    • Energetic nutrients are oxidized to sustain high intracellular NADPH/NADP(+) ratios. NADPH-dependent reduction of thioredoxin-1 (Trx1) disulfide and glutathione disulfide by thioredoxin reductase-1 (TrxR1) and glutathione reductase (Gsr), respectively, fuels antioxidant systems and deoxyribonucleotide synthesis. Mouse livers lacking both TrxR1 and Gsr sustain these essential activities using an NADPH-independent methionine-consuming pathway; however, it remains unclear how this reducing power is distributed. Here, we show that liver-specific co-disruption of the genes encoding Trx1, TrxR1, and Gsr (triplenull) causes dramatic hepatocyte hyperproliferation. Thus, even in the absence of Trx1, methionine-fueled glutathione production supports hepatocyte S phase deoxyribonucleotide production. Also, Trx1 in the absence of TrxR1 provides a survival advantage to cells under hyperglycemic stress, suggesting that glutathione, likely via glutaredoxins, can reduce Trx1 disulfide in vivo. In triple-null livers like in many cancers, deoxyribonucleotide synthesis places a critical yet relatively low-volume demand on these reductase systems, thereby favoring high hepatocyte turnover over sustained hepatocyte integrity.

    • Chronic, low intensity herbivory by invertebrates, termed background herbivory, has been understudied in tundra, yet its impacts are likely to increase in a warmer Arctic. The magnitude of these changes is however hard to predict as we know little about the drivers of current levels of invertebrate herbivory in tundra. We assessed the intensity of invertebrate herbivory on a common tundra plant, the dwarf birch (Betula glandulosa-nana complex), and investigated its relationship to latitude and climate across the tundra biome. Leaf damage by defoliating, mining and gall-forming invertebrates was measured in samples collected from 192 sites at 56 locations. Our results indicate that invertebrate herbivory is nearly ubiquitous across the tundra biome but occurs at low intensity. On average, invertebrates damaged 11.2% of the leaves and removed 1.4% of total leaf area. The damage was mainly caused by external leaf feeders, and most damaged leaves were only slightly affected (12% leaf area lost). Foliar damage was consistently positively correlated with mid-summer (July) temperature and, to a lesser extent, precipitation in the year of data collection, irrespective of latitude. Our models predict that, on average, foliar losses to invertebrates on dwarf birch are likely to increase by 6-7% over the current levels with a 1 degrees C increase in summer temperatures. Our results show that invertebrate herbivory on dwarf birch is small in magnitude but given its prevalence and dependence on climatic variables, background invertebrate herbivory should be included in predictions of climate change impacts on tundra ecosystems.

    • We have isolated a cDNA sequence coding for a part of human tissue plasminogen activator. mRNA coding for tissue plasminogen activator was partially purified, copied into double-stranded cDNA, and cloned into Escherichia coli. Two sets of partially overlapping oligodeoxynucleotide mixtures corresponding to all possible coding sequences for a known portion of the tissue plasminogen activator gene were prepared. One set was used as a probe to screen cDNA containing bacterial clones and both were used as probes in hybridization against purified plasmid DNA. Of 4,200 bacterial clones examined, 1 carried a plasmid that hybridized to both sets of oligonucleotides. This plasmid contained a 370-base-pair cDNA insert, which was shown by nucleotide sequence analysis to code for the cleavage site region in the one-chain form of the human tissue plasminogen activator.

    • Fragile-X mental retardation (FRAX-MR) is one of the more common X-linked disorders affecting 1 in 1,500 newborn males. This disease is characterized by the expression of fragile site in the region q27.3 of the X-chromosome of affected boys when their lymphocytes are cultured in folate deficient medium. In most patients there is macroorchidism postpubertally. The clinical diagnosis of carrier females based on the expression of fragile site in Xq27.3 is usually difficult and sometimes impossible. About half of the carrier females escape diagnosis by this method. Furthermore, prenatal diagnosis is not always feasible. Using Restriction Fragment Length Polymorphism (RFLP) and cloned DNA segments from the region Xq27-Xqter as probes, we have investigated Swedish families with FRAX-MR in three generations. Interesting observations, previously unreported to our knowledge, have been made in some patients and carrier mothers, using one of the probes which is localized to the distal end of Xq. The significance of these findings and the linkage of the disease locus to the different probes used in this study is presented.

  • Swedish forest commons : A matter of governance?

    • Around 100 years ago, when Crown land in the interior of northern Sweden was privatized, part of the forest land was set aside as forest commons. Today, there are 33 such forest commons jointly managed and owned in common mainly by private forest owners. The forest commons may be looked upon as a means by which the state controls the production of and returns from the forests belonging to small and less affluent forest owners. Further, an attempt has been made to use the forests as a tool to move the self-interests of these small forest owners closer to providing public goods. Forest commons thus hold a contested status, as private lands under public control and as a partly de-regulated form of ownership. This paper examines the extent to which forest commons are currently managed directly by the government, comparing this with the general trend in forest policy towards governance and less prescriptive measures, which often take account of market and participative goals. Building upon Appelstrand (2007), this paper describes the major policy instruments relevant for forest commons from 1861 to 2005. We conclude that direct government management remains dominant, with the major legislation pertaining to forest commons dating back to the 1950s. While governance may seem to be inherent in the forest commons concept, the development of governance has not been fully realised given the relatively strict government-steered framework.

  • Shareholder perceptions of individual and common benefits in Swedish forest commons

    • In the period 1861 to 1918, thirty-three commons were established in Northern Sweden. This was linked to the finalisation of the Great Redistribution of Forest Holdings in Dalarna and the delimitation process in Västerbotten and Norrbotten. They were intended to serve as an instrument for improved and sustained forest production, the viability of farmers and the liveability of the rural communities in the areas where they were established. The aim of this paper is to describe the results of a study examining how three of these forest commons, one from each region, have benefitted the local shareholders and their community. The perceptions among forest common shareholders were assessed using a questionnaire. The study also assessed economic impact on shareholders in terms of extent and use of the dividend from each of the commons for the period 1958-2007, highlighting the extent of the economic support to individual shareholders and to the local community. Results reveal large differences between the three cases; there was a positive correlation between the extent of the economic support and contentment among the shareholders.

    • Objective: The aim of the present study was to illuminate the contemporary cultural reality of being a young Sami in Sweden, with special reference to issues such as identity/self-perception, autonomy, and experiences of being ill treated and discrimination.Design: The study comprises a qualitative and a quantitative part. The qualitative part includes meetings, discussions and dialogues with young Sami and others. The quantitative part includes a questionnaire on socioeconomic conditions, Sami ethnicity, experiences of being ill-treated because of a Sami background, specific questions on identity and self-perception, questions about self-determination, and thoughts and expectations of the future. The sample consists of 876 young Sami aged 18–28, of whom 516 (59 per cent) responded to the questionnaire.Results: A majority are proud to be Sami and wish to preserve their culture. 71 per cent have a close connection to a Sami community. Most of the young Sami have had to explain and defend their culture and way of life. Nearly half had perceived discrimination or ill-treatment because of their ethnicity, with reindeer herders reporting a higher degree of ill-treatment (70 per cent). Reindeer herders exist in a severe environment with an insecure legacy. Most of the young Sami in this study have a positive self-perception and think that their lives are meaningful. Very few dropped out of school and very few are unemployed.Conclusion: We believe that there are protective factors that potentially explain the well-being of this group; a strong feeling of belonging among the Sami, strong connections to family, relatives and friends and good sociocultural adaptation (to have a job, completed school).
