    • Proteins are key components in a multitude of biological processes, of which the functions carried out by transmembrane (membrane-spanning) proteins are especially demanding for investigations. This is because this class of protein needs to be incorporated into a lipid bilayer representing its native environment, and in addition, many experimental conditions also require a solid support for stabilization and analytical purposes. The solid support substrate may, however, limit the protein functionality due to protein material interactions and a lack of physical space. We have in this work tailored the pore size and pore ordering of a mesoporous silica thin film to match the native cell-membrane arrangement of the transmembrane protein human aquaporin 4 (hAQP4). Using neutron reflectivity (NR), we provide evidence of how substrate pores host the bulky water-soluble domain of hAQP4, which is shown to extend 7.2 nm into the pores of the substrate. Complementary surface analytical tools, including quartz crystal microbalance with dissipation monitoring (QCM-D) and fluorescence microscopy, revealed successful protein-containing supported lipid bilayer (pSLB) formation on mesoporous silica substrates, whereas pSLB formation was hampered on nonporous silica. Additionally, electron microscopy (TEM and SEM), light scattering (DLS and stopped-flow), and small-angle X-ray scattering (SAXS) were employed to provide a comprehensive characterization of this novel hybrid organic-inorganic interface, the tailoring of which is likely to be generally applicable to improve the function and stability of a broad range of membrane proteins containing water-soluble domains.

  • Human Saliva Forms a Complex Film Structure on Alumina Surfaces

    • Films formed from saliva on surfaces are important for the maintenance of oral health and integrity by protection against chemical and/or biological agents. The aim of the present study was to investigate adsorbed amounts, thickness, and structure of films formed from human whole saliva on alumina surfaces by means of in situ ellipsometry, neutron reflectivity, and atomic force microscopy. Alumina (Al2O3, synthetic sapphire) is a relevant and interesting substrate for saliva adsorption studies as it has an isoelectric point close to that of tooth enamel. The results showed that saliva adsorbs rapidly on alumina. The film could be modeled in two layers:  an inner and dense thin region that forms a uniform layer and an outer, more diffuse and thicker region that protrudes toward the bulk of the solution. The film morphology described a uniformly covering dense layer and a second outer layer containing polydisperse adsorbed macromolecules or aggregates.

    • Socio-hydrology has expanded and been effective in exposing the hydrological community to ideas and approaches from other scientific disciplines, and social sciences in particular. Yet it still has much to explore regarding how to capture human agency and how to combine different methods and disciplinary views from both the hydrological and the social sciences to develop knowledge. A useful starting ground is noting that the complexity of human–water relations is due to interactions not only across spatial and temporal scales but also across different organizational levels of social systems. This calls for consideration of another analytical scale, the human organizational scale, and interdisciplinarity in study methods. Based on the papers published in this journal’s Special Issue Advancing Socio-hydrology over 2019–2022, this paper illuminates how the understanding of coupled human–water systems can be strengthened by capturing the multi-level nature of human decision making and by applying an interdisciplinary multi-method approach.

    • Hfszeeman95 is an updated and extended Fortran 95 version of the Hfszeeman program (Andersson and Jönsson, 2008). Given relativistic atomic state functions generated by the Grasp2018 package (Fischer et al., 2019), Hfszeeman95 together with the accompanying Matlab/GNU Octave program Mithit allows for: (1) the computation and plotting of Zeeman energy splittings of magnetic fine- and hyperfine structure substates as functions of the strength of an external magnetic field, (2) the computation of transition rates between different magnetic fine- and hyperfine structure substates in the presence of an external magnetic field and rates of hyperfine-induced transitions in the field free limit, (3) the synthesization of spectral profiles for transitions obtained from (2). With the new features, Hfszeeman95 and the accompanying Matlab/GNU Octave program Mithit are useful for the analysis of observational spectra and to resolve the complex features due to the splitting of the fine and hyperfine levels.

    • Dental implant abutments that emerge through the mucosa are rapidly covered with a salivary protein pellicle to which bacteria bind, initiating biofilm formation. In this study, adherence of early colonizing streptococci, Streptococcus gordonii, Streptococcus oralis, Streptococcus mitis and Streptococcus sanguinis to two saliva-coated anodically oxidized surfaces was compared with that on commercially pure titanium (CpTi). Near edge X-ray absorption (NEXAFS) showed crystalline anatase was more pronounced on the anodically oxidized surfaces than on the CpTi. As revealed by fluorescence microscopy, a four-species mixture, as well as individual bacterial species, exhibited lower adherence after 2 h to the saliva-coated, anatase-rich surfaces than to CpTi. Since wettability did not differ between the saliva-coated surfaces, differences in the concentration and/or configuration of salivary proteins on the anatase-rich surfaces may explain the reduced bacterial binding effect. Anatase-rich surfaces could thus contribute to reduced overall biofilm formation on dental implant abutments through diminished adherence of early colonizers.

