    • Lack of user acceptance of telemedicine services is an important barrier to deployment and stresses the need for involving users, i.e. medical professionals. However, the involvement of users in the service development process of telemedicine services is difficult because of (a) the knowledge gap between the expertise of medical and technical experts; (b) the language gap, i.e. the use of different terminologies between the medical and the technical professions; and (c) the methodological gap in applying requirement methods to multidisciplinary scientific matters. We have developed a guideline in which the medical and technical domains meet. The guideline can be used to develop a scenario from which requirements can be elicited. In a retrospective analysis of a myofeedback-based teletreatment service, the technically-oriented People-Activities-Context-Technology (PACT) framework and medically-oriented principles of evidence-based medicine were incorporated into a guideline. The guideline was developed to construct the content of a scenario which describes the new teletreatment service. This allows the different stakeholders to come together and develop the service. Our approach provides an arena for different stakeholders to take part in the early stages of the design process. This should increase the chance of user acceptance and thus adoption of the service being developed.

    • Ekosystemtjänster kan bidra till att synliggöra värden i naturen som människor är beroende av för sin överlevnad och sin välfärd. Värdet av ekosystemtjänster behöver också tydligt och systematiskt integreras i politiska beslut och i förvaltningen av våra gemensamma naturresurser, på alla dess nivåer.Övergödningen av Östersjön är ett fortsatt stort problem. Projektet Values har visat hur kartering, kvantifiering och värdering av ekosystemfunktioner kan användas för att visa på värde och nytta av olika typer av ekosystemtjänster i relation till mänsklig påverkan i form av invasiva arter och övergödning i två studiesystem, djupa sedimentbottnar och grunda kustområden.I rapporten visas att vi för att kunna ta hänsyn till ekosystemtjänster i beslutsfattande och förvaltning, behöver kartlägga dem och övervaka förändringar i deras tillstånd. Studien visar på ett konkret sätt hur begreppet ekosystemtjänster kan komma till praktisk nytta.Den här rapporten är ett resultat från ett av sju projekt inom forskningssatsningen Värdet av ekosystemtjänster och har författats av forskare vid AquaBiota Water Research, SLU och SU Östersjöcentrum. Projektet finansierades av Naturvårdsverkets miljöforskningsanslag.

  • Comparative biogeography and the evolution of population structure for bottlenose and common dolphins in the Indian Ocean

    • Aim: In the marine environment, where there are few physical boundaries to gene flow, there is often nevertheless intraspecific diversity with consequences for effective conservation and management. Here, we compare two closely related dolphin species with a shared distribution in the Indian Ocean (IO) to better understand the biogeographic drivers of their population structure.Location: Global oceans and seas with a focus on the Indian OceanTaxon: Tursiops sp. and Delphinus sp.Methods: Bayesian, ordination, assignment, statistical and phylogenetic analyses to assess phylogeography, connectivity and population structure using microsatellite and mitochondrial DNA genetic markers.Results: Both Tursiops sp. and Delphinus sp. showed population structure across the western IO and, in each case, populations in the Arabian Sea (off India, Pakistan and Oman) were most differentiated. Comparisons with other populations worldwide revealed independent lineages in this geographic region for both genera. For T. aduncus, (for which multiple sites within the IO could be compared), Bayesian modelling best supported a scenario of expansion southwards following a bottleneck event resulting in differentiation between the northern and western IO. For Delphinus, the same pattern is even more pronounced. Populations in the Arabian Sea region of the north-western IO show genetic isolation for each of the two genera, consistent with other studies of cetacean species in this region.Main conclusions: We propose that changes in the intensity of the southwest monsoon during the climate cycles of the Pleistocene could have affected regional patterns of productivity and represent an important biogeographic driver promoting the observed patterns of differentiation and population dynamics seen in our focal species. Patterns of population genetic structure are consistent with phenotypic differences, suggesting an influence from distinct habitats and resources, and emphasising the need for effective conservation measures in this geographic region.

  • S100A4 regulates the Src-tyrosine kinase dependent differentiation of Th17 cells in rheumatoid arthritis

    • Objectives: To evaluate the role of S100A4, a calcium-binding regulator of nonmuscle myosin assembly, for T-cell responses in rheumatoid arthritis. Methods: Arthritis was induced in the methylated bovine serum albumin (mBSA)-immunized mice lacking the entire S100A4 protein (S100A4KO) and in wild-type counterparts treated with short hairpin ribonucleic acid (shRNA)-lentiviral constructs targeting S100A4 (S100A4-shRNA). The severity of arthritis was evaluated morphologically. T-cell subsets were characterized by the expression of master transcription factors, and functionally by proliferation activity and cytokine production. The activity of the Scr-kinases Fyn and Lck was assessed by the autophosphorylation of C-terminal thyrosine and by the phosphorylation of the CD5 cytodomain. The interaction between S100A4 and the CD5 cytodomain was analysed by nuclear magnetic resonance spectrophotometry. Results: S100A4-deficient mice (S100A4KO and S100A4-shRNA) had significantly alleviated morphological signs of arthritis and joint damage. Leukocyte infiltrates in the arthritic joints of S100A4-deficient mice accumulated Foxp3(+) Treg cells, while the number of ROR gamma t(+) and (pTyr705)STAT3(+) cells was reduced. S100A4-deficient mice had a limited formation of Th17-cells with low retinoic acid orphan receptor gamma t (ROR gamma t) mRNA and IL17 production in T-cell cultures. S100A4-deficient mice had a low expression and activity of T-cell receptor (TCR) inhibitor CD5 and low (pTyr705)STAT3 (signal transducer and activator of transcription 3), which led to increased (pTyr352)ZAP-70 (theta-chain associated protein kinase of 70 kDa), lymphocyte proliferation and production of IL2. In vitro experiments showed that S100A4 directly binds Lck and Fyn and reciprocally regulates their kinase activity towards the CD5 cytodomain. Spectrometry demonstrates an interaction between the CD5 cytodomain and EF2-binding sites of S100A4. Conclusion: The present. study demonstrates that S100A4 plays an important part in the pathogenesis of arthritis. It controls CD5-dependent differentiation of Th17 cells by regulating the activity of the Src-family kinases Lck and Fyn. (C) 2014 Elsevier B.V. All rights reserved.

    • There are several environmental and ecological effects of shipping. However, these are rarely assessed in total in the scientific literature. Thus, the aim of this study was to summarize the different impacts of water-based transport on aquatic and nearshore ecosystems and to identify knowledge gaps and areas for future research. The review identified several environmental and ecological consequences within the main impact categories of water discharges, physical impacts, and air emissions. However, although quantitative data on these consequences are generally scarce the shipping contribution to acidification by SOx- and NOx-emissions has been quantified to some extent. There are several knowledge gaps regarding the ecological consequences of, for example, the increasing amount of chemicals transported on water, the spread of non-indigenous species coupled with climate change, and physical impacts such as shipping noise and artificial light. The whole plethora of environmental consequences, as well as potential synergistic effects, should be seriously considered in transport planning.
