  • Identification and functional characterization of a novel susceptibility locus for small vessel vasculitis with MPO-ANCA

    • Objective To identify and characterize genetic loci associated with the risk of developing ANCA-associated vasculitides (AAV). Methods Genetic association analyses were performed after Illumina sequencing of 1853 genes and subsequent replication with genotyping of selected single nucleotide polymorphisms in a total cohort of 1110 Scandinavian cases with granulomatosis with polyangiitis or microscopic polyangiitis, and 1589 controls. A novel AAV-associated single nucleotide polymorphism was analysed for allele-specific effects on gene expression using luciferase reporter assay. Results PR3-ANCA(+) AAV was significantly associated with two independent loci in the HLA-DPB1/HLA-DPA1 region [rs1042335, P = 6.3 x 10(-61), odds ratio (OR) 0.10; rs9277341, P = 1.5 x 10(-44), OR 0.22] and with rs28929474 in the SERPINA1 gene (P = 2.7 x 10(-10), OR 2.9). MPO-ANCA(+) AAV was significantly associated with the HLA-DQB1/HLA-DQA2 locus (rs9274619, P = 5.4 x 10(-25), OR 3.7) and with a rare variant in the BACH2 gene (rs78275221, P = 7.9 x 10(-7), OR 3.0), the latter a novel susceptibility locus for MPO-ANCA(+) granulomatosis with polyangiitis/microscopic polyangiitis. The rs78275221-A risk allele reduced luciferase gene expression in endothelial cells, specifically, as compared with the non-risk allele. Conclusion We identified a novel susceptibility locus for MPO-ANCA(+) AAV and propose that the associated variant is of mechanistic importance, exerting a regulatory function on gene expression in specific cell types.

  • Sex differences in academic strengths contribute to gender segregation in education and occupation : A longitudinal examination of 167,776 individuals

    • We investigate whether sex differences in academic strengths have an impact on society by affecting the career choices made by women and men. By longitudinally following 167,776 individuals from Sweden, we found that (1) more 16-year old girls than boys had a relative strength in verbal/language school subjects than in technical/numerical ones, whereas more boys than girls had a relative strength in technical/numerical school subjects than in verbal/language ones; (2) when these girls and boys attained higher education and entered employment, they largely pursued careers cognitively matching their initial academic strengths; (3) while individuals generally made career choices in line with their academic strengths, men and women matched on these strengths nevertheless made rather distinct career choices, in particular women with technical/numerical strengths who largely avoided careers demanding these skills; (4) sex distribution in education and occupation was related to the extent these career paths were perceived as either numerically or verbally demanding. Taken together, although gender segregation is to some extent associated with individuals making choices matching their academic strengths, the vast discrepancies in career outcomes between men and women can be only in part attributed to sex differences in academic performance.

    • Sex differences favoring women have been found in a number of studies of episodic memory. This study examined sex differences in verbal, nonverbal, and visuospatial episodic memory tasks. Results showed that although women performed at a higher level on a composite verbal and nonverbal episodic memory score, men performed at a higher level on a composite score of episodic memory tasks requiring visuospatial processing. Thus, men can use their superior visuospatial abilities to excel in highly visuospatial episodic memory tasks, whereas women seem to excel in episodic memory tasks in which a verbalization of the material is possible.

  • Sex differences in face recognition – Women’s faces make the difference

    • Sex differences favoring women have been found in face recognition tasks as well as in verbal episodic memory tasks. Women's higher face recognition performance was hypothesized to be related to either their higher verbal ability or to their superiority in recognizing female faces, rather than faces in general. Results showed that whereas there were no differences between Swedish men and women (aged 20-40 yrs) in the recognition of male faces, or in verbal ability, women performed at a higher level than men in the recognition of female faces. Verbal ability did not influence women's face recognition performance. Potential explanations for this pattern of data, such as sex differences in interest and prior knowledge, are discussed.

  • Medial temporal lobe is vulnerable to vascular risk factors in men : a population based study

    • Background and purpose: Vascular risk factors (VRFs) are known to cause cerebral microvascular disease, but evidence supporting an effect of VRFs on regional brain atrophy is mixed. We investigate whether an aggregation of VRFs is associated with volume of hippocampus and entorhinal cortex in elderly people living in the community. Methods: This cross-sectional study consists of 523 participants (age =60 years, 59.3% women) of the SNAC-K Study in central Stockholm, Sweden, who were free of clinical stroke and cognitive impairment. We collected data on VRFs through interviews, clinical examination and inpatient register system. Hippocampal and entorhinal cortex volume was manually measured on magnetic resonance images. Data were analysed with general linear regression models controlling for demographics and total intracranial volume. Results: In men, high total cholesterol and diabetes were significantly or marginally associated with smaller hippocampus and entorhinal cortex; when current smoking, binge alcohol drinking, high cholesterol and diabetes were aggregated, an increasing number of VRFs were significantly associated with decreasing volume of hippocampus and entorhinal cortex (P for linear trend <0.01). In women, none of individual VRFs or their aggregation was significantly associated with the volume of these brain regions, except former smoking that was significantly associated with a larger volume of these regions. Conclusions: Aggregation of VRFs is associated with reduced hippocampal and entorhinal cortex volume in apparently healthy elderly men, but not in women. This implies that in men, the medial temporal lobe is vulnerable to cardiovascular risk factors.