    • Arrays of nanoparticle-supported lipid bilayers (nanoSLB) are lipid-coated nanopatterned interfaces that provide a platform to study curved model biological membranes using surface-sensitive techniques. We combined scattering techniques with direct imaging, to gain access to sub-nanometer scale structural information on stable nanoparticle monolayers assembled on silicon crystals in a noncovalent manner using a Langmuir-Schaefer deposition. The structure of supported lipid bilayers formed on the nanoparticle arrays via vesicle fusion was investigated using a combination of grazing incidence X-ray and neutron scattering techniques complemented by fluorescence microscopy imaging. Ordered nanoparticle assemblies were shown to be suitable and stable substrates for the formation of curved and fluid lipid bilayers that retained lateral mobility, as shown by fluorescence recovery after photobleaching and quartz crystal microbalance measurements. Neutron reflectometry revealed the formation of high-coverage lipid bilayers around the spherical particles together with a flat lipid bilayer on the substrate below the nanoparticles. The presence of coexisting flat and curved supported lipid bilayers on the same substrate, combined with the sub-nanometer accuracy and isotopic sensitivity of grazing incidence neutron scattering, provides a promising novel approach to investigate curvature-dependent membrane phenomena on supported lipid bilayers.

    • In situ structural and chemical state characterization of Rh/CeO2 and Ni/CeO2 catalysts during atmospheric pressure CO2 methanation has been performed by a combined array of time-resolved analytical techniques including ambient-pressure X-ray photoelectron spectroscopy, high-energy X-ray diffraction and diffuse reflectance infrared Fourier transform spectroscopy. The ceria phase is partially reduced during the CO2 methanation and in particular Ce3+ species seem to facilitate activation of CO2 molecules. The activated CO2 molecules then react with atomic hydrogen provided from H-2 dissociation on Rh and Ni sites to form formate species. For the most active catalyst (Rh/CeO2), transmission electron microscopy measurements show that the Rh nanoparticles are small (average 4 nm, but with a long tail towards smaller particles) due to a strong interaction between Rh particles and the ceria phase. In contrast, larger nanoparticles were observed for the Ni/CeO2 catalyst (average 6 nm, with no crystallites below 5 nm found), suggesting a weaker interaction with the ceria phase. The higher selectivity towards methane of Rh/CeO2 is proposed to be due to the stronger metal-support interaction.

    • In this paper, we examine decay and fragmentation of core-excited and core-ionized water molecules combining quantum chemical calculations and electron-energy-resolved electron–ion coincidence spectroscopy. The experimental technique allows us to connect electronic decay from core-excited states, electronic transitions between ionic states, and dissociation of the molecular ion. To this end, we calculate the minimum energy dissociation path of the core-excited molecule and the potential energy surfaces of the molecular ion. Our measurements highlight the role of ultra-fast nuclear motion in the 1a1-14a1 core-excited molecule in the production of fragment ions. OH+ fragments dominate for spectator Auger decay. Complete atomization after sequential fragmentation is also evident through detection of slow H+ fragments. Additional measurements of the non-resonant Auger decay of the core-ionized molecule (1a1-1) to the lower-energy dication states show that the formation of the OH+ + H+ ion pair dominates, whereas sequential fragmentation OH+ + H+ → O + H+ + H+ is observed for transitions to higher dication states, supporting previous theoretical investigations.

    • Remote health monitoring is a trend for better health management which necessitates the need for secure monitoring and privacy-preservation of patient data. Moreover, accurate and continuous monitoring of personal health status may require expert validation in an active learning strategy. As a result, this paper proposes a Federated Interactive Learning IoT-based Health Monitoring Platform (FIL-IoT-HMP) which incorporates multi-expert feedback as ‘Human-in-the-loop’ in an active learning strategy in order to improve the clients’ Machine Learning (ML) models. The authors have proposed an architecture and conducted an experiment as a proof of concept. Federated learning approach has been preferred in this context given that it strengthens privacy by allowing the global model to be trained while sensitive data is retained at the local edge nodes. Also, each model’s accuracy is improved while privacy and security of data has been upheld.

    • An overview of recent experimental results of studies of negative atomic and molecular ions in the Double ElectroStatic Ion-Ring ExpEriment, DESIREE is given. Metastable level lifetimes in atomic negative ions have been measured by time-dependent laser photodetachment. Rotational relaxation of diatomic anions is studied by near-threshold photodetachment. Spontaneous decays of small metal cluster anions by electron emission and fragmentation is studied with decay-channel specificity. Finally, mutual neutralisation of pairs of negative and positive ions has been investigated with initial and final state selectivity.
