  • From agroindustries to integrated biomass logistics centres. Agroinlog project : Summary of final results

    • AGROinLOG project has tested the integrated biomass logistics centres (IBLC) concept in three real agro-industries in Europe. The relevance of the IBLC strategy relies on the fact that it allows agro-industries to create a new activity with lower investment, increasing incomes, stabilizing their annual activity (avoiding idle periods) and maintaining or creating new jobs. The demos’ studies were performed in Spain at a fodder industry, in Greece at an olive oil industry, and in Sweden inside a cereal processing industry. AGROinLOG validated these demos´ business models from a holistic perspective, also studying the replicability of the IBLC business model in other agro-industries from different sectors (vegetable oil extraction, olive oil chain, feed & fodder, wine, grain chain and sugar industry). Sectorial analysis was carried out as well, allowing the identification of opportunities among the targeted sector to replicate the IBLC concept, drawing barriers to overcome in each case. Thus, technical, economic and environmental feasibility of integrated biomass logistics centers (IBLCs) for food and non-food products have been assessed in detail.

  • Magnetic, Structural, and Particle Size Analysis of Single- and Multi-Core Magnetic Nanoparticles

    • We have measured and analyzed three different commercial magnetic nanoparticle systems, both multi-core and single-core in nature, with the particle (core) size ranging from 20 to 100 nm. Complementary analysis methods and same characterization techniques were carried out in different labs and the results are compared with each other. The presented results primarily focus on determining the particle size-both the hydrodynamic size and the individual magnetic core size-as well as magnetic and structural properties. The used analysis methods include transmission electron microscopy, static and dynamic magnetization measurements, and Mossbauer spectroscopy. We show that particle (hydrodynamic and core) size parameters can be determined from different analysis techniques and the individual analysis results agree reasonably well. However, in order to compare size parameters precisely determined from different methods and models, it is crucial to establish standardized analysis methods and models to extract reliable parameters from the data.

  • Nitrogen losses following application of pig slurry to arable land

    • Emissions of ammonia (NH3) and nitrous oxide (N2O), and nitrate (NO3/-) leaching were measured in two field experiments following application of pig slurry at rates corresponding to 83-96kg NH4-N ha-1 before sowing. In spring and in autumn 1994, slurry was applied by four methods: trenching (T), shallow injection (S), band spreading immediately followed by harrowing (B/H) and band spreading (B). NH3 emission measurements were made during the first week after application in both experiments. In the spring experiment N2O emissions and NO3/- leaching were measured during 6 and 52 weeks after spreading respectively, and during 11-and 33 weeks after spreading in the autumn experiment. In spring, the increased N2O emissions (i.e. control subtracted) ranged from 0.27% (T) to 0.45% (B/H), and in the autumn study from 0.92% (T) to 1.14% (B/H), of applied NH4-N, although showing no statistically significant differences. In order to validate the chamber measurements, a 'megachamber' (21 m2) was used together with an infrared spectrometer. The emissions agreed well for (B/H), while (B) resulted in lower emissions compared with the smaller chambers. Emissions of NH3 were about one order of magnitude higher. In spring, (B) gave the highest emission, reaching 19.5% of applied NH4-N, whereas (S), and (B/H) gave the lowest emissions, reaching 1.2 and 3.5% of applied NH4-N, respectively. NH3 emissions in autumn were 15-20% lower compared with spring. In spring the increased nitrate leaching ranged from 10.1 (T) to 24.9 kg ha-1 (B/H) and from 29.5 (B) to 37.8 kg ha-1 (T) in the autumn experiment, showing no statistically significant differences. Estimations of indirect N2O emissions due to ammonia deposition and nitrate leaching, suggested that the N2O contribution from NH3 deposition was relatively small, while the indirect N2O emissions from NO3/- leaching were of the same order of magnitude or higher than the direct N2O emissions.

