  • Long‐term changes in trophic ecology of blue mussels in a rapidly changing ecosystem

    • Ocean climate change strongly affects organisms and ecosystems, and the causes, consequences, and underlying mechanisms need to be documented. In the Baltic Sea, a marginal sea under severe eutrophication stress, a longer productive season, and changes in the phytoplankton community over the last few decades have likely impacted diet and condition of keystone species, from individual to population level. This study uses stable isotopes (δ13C, δ15N, and derived isotope niche metrics) to trace energy and nutrient flows in archived samples of blue mussel (Mytilus edulis trossulus) spanning 24 yr (1993–2016). We test if long-term changes in isotope and elemental composition in mussels, as well as population abundance and biomass, can be explained by changes in abiotic and biotic variables, using partial least square regressions and structural equation modeling. We found decreasing trends in δ13C and δ15N as well as in mean size and total biomass of mussels, but no unidirectional changes in their stoichiometry or condition index. Changes in isotope composition were best explained by nitrogen-fixing cyanobacteria, by increased terrestrial organic carbon from land runoff (reflecting precipitation) and by decreases in dissolved inorganic nitrogen (indicative of successful eutrophication mitigation) and in biomass of a mixotrophic ciliate species. The trophic niche (assessed from isotope niche) was included as the best predictor for both mussel body condition and the observed decline in their total biomass. This study reveals that altered trophic relationships from climate-induced changes in the productivity base may strongly impact keystone species, with potential knock-on effects on ecosystem functions.

  • Power mapping and noise reduction for financial correlations

    • The spectral properties of financial correlation matrices can show features known from completely random matrices. A major reason is noise originating from the finite lengths of the financial time series used to compute the correlation matrix elements. In recent years, various methods have been proposed to reduce this noise, i.e. to clean the correlation matrices. This is of direct practical relevance for risk management in portfolio optimization. In this contribution, we discuss in detail the power mapping, a new shrinkage method. We show that the relevant parameter is, to a certain extent, self-determined. Due to the "chirality" and the normalization of the correlation matrix, the optimal shrinkage parameter is fixed. We apply the power mapping and the well-known filtering method, to market data and compare them by optimizing stock portfolios. We address the rle of constraints by excluding short selling in the optimization.

  • Speaking of Geometry : A study of geometry textbooks and literature on geometry instruction for elementary and lower secondary levels in Sweden, 1905-1962, with a special focus on professional debates

    • This dissertation deals with geometry instruction in Sweden in the period 1905-1962. The purpose is to investigate textbooks and other literature used by teachers in elementary schools (ES) and lower secondary schools (LSS) – Folkskolan and Realskolan – connection to geometry instruction. Special attention is given to debates about why a course should be taught and how the content should be communicated.In the period 1905-1962, the Swedish school system changed greatly. Moreover, in this period mathematics instruction was reformed in several countries and geometry was a major issue; especially, classical geometry based on the axiomatic method. However, we do not really know how mathematics instruction changed in Sweden. Moreover, in the very few works where the history of mathematics instruction in Sweden is mentioned, the time before 1950 is often described in terms of “traditional”, “static” and “isolation”.In this dissertation, I show that geometry instruction in Sweden did change in the period 1905-1962: geometry instruction in LSS was debated; the axiomatic method and spatial intuition were major issues. Textbooks for LSS not following Euclid were produced also, but the axiomatic method was kept. By 1930, these alternative textbooks were the most popular.Also the textbooks in ES changed. In the debate about geometry instruction in ES, visualizability was a central concept.Nonetheless, some features did not change. Throughout the period, the rationale for keeping axiomatic geometry in LSS was to provide training in reasoning. An important aspect of the debate on geometry instruction in LSS is that the axiomatic method was the dominating issue; other issues, e.g. heuristics, were not discussed. I argue that a discussion on heuristics would have been relevant considering the final exams in the LSS; in order to succeed, it was more important to be a skilled problem solver than a master of proof.

