    • A rapid and easy-to-use test kit, EPO WGA MAIIA, which can be used for distinguishing various endogenous human erythropoietins (hEPO) and several recombinant hEPOs and EPO analogues, has been evaluated. The test is based on chromatographic separation of the glycosylated isoforms of EPO using wheat germ agglutinin (WGA), and a sensitive immunoassay utilizing anti-EPO carbon black nanostrings and image scanning for quantification. All the reactions take place along the porous layer of a lateral flow micro-column containing WGA and anti-EPO zones. The presence of molecules resembling hEPO, like Mircera, was detected by the aberrant affinity interaction with the antibody zone on the strip. It was possible to distinguish nine recombinant hEPO expressed in hamster and human cell-lines, and also Aranesp and Mircera, from endogenous urine hEPO. The required amount of EPO in the samples, a few pg, is very low compared to other methods for EPO isoform identification. This EPO isoform determination method opens the possibility to monitor recombinant EPO therapy for clinical research and seems to be a valuable candidate to the arsenal of EPO doping control tests.

  • A genome-wide association study identifies rs2000999 as a strong genetic determinant of circulating haptoglobin levels

    • Haptoglobin is an acute phase inflammatory marker. Its main function is to bind hemoglobin released from erythrocytes to aid its elimination, and thereby haptoglobin prevents the generation of reactive oxygen species in the blood. Haptoglobin levels have been repeatedly associated with a variety of inflammation-linked infectious and non-infectious diseases, including malaria, tuberculosis, human immunodeficiency virus, hepatitis C, diabetes, carotid atherosclerosis, and acute myocardial infarction. However, a comprehensive genetic assessment of the inter-individual variability of circulating haptoglobin levels has not been conducted so far. We used a genome-wide association study initially conducted in 631 French children followed by a replication in three additional European sample sets and we identified a common single nucleotide polymorphism (SNP), rs2000999 located in the Haptoglobin gene (HP) as a strong genetic predictor of circulating Haptoglobin levels (P overall = 8.1×10 -59), explaining 45.4% of its genetic variability (11.8% of Hp global variance). The functional relevance of rs2000999 was further demonstrated by its specific association with HP mRNA levels (β = 0.23±0.08, P = 0.007). Finally, SNP rs2000999 was associated with decreased total and low-density lipoprotein cholesterol in 8,789 European children (P total cholesterol = 0.002 and P LDL = 0.0008). Given the central position of haptoglobin in many inflammation-related metabolic pathways, the relevance of rs2000999 genotyping when evaluating haptoglobin concentration should be further investigated in order to improve its diagnostic/therapeutic and/or prevention impact. © 2012 Froguel et al.

  • Modelling pesticide transport in a shallow groundwater catchment using tritium and helium-3 data

