    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • A scoping review of trials of interventions led or delivered by cancer nurses

    • BACKGROUND: Advances in research and technology coupled with an increased cancer incidence and prevalence have resulted in significant expansion of cancer nurse role, in order to meet the growing demands and expectations of people affected by cancer (PABC). Cancer nurses are also tasked with delivering an increasing number of complex interventions as a result of ongoing clinical trials in cancer research. However much of this innovation is undocumented, and we have little insight about the nature of novel interventions currently being designed or delivered by cancer nurses.OBJECTIVES: To identify and synthesise the available evidence from clinical trials on interventions delivered or facilitated by cancer nurses.DATA SOURCES AND REVIEW METHODS: A systematic review of randomised controlled trials (RCT), quasi-RCTs and controlled before and after studies (CBA) of cancer nursing interventions aimed at improving the experience and outcomes of PABC. Ten electronic databases (CENTRAL, MEDLINE, AMED, CINAHL, EMBASE, Epistemonikos, CDSR, DARE, HTA, WHO ICTRP) were searched between 01 January 2000 and 31 May 2016. No language restrictions were applied. Bibliographies of selected studies and relevant Cochrane reviews were also hand-searched. Interventions delivered by cancer nurses were classified according to the OMAHA System. Heat maps were used to highlight the volume of evidence available for different cancer groups, intervention types and stage of cancer care continuum.RESULTS: The search identified 22,450 records; we screened 16,169 abstracts and considered 925 full papers, of which 214 studies (247,550 participants) were included in the evidence synthesis. The majority of studies were conducted in Europe (n = 79) and USA (n = 74). Interventions were delivered across the cancer continuum from prevention and risk reduction to survivorship, with the majority of interventions delivered during the treatment phase (n = 137). Most studies (131/214) had a teaching, guidance or counselling component. Cancer nurse interventions were targeted at primarily breast, prostate or multiple cancers. No studies were conducted in brain, sarcoma or other rare cancer types. The majority of the studies (n = 153) were nurse-led and delivered by specialist cancer nurses (n = 74) or advanced cancer nurses (n = 29), although the quality of reporting was poor.CONCLUSIONS: To the best of our knowledge, this is the first review to synthesise evidence from intervention studies across the entire cancer spectrum. As such, this work provides new insights into the nature of the contribution that cancer nurses have made to evidence-based innovations, as well as highlighting areas in which cancer nursing trials can be developed in the future.

  • AusTraits, a curated plant trait database for the Australian flora

    • We introduce the AusTraits database - a compilation of values of plant traits for taxa in the Australian flora (hereafter AusTraits). AusTraits synthesises data on 448 traits across 28,640 taxa from field campaigns, published literature, taxonomic monographs, and individual taxon descriptions. Traits vary in scope from physiological measures of performance (e.g. photosynthetic gas exchange, water-use efficiency) to morphological attributes (e.g. leaf area, seed mass, plant height) which link to aspects of ecological variation. AusTraits contains curated and harmonised individual- and species-level measurements coupled to, where available, contextual information on site properties and experimental conditions. This article provides information on version 3.0.2 of AusTraits which contains data for 997,808 trait-by-taxon combinations. We envision AusTraits as an ongoing collaborative initiative for easily archiving and sharing trait data, which also provides a template for other national or regional initiatives globally to fill persistent gaps in trait knowledge.

  • The Holocene vegetation cover of Britain and Ireland : overcoming problems of scale and discerning patterns of openness

    • The vegetation of Europe has undergone substantial changes during the course of the Holocene epoch, resulting from range expansion of plants following climate amelioration, competition between taxa and disturbance through anthropogenic activities. Much of the detail of this pattern is understood from decades of pollen analytical work across Europe, and this understanding has been used to address questions relating to vegetation-climate feedback, biogeography and human impact. Recent advances in modelling the relationship between pollen and vegetation now make it possible to transform pollen proportions into estimates of vegetation cover at both regional and local spatial scales, using the Landscape Reconstruction Algorithm (LRA), i.e. the REVEALS (Regional Estimates of VEgetation Abundance from Large Sites) and the LOVE (Local VEgetation) models. This paper presents the compilation and analysis of 73 pollen stratigraphies from the British Isles, to assess the application of the LRA and describe the pattern of landscape/woodland openness (i.e. the cover of low herb and bushy vegetation) through the Holocene. The results show that multiple small sites can be used as an effective replacement for a single large site for the reconstruction of regional vegetation cover. The REVEALS vegetation estimates imply that the British Isles had a greater degree of landscape/woodland openness at the regional scale than areas on the European mainland. There is considerable spatial bias in the British Isles dataset towards wetland areas and uplands, which may explain higher estimates of landscape openness compared with Europe. Where multiple estimates of regional vegetation are available from within the same region inter-regional differences are greater than intra-regional differences, supporting the use of the REVEALS model to the estimation of regional vegetation from pollen data.   • Microvascular stabilization via blood-brain barrier regulation prevents seizure activity

    • Blood-brain barrier (BBB) dysfunction is associated with worse epilepsy outcomes however the underlying molecular mechanisms of BBB dysfunction remain to be elucidated. Tight junction proteins are important regulators of BBB integrity and in particular, the tight junction protein claudin-5 is the most enriched in brain endothelial cells and regulates size-selectivity at the BBB. Additionally, disruption of claudin-5 expression has been implicated in numerous disorders including schizophrenia, depression and traumatic brain injury, yet its role in epilepsy has not been fully deciphered. Here we report that claudin-5 protein levels are significantly diminished in surgically resected brain tissue from patients with treatment-resistant epilepsy. Concomitantly, dynamic contrast-enhanced MRI in these patients showed widespread BBB disruption. We show that targeted disruption of claudin-5 in the hippocampus or genetic heterozygosity of claudin-5 in mice exacerbates kainic acid-induced seizures and BBB disruption. Additionally, inducible knockdown of claudin-5 in mice leads to spontaneous recurrent seizures, severe neuroinflammation, and mortality. Finally, we identify that RepSox, a regulator of claudin-5 expression, can prevent seizure activity in experimental epilepsy. Altogether, we propose that BBB stabilizing drugs could represent a new generation of agents to prevent seizure activity in epilepsy patients.