    • Organohalogens, a class of molecules that contain at least one halogen atom bonded to carbon, are abundant on the Earth where they are mainly produced through industrial and biological processes(1). Consequently, they have been proposed as biomarkers in the search for life on exoplanets(2). Simple halogen hydrides have been detected in interstellar sources and in comets, but the presence and possible incorporation of more complex halogen-containing molecules such as organohalogens into planet-forming regions is uncertain(3,4). Here we report the interstellar detection of two isotopologues of the organohalogen CH3Cl and put some constraints on CH3F in the gas surrounding the low-mass protostar IRAS 16293-2422, using the Atacama Large Millimeter/submillimeter Array (ALMA). We also find CH3Cl in the coma of comet 67P/Churyumov-Gerasimenko (67P/C-G) by using the Rosetta Orbiter Spectrometer for Ion and Neutral Analysis (ROSINA) instrument. The detections reveal an efficient pre-planetary formation pathway of organohalogens. Cometary impacts may deliver these species to young planets and should thus be included as a potential abiotical production source when interpreting future organohalogen detections in atmospheres of rocky planets.

    • Objectives: Genetic variations in TNFAIP3 (A20) de-ubiquitinase (DUB) domain increase the risk of systemic lupus erythematosus (SLE) and rheumatoid arthritis. A20 is a negative regulator of NF-κB but the role of its DUB domain and related genetic variants remain unclear. We aimed to study the functional effects of A20 DUB-domain alterations in immune cells and understand its link to SLE pathogenesis. Methods: CRISPR/Cas9 was used to generate human U937 monocytes with A20 DUB-inactivating C103A knock-in (KI) mutation. Whole genome RNA-sequencing was used to identify differentially expressed genes between WT and C103A KI cells. Functional studies were performed in A20 C103A U937 cells and in immune cells from A20 C103A mice and genotyped healthy individuals with A20 DUB polymorphism rs2230926. Neutrophil extracellular trap (NET) formation was addressed ex vivo in neutrophils from A20 C103A mice and SLE-patients with rs2230926. Results: Genetic disruption of A20 DUB domain in human and murine myeloid cells did not give rise to enhanced NF-κB signalling. Instead, cells with C103A mutation or rs2230926 polymorphism presented an upregulated expression of PADI4, an enzyme regulating protein citrullination and NET formation, two key mechanisms in autoimmune pathology. A20 C103A cells exhibited enhanced protein citrullination and extracellular trap formation, which could be suppressed by selective PAD4 inhibition. Moreover, SLE-patients with rs2230926 showed increased NETs and increased frequency of autoantibodies to citrullinated epitopes. Conclusions: We propose that genetic alterations disrupting the A20 DUB domain mediate increased susceptibility to SLE through the upregulation of PADI4 with resultant protein citrullination and extracellular trap formation.

    • Three efficient routes to 3-azido-3-deoxy-beta-d-galactopyranosides were developed relying on a double inversion protocol at C3. Two of the routes were demonstrated to work with both O- and S-glycosides. In all three routes, the 2-O-acetyl-3-azido-4,6-O-benzylidene-3-deoxy-beta-d-galactopyranosides were obtained by an azide inversion of the key intermediates 2-O-acetyl-4,6-O-benzylidene-3-O-trifluoromethanesulfonyl-beta-d-gulopyranosides. The intermediate gulopyranosides were in turn obtained from 2-O-acetyl-4,6-O-benzylidene-3-O-trifluoromethanesulfonyl-beta-d-galactopyranosides, installed in one pot from the 4,6-O-benzylidene-beta-d-galactopyranosides, by inversion with nitrite or acetate. For O-glycosides, the gulopyranoside configuration could alternatively be obtained from the 4,6-O-benzylidene-beta-d-galactopyranoside by elimination to give the 2,3-dianhydro derivative followed by a highly stereoselective cis-dihydroxylation.
