radiolysis, sonolysis, two-ultraviolet-photon laser excitation of liquid water, or photodetachment of suitable electron donors. Here we report a method for the generation of hydrated electrons via single-visible-photon excitation of localized surface plasmon resonances (LSPRs) of supported sub-3 nm copper nanoparticles in contact with water. Only excitations at the LSPR maximum resulted in the formation of hydrated electrons, suggesting that plasmon excitation plays a crucial role in promoting electron transfer from the nanoparticle into the solution. The reactivity of the hydrated electrons was confirmed via proton reduction and concomitant H-2 evolution in the presence of a Ru/TiO2 catalyst.
