    • The vertical dynamic interaction between vehicle and railway track is simulated in the time domain using an extended state space vector approach. The track model includes a transition zone between slab track on a bridge and ballasted track on an embankment. By considering a multi-objective optimisation problem, solved using a genetic algorithm, selected vehicle and track responses are simultaneously minimised by optimising the distributions of rail pad stiffness and sleeper spacing in the transition zone. It is shown that the magnitudes of the maximum dynamic loads in the optimised transition zone can be reduced to be similar as the magnitudes far away from the transition zone.

  • Projected sensitivity to sub-GeV dark matter of next-generation semiconductor detectors

    • We compute the projected sensitivity to dark matter (DM) particles in the sub-GeV mass range of future direct detection experiments using germanium and silicon semiconductor targets. We perform this calculation within the dark photon model for DM-electron interactions using the likelihood ratio as a test statistic, Monte Carlo simulations, and background models that we extract from recent experimental data. We present our results in terms of DM-electron scattering cross section values required to reject the background only hypothesis in favour of the background plus DM signal hypothesis with a statistical significance, Z, corresponding to 3 or 5 standard deviations. We also test the stability of our conclusions under changes in the astrophysical parameters governing the local space and velocity distribution of DM in the Milky Way. In the best-case scenario, when a high-voltage germanium detector with an exposure of 50 kg-year and a CCD silicon detector with an exposure of 1 kg-year and a dark current rate of 1×10-7 counts/pixel/day have simultaneously reported a DM signal, we find that the smallest cross section value compatible with Z=3 (Z=5) is about 4×10-42 cm2 (6×10-42 cm2) for contact interactions, and 2.5×10-41 cm2 (4×10-41 cm2) for long-range interactions. Our sensitivity study extends and refine previous works in terms of background models, statistical methods, and treatment of the underlying astrophysical uncertainties.

    • It is generally believed that estrogens exert their bone sparing effects directly on the cells within the bone compartment. The aim of the present study was to investigate if central mechanisms might be involved in the bone sparing effect of estrogens. The dose-response of central (i.c.v) 17beta-estradiol (E2) administration was compared with that of peripheral (s.c.) administration in ovariectomized (ovx) mice. The dose-response curves for central and peripheral E2 administration did not differ for any of the studied estrogen-responsive tissues, indicating that these effects were mainly peripheral. In addition, ovx mice were treated with E2 and/or the peripheral estrogen receptor antagonist ICI 182,780. ICI 182,780 attenuated most of the estrogenic response regarding uterus weight, retroperitoneal fat weight, cortical BMC and trabecular bone mineral content (P<0.05). These findings support the notion that the primary target tissue that mediates the effect of E2 on bone is peripheral and not central.

    • To investigate the role of the central neuromedin U (NMU) signaling system in body weight and energy balance regulation, we examined the effects of long-term intracerebroventricular (icv) infusion of NMU in C57Bl/6 mice and in mice lacking the gene encoding NMU receptor 2. In diet-induced obese male and female C57BL/6 mice, icv infusion of NMU (8 microg x day(-1) x mouse(-1)) for 7 days decreased body weight and total energy intake compared with vehicle treatment. However, these parameters were unaffected by NMU treatment in lean male and female C57BL/6 mice fed a standard diet. In addition, female (but not male) NMUR2-null mice had increased body weight and body fat mass when fed a high-fat diet but lacked a clear body weight phenotype when fed a standard diet compared with wild-type littermates. Furthermore, female (but not male) NMUR2-null mice fed a high-fat diet were protected from central NMU-induced body weight loss compared with littermate wild-type mice. Thus, we provide the first evidence that long-term central NMU treatment reduces body weight, food intake, and adiposity and that central NMUR2 signaling is required for these effects in female but not male mice.

