    • Liquid biopsies based on extracellular vesicles (EVs) represent a promising tool for treatment monitoring of tumors, including non-small-cell lung cancers (NSCLC). In this study, we report on a multiplexed electrokinetic sensor for surface protein profiling of EVs from clinical samples. The method detects the difference in the streaming current generated by EV binding to the surface of a functionalized microcapillary, thereby estimating the expression level of a marker. Using multiple microchannels functionalized with different antibodies in a parallel fluidic connection, we first demonstrate the capacity for simultaneous detection of multiple surface markers in small EVs (sEVs) from NSCLC cells. To investigate the prospects of liquid biopsies based on EVs, we then apply the method to profile sEVs isolated from the pleural effusion (PE) fluids of five NSCLC patients with different genomic alterations (ALK, KRAS or EGFR) and applied treatments (chemotherapy, EGFR- or ALKtyrosine kinase inhibitors). The vesicles were targeted against CD9, as well as EGFR and PD-L1, two treatment targets in NSCLC. The electrokinetic signals show detection of these markers on sEVs, highlighting distinct interpatient differences, e.g., increased EGFR levels in sEVs from a patient with EGFR mutation as compared to an ALK-fusion one. The sensors also detect differences in PD-L1 expressions. The analysis of sEVs from a patient prior and post ALK-TKI crizotinib treatment reveals significant increases in the expressions of some markers (EGFR and PD-L1). These results hold promise for the application of the method for tumor treatment monitoring based on sEVs from patient liquid biopsies.

    • The Maribot Vane is an autonomous sailing drone project that has been in the prototype testing phase for two years, developing a fully capable platform to aid in long-term ocean research projects. With longer voyages required for further development, a more reliable physical platform is a high priority of the next-generation design. This paper applies a multi-objective, Reliability-Based Design Optimization (RBDO) approach to minimize the system cost and the probability of failure under anticipated operational conditions. The results will inform designers of the new platform about their trade-offs between the cost and reliability objectives.

    • The approach of inducing protective immunity against cholera by oral vaccination with killed whole Vibrio cholerae cells is effective, but the complexity of current cholera vaccines makes them difficult and relatively expensive to manufacture, especially if recombinant cholera toxin B subunit is included in the formulation. In an effort to simplify the composition of a new generation of oral cholera vaccines we have generated a novel non-toxigenic candidate vaccine strain of V. cholerae O1 that stably expresses both the Ogawa and Inaba serotype antigens on its surface. This was done by introducing a functional wbeT gene without a functional promoter into the chromosome of an O1 Inaba strain. The resulting low levels of expression of the wbeT gene product allowed for the desired partial serotype switching. This strain (MS1342) can potentially replace the three virulent strains used in currently manufactured cholera vaccines. Oral immunization of mice with formalin-killed MS1342 bacteria gave rise to Ogawa-specific, Inaba-specific and cross-reactive serum antibodies that were detectable both by lipopolysaccharide (LPS)-specific ELISAs and as vibriocidal antibodies that are considered to predict protective efficacy. These responses as well as intestinal mucosal IgA anti-LPS antibody responses were fully comparable with those obtained by immunization with the internationally licensed oral cholera vaccine Dukoral ®. We propose that such a strain may form the basis of a single strain killed whole cell cholera vaccine protecting against cholera caused by either the Inaba or Ogawa serotype of V. cholerae O1.

  • Making energy metrics relevant to service firms : from energy conservation to energy productivity

    • Although energy conservation and reduction in environmental impact are on the international and most national agendas, service firms rarely include energy consumption metrics in their strategic decision-making. One reason for the omission is that for service industries, firm level energy utilization is most commonly measured in kilowatt hours per square meter of office space where changes often related to the space rather than the firm performance. The measure also presents several problems for firms in service industries. First, energy conservation and reduction may be counterproductive for service firms that are growing and require energy to sustain that growth. Second, it may not relate to national and international goals which are often focused on the amount of carbon dioxide produced generating energy than the total amount of energy consumed. Third, it treats energy as a utility rather than a resource in firms’ value creation. Results from a field study focused on service firms in Sweden suggests that focusing on energy productivity overcomes the limitations of existing measures and produces positive results. By conceptualizing energy productivity as output per unit of energy, we create a conservation metric that enables service firms to measure their contributions to energy consumption relative to national economic growth. As a result, energy productivity aligns the interests of service organizations with those of policy makers and conservationists.

  • Relativistic radial electron density functions and natural orbitals from GRASP2018

    • A new module, RDENSITY, of the GRASP2018 package [1] is presented for evaluating the radial electron density function of an atomic state described by a multiconfiguration Dirac-Hartree-Fock or configuration interaction wave function in the fully relativistic scheme. The present module is the relativistic version of DENSITY [2] that was developed for the ATSP2K package [3]. The calculation of the spin-angular factors entering in the expression of the expectation value of the density operator is performed using the angular momentum theory in orbital, spin, and quasispin spaces, adopting a generalized graphical technique [4]. The natural orbitals (NOs) are evaluated from the diagonalization of the density matrix, taking advantage of its κ-block structure. The features of the code are discussed in detail, focusing on the advantages and properties of the NOs and on the electron radial density picture as a mean for investigating electron correlation and relativistic effects.

  • Benefits and Challenges of Integration of High-Capacity Autonomous Buses to Public Transport Operations

    • High-capacity autonomous buses can make bus travel safer, smoother, and more energy efficient. However, despite clear societal and environmental benefits there are still several challenges to be addrebed. In this article we present the outcomes of two research projects focusing on the public acceptance of high-capacity autonomous buses. Results indicate high degree of trust towards automation safety. However, in certain conditions, such as driving alone at night, perceived security appears to be an issue. In such circumstances digital forms of assistance replacing drivers will most likely not guarantee a sufficient degree of security. The paper also argues that bus automation is not a silver bullet to make PT more attractive. Instead, it shall be seen as rather an enabler of new applications allowing to efficiently prioritize PT in traffic flows to make it an attractive choice.

  • The green fingerprint : Decreasing energy consumption with decision support systems

    • Sustainable business development is a challenge that requires both organizational change and changes to individual behavior. We report on the design and implementation of an app-based decision support system designed to promote environmentally responsible behavior among individuals in a B2B setting. The app, known as the "Green Fingerprint" was designed in collaboration with a commercial real estate firm and subsequently deployed in several tenant sites in the greater Stockholm area. We measured energy productivity defined as revenue in Swedish Kronor/kilowatt hours of electricity consumption. Over a two year period participating firms were able to significantly increase energy productivity by as much as much as 100% as a result of direct feedback and adopting conservation techniques recommended by the system. Increased energy productivity resulted in annual energy costs that were substantially less than if energy productivity had not increased.