  • Plant water use related to leaf traits and CSR strategies of 10 common European green roof species

    • The vegetation layer contributes to multiple functions of green roofs including their hydrological function as plants remove water from substrates between rainfall events through evapotranspiration, restoring the green roofs storage capacity for rainfall retention. While individual traits have been related to water use strategies of green roof plants, these traits are inconsistent, suggesting the importance of trait combinations which may be reflected in CSR (competitor, stress tolerator, ruderal) strategies. Therefore, relating plant water use to leaf traits and CSR strategies could help facilitate green roof plant selection into new geographical regions where green roof technology is developing. For example, in high latitude northern European regions with long daylight during the growing season. Growth (shoot biomass, relative growth rate and leaf area), leaf traits (leaf dry matter content, specific leaf area and succulence) and CSR strategies were determined of 10 common European green roof plants and related to their water use under well-watered (WW) and water-deficit (WD) conditions. All three succulent species included in the experiment showed mostly stress tolerant traits and their water loss was less than the bare unplanted substrate, likely due to mulching of the substrate surface. Plants with greater water use under WW conditions had more ruderal and competitive strategies, and greater leaf area and shoot biomass, than species with lower WW water use. However, the four species with the highest water use under WW conditions were able to downregulate their water use under WD, indicating that they could both retain rainfall and survive periods of water limitations. This study indicates that, for optimal stormwater retention, green roof plant selection in high latitude regions like northern Europe, should focus on selecting non-succulent plants with predominantly competitive or ruderal strategies to make the most of the long daylight during the short growing season.

  • Socio-Ecological Dimensions of Spontaneous Plants on Green Roofs

    • Green roofs have the potential to provide socio-ecological services in urban settings that lack vegetation and open space. However, implementation of green roofs is limited by high construction and maintenance costs. Consequently, green roof projects often disproportionately benefit wealthy communities and can further marginalise disadvantaged communities by increasing property values and housing costs. Vegetation cover on green roofs is crucial to their provisioning of socio-ecological services. Evidence suggests that green roof plantings change over time, especially with limited maintenance, and are replaced with spontaneous “weedy” species. This is often perceived as a failure of the original green roof design intent and spontaneous species are usually removed. However, where good coverage is achieved, spontaneous vegetation could provide beneficial services such as stormwater mitigation, habitat provision, and climate regulation. While social norms about “weediness” may limit the desirability of some spontaneous species, research suggests that their acceptability on green roofs increases with coverage. As spontaneous species can establish on green roofs without irrigation and fertiliser, reduced input costs could help facilitate adoption particularly in markets without an established green roof industry. Construction costs may also be reduced in hot and dry climates where deeper substrates are necessary to ensure plant survival, as many spontaneous species are able to colonise shallow substrates and can regenerate from seed. If implemented based on socio-ecological need, green roofs with spontaneous vegetation coverage may apply less pressure to property values and housing costs than conventionally planted green roofs, increasing the resilience of urban communities while limiting gentrification.

  • Selecting tree species with high transpiration and drought avoidance to optimise runoff reduction in passive irrigation systems

    • Rainfall in cities can generate large volumes of stormwater runoff which degrades receiving waterways. Irrigating trees with runoff (passive irrigation) has the potential to increase transpiration and contribute to stormwater management by reducing runoff received by downstream waterways, but the stochastic nature of rainfall may expose trees with high transpiration to drought stress. We hypothesized that for success in passive irrigation systems, tree species should exhibit i) high maximum transpiration rates under well-watered conditions, ii) drought avoidance between rainfall events, and iii) high recovery of transpiration with rainfall following a drought. We assessed 13 commonly planted urban tree species in Melbourne, Australia against three metrics representing these behaviours (crop factor, hydroscape area, and transpiration recovery, respectively) in a glasshouse experiment. To aid species selection, we also investigated the relationships between these three metrics and commonly measured plant traits, including leaf turgor loss point, wood density, and sapwood to leaf area ratio (Huber value). Only one species (Tristaniopsis laurina) exhibited a combination of high crop factor (>1.1 mm mm−1 d−1) indicating high transpiration, small hydroscape area (<3 MPa2) indicating drought avoidance, and high transpiration recovery (>85%) following water deficit. Hence, of the species measured, it had the greatest potential to reduce runoff from passive irrigation systems while avoiding drought stress. Nevertheless, several other species showed moderate transpiration, hydroscape areas and transpiration recovery, indicating a balanced strategy likely suitable for passive irrigation systems. Huber values were negatively related to crop factor and transpiration recovery and may therefore be a useful tool to aid species selection. We propose that selecting tree species with high transpiration rates that can avoid drought and recover well could greatly reduce stormwater runoff, while supporting broader environmental benefits such as urban cooling in cities.