  • Understanding interactions of plasticisers with a phospholipid monolayer

    • The use of DEHP (diethylhexyl phthalate) is now banned for most applications in Europe; the exception is for blood bags, where its toxicity is overshadowed by its ability to extend the storage life of red blood cells. Another plasticiser, BTHC (butanoyl trihexyl citrate), is used in paediatric blood bags but does not stabilise blood cells as effectively. Interactions between plasticisers and lipids are investigated with a phospholipid, DMPC, to understand the increased stability of blood cells in the presence of DEHP as well as bioaccumulation and identify differences with BTHC. Mixed monolayers of DMPC and DEHP or BTHC were studied on Langmuir troughs where surface pressure/area isotherms can be measured. Neutron reflection measurements were made to determine the composition and structure of these mixed layers. A large amount of plasticiser can be incorporated into a DMPC monolayer but once an upper limit is reached, plasticiser is selectively removed from the interface at high surface pressures. The upper limit is found to occur between 40-60 mol% for DEHP and 20-40 mol% for BTHC. The areas per molecule are also different with DEHP being in the range of 50-100 Å2 and BTHC being 65-120 Å2. Results indicate that BTHC does not fit as well as DEHP in DMPC monolayers which could help explain the differences observed with regards to the stability of blood cells.

    • A first-generation oral inactivated whole-cell enterotoxigenic Escherichia coli (ETEC) vaccine, comprising formalin-killed ETEC bacteria expressing different colonization factor (CF) antigens combined with cholera toxin B subunit (CTB), when tested in phase III studies did not significantly reduce overall (generally mild) ETEC diarrhea in travelers or children although it reduced more severe ETEC diarrhea in travelers by almost 80%. We have now developed a novel more immunogenic ETEC vaccine based on recombinant non-toxigenic E. coli strains engineered to express increased amounts of CF antigens, including CS6 as well as an ETEC-based B subunit protein (LCTB. A), and the optional combination with a nontoxic double-mutant heat-labile toxin (LT) molecule (dmLT) as an adjuvant.Two test vaccines were prepared under GMP: (1) A prototype E. coli CFA/I-only formalin-killed whole-cell. +. LCTB. A vaccine, and (2) A "complete" inactivated multivalent ETEC-CF (CFA/I, CS3, CS5 and CS6 antigens) whole-cell. +. LCTB. A vaccine. These vaccines, when given intragastrically alone or together with dmLT in mice, were well tolerated and induced strong intestinal-mucosal IgA antibody responses as well as serum IgG and IgA responses to each of the vaccine CF antigens as well as to LT B subunit (LTB). Both mucosal and serum responses were further enhanced (adjuvanted) when the vaccines were co-administered with dmLT. We conclude that the new multivalent oral ETEC vaccine, both alone and especially in combination with the dmLT adjuvant, shows great promise for further testing in humans.

  • Individualized tissue-engineered veins as vascular grafts: A proof of concept study in pig

    • Personalized tissue engineered vascular grafts are a promising advanced therapy medicinal product alternative to autologous or synthetic vascular grafts utilized in blood vessel bypass or replacement surgery. We hypothesized that an individualized tissue engineered vein (P-TEV) would make the body recognize the transplanted blood vessel as autologous, decrease the risk of rejection and thereby avoid lifelong treatment with immune suppressant medication as is standard with allogenic organ transplantation. To individualize blood vessels, we decellularized vena cava from six deceased donor pigs and tested them for cellular removal and histological integrity. A solution with peripheral blood from the recipient pigs was used for individualized reconditioning in a perfusion bioreactor for seven days prior to transplantation. To evaluate safety and functionality of the individualized vascular graft in vivo, we transplanted reconditioned porcine vena cava into six pigs and analyzed histology and patency of the graft at different time points, with three pigs at the final endpoint 4-5 weeks after surgery. Our results showed that the P-TEV was fully patent in all animals, did not induce any occlusion or stenosis formation and we did not find any signs of rejection. The P-TEV showed rapid recellularization in vivo with the luminal surface covered with endothelial cells. In summary, the results indicate that P-TEV is functional and have potential for use as clinical transplant grafts.