    • To study the role of the growth hormone receptor (GHR) in the development of cardiovascular structure and function, female GHR gene-disrupted or knockout (KO) and wild-type (WT) mice at age 18 wk were used. GHR KO mice had lower plasma renin levels (12 ± 2 vs. 20 ± 4 mGU/ml, P < 0.05) and increased aortic endothelial NO synthase (eNOS) expression (146%, P < 0.05) accompanied by a 25% reduction in systolic blood pressure (BP, 110 ± 4 vs. 147 ± 3 mmHg, P < 0.001) compared with WT mice. Aldosterone levels were unchanged, whereas the plasma potassium concentration was elevated by 14% ( P < 0.05) in GHR KO. Relative left ventricular weight was 14% lower in GHR KO mice ( P < 0.05), and cardiac dimensions as analyzed by echocardiography were similarly reduced. Myograph studies revealed a reduced maximum contractile response in the aorta to norepinephrine (NE) and K+ ( P < 0.05), and aorta media thickness was decreased in GHR KO ( P < 0.05). However, contractile force was normal in mesenteric arteries, whereas sensitivity to NE was increased ( P < 0.05). Maximal acetylcholine-mediated dilatation was similar in WT and GHR KO mice, whereas the aorta of GHR KO mice showed an increased sensitivity to acetylcholine ( P < 0.05). In conclusion, loss of GHR leads to low BP and decreased levels of renin in plasma as well as increase in aortic eNOS expression. Furthermore, GHR deficiency causes functional and morphological changes in both heart and vasculature that are beyond the observed alterations in body size. These data suggest an important role for an intact GH/IGF-I axis in the maintenance of a normal cardiovascular system.

    • Understanding the microstructural behaviour of materials during thermomechanical processing is a vital step towards optimizing the mechanical properties. One important aspect during forming processes, such as forging, is dynamic recrystallization (DRX), which sets the starting microstructure for the subsequent manufacturing steps. Here we investigated the DRX behaviour of Ni-base superalloy Haynes 282 during hot compression with a strain rate of 0.05 s−1 at 1080 °C, with care taken to minimize the effects of meta-dynamic recrystallization (mDRX) and adiabatic heating. Small DRX grains could be observed already at ε = 0.1, i.e. before the peak strain εp = 0.15. The DRX process accelerated significantly above ε = 0.2, and the material was fully recrystallized at ε = 1.5. Up to ε = 0.8 DRX occurred through continuous nucleation of new grains, whereas above ε = 0.8 the number density of DRX grains decreased and the increase in recrystallized fraction was due to growth of existing grains. Contrary to common assumptions of DRX nuclei being essentially dislocation free, many of the DRX grains contained pronounced dislocation substructures, even at small strains where they are not expected to have undergone deformation.

    • Forging on an industrial scale often involves slow, size-limited cooling rates or high temperature hold times between, or after, deformation. This enables the dynamic recrystallization (DRX) initiated during forging to further progress under static conditions, a phenomenon called meta-dynamic recrystallization (mDRX). As mDRX will influence the final grain size, and thus properties, it is critical to understand and control it during processing. Here, we study the mDRX evolution in Ni-based superalloy Haynes 282 during post-deformation hold times of up to 120 s at 1080 °C after partial DRX. We find that mDRX is the dominating mechanisms responsible for the microstructure evolution the hold time. The very rapid mDRX kinetics in the initial stages suggest that quench delays (the time between the end of the deformation and the onset of the quenching intended to arrest the microstructure evolution) must be kept well below 1 s in order to allow reliable conclusions to be drawn from post-deformation microstructure investigations. A larger prior strain (larger DRX fraction) leads to faster mDRX kinetics and a larger final grain size. Larger strains leads to earlier impingement of the growing grains, which, in combination with smaller remaining deformed regions into which the grains can grow, limits the maximum size of the mDRX grains. We also note a close correlation between static recovery and stress relaxation during the hold time, whereas no such correlation between mDRX and stress relaxation can be observed.

    • In alloys where carbides are the main grain boundary phase, the role of carbides during hot working is not known. Here, we address the effect of grain boundary carbides on the dynamic recrystallization during hot compression of Ni-base superalloy Haynes 282. When excluding variations from experimental factors neither stress evolution nor final microstructure indicated that carbides exerted a significant influence on the dynamic recrystallization. The carbide solvus temperature is not a critical limit during thermomechanical processes.