    • Using tritium and helium-3 data for calibration, a 2-D transport model was set up to explain the occurrence of bentazone, dichlorprop, glyphosate, isoproturon, MCPA and metamitron in a small groundwater catchment in southern Sweden. The model was parameterised with site-specific degradation and sorption data to enable transport simulations. Local climatological data and a 21-year record of agricultural pesticide use within the study area were used as boundary conditions. Model output was evaluated against a 7-year long pesticide monitoring data-series from two monitoring wells within the study area. The model successfully predicts observed breakthrough of bentazone, dichlorprop, isoproturon and MCPA. However, it fails to simulate observed occurrences of glyphosate and metamitron. Glyphosate and metamitron exhibit relatively high sorption potential, and their occurrence is suggested to be the result of non-equilibrium preferential flow paths which the model cannot reproduce due the conceptualisation of the system as homogenous and isotropic. The results indicate a promising methodological approach applicable to groundwater contamination risk assessment, and demonstrate the potential for transport model calibration by means of tritium and helium-3 data. Main constraints of the study relate to the relatively simple system conceptualisation, indicating a need for further consideration of physical and chemical heterogeneity. (C) 2014 Elsevier Ltd.     • The demand for vegetable proteins increases globally and seaweeds are considered novel and promising protein sources. However, the tough polysaccharide-rich cell walls and the abundance of polyphenols reduce the extractability and digestibility of seaweed proteins. Therefore, food grade, scalable, and environmentally friendly protein extraction techniques are required. To date, little work has been carried out on developing such methods taking into consideration the structural differences between seaweed species. In this work, three different protein extraction methods were applied to three Swedish seaweeds (Porphyra umbilicalis, Ulva lactuca, and Saccharina latissima). These methods included (I) a traditional method using sonication in water and subsequent ammonium sulfate-induced protein precipitation, (II) the pH-shift protein extraction method using alkaline protein solubilization followed by isoelectric precipitation, and (III) the accelerated solvent extraction (ASE (R)) method where proteins are extracted after pre-removal of lipids and phlorotannins. The highest protein yields were achieved using the pH-shift method applied to P. umbilicalis (22.6 +/- 7.3%) and S. latissima (25.1 +/- 0.9%). The traditional method resulted in the greatest protein yield when applied to U. lactuca (19.6 +/- 0.8%). However, the protein concentration in the produced extracts was highest for all three species using the pH-shift method (71.0 +/- 3.7%, 51.2 +/- 2.1%, and 40.7 +/- 0.5% for P. umbilicalis, U. lactuca, and S. latissima, respectively). In addition, the pH-shift method was found to concentrate the fatty acids in U. lactuca and S. latissima by 2.2 and 1.6 times, respectively. The pH-shift method can therefore be considered a promising strategy for producing seaweed protein ingredients for use in food and feed.

  • Structural and Electrochemical Properties of Functionalized Nanocellulose Materials and Their Biocompatibility

    • Nanocellulose has received considerable interest during the last decade because it is renewable and biodegradable, and has excellent mechanical properties, nanoscale dimensions and wide functionalization possibilities. It is considered to be a unique and versatile platform on which new functional materials can be based.This thesis focuses on nanocellulose from wood (NFC) and from Cladophora algae (CNC), functionalized with surface charges or coated with the conducting polymer polypyrrole (PPy), aiming to study the influence of synthesis processes on structural and electrochemical properties of such materials and assess their biocompatibility.The most important results of the work demonstrated that 1) CNC was oxidized to the same extent using electrochemical TEMPO-mediated oxidation as with conventional TEMPO processes, which may facilitate easier reuse of the reaction medium; 2) NFC and CNC films with or without surface charges were non-cytotoxic as assessed by indirect in vitro testing. Anionic TEMPO-CNC films promoted fibroblast adhesion and proliferation in direct in vitro cytocompatibility testing, possibly due to its aligned fibril structure; 3) Rinsing of PPy-coated nanocellulose fibrils, which after drying into free-standing porous composites are applicable for energy storage and electrochemically controlled ion extraction, significantly degraded the PPy coating, unless acidic rinsing was employed. Only minor degradation was observed during long-term ambient storage; 4) Variations in the drying method as well as type and amount of nanocellulose offered ways of tailoring the porosities of nanocellulose/PPy composites between 30% and 98%, with increments of ~10%. Supercritical CO2-drying generated composites with the largest specific surface area yet reported for nanocellulose/conducting polymer composites (246 m2/g). The electrochemical oxidation rate was found to be controlled by the composite porosity; 5) In blood compatibility assessments for potential hemodialysis applications, heparinization of CNC/PPy composites was required to obtain thrombogenic properties comparable to commercial hemodialysis membranes. The pro-inflammatory characteristics of non-heparinized and heparinized composites were, to some extent, superior to commercial membranes. The heparin coating did not affect the solute extraction capacity of the composite.The presented results are deemed to be useful for tuning the properties of systems based on the studied materials in e.g. energy storage, ion exchange and biomaterial applications.

    • Consider a constraint on a sequence of variables functionally determining a result variable that is unchanged under reversal of the sequence. Most such constraints have a compact encoding via an automaton augmented with accumulators, but it is unknown how to maintain domain consistency efficiently for most of them. Using such an automaton for such a constraint, we derive an implied constraint between the result variables for a sequence, a prefix thereof, and the corresponding suffix. We show the usefulness of this implied constraint in constraint solving, both by local search and by propagation-based systematic search.
