  • Bridging the gap between analysis and design : Improving existing driver interfaces with tools from the framwork of cognitive work analysis

    • Two studies of train driving are presented, both within the framework of cognitive work analysis. In the first study, the modelling tool abstraction–decomposition space is adapted to routine conditions, making the analysis more representative for normal procedures. A major contribution to these analyses was the use of the method for ‘collegial verbalisation’. One particular advantage with this method over other verbalisation methods is that it supplies the analysts with data that contain much more information, but not at the expense of being more subjective. On the contrary, this method produces think-aloud protocols from video-recordings that do not have to be interpreted by the researcher. From these analyses, it was possible to distinguish information that is an intrinsic part of the train driver task from information that is dependent on the configuration and design of the current support system. The analyses show that the driver works in three rather separate time intervals with a long-range, a short-term and an immediate sense of perspective. The driver switches between these while travelling between two stations. Based on these behaviour-shaping constraints, a prototype of a planning area of a driver interface was developed, making feed-forward planning possible for the driver. Four design iterations were completed, using a user-centred system design (UCSD) approach. Early tests show that the planning area of the interface supports the feed-forward decision strategy used by drivers who prefer an active driving style. However, the driver group also made substantial changes in the design, indicating that UCSD is an efficient tool in order to capture user competencies, and to bridge the gap between analysis and design.

  • Monitoring the osmotic response of single yeast cells through force measurement in the environmental scanning electron microscope

    • We present a measurement system that combines an environmental scanning electron microscope (ESEM) and an atomic force microscope (AFM). This combination enables studies of static and dynamic mechanical properties of hydrated specimens, such as individual living cells. The integrated AFM sensor provides direct and continuous force measurement based on piezoresistive force transduction, allowing the recording of events in the millisecond range. The in situ ESEM-AFM setup was used to study Pichia pastoris wild-type yeast cells. For the first time, a quantified measure of the osmotic response of an individual yeast cell inside an ESEM is presented. With this technique, cell size changes due to humidity variations can be monitored with nanometre accuracy. In addition, mechanical properties were extracted from load-displacement curves. A Young's modulus of 13-15 MPa was obtained for the P. pastoris yeast cells. The developed method is highly interesting as a complementary tool for the screening of drugs directed towards cellular water transport activity and provides new possibilities of studying mechanosensitive regulation of aquaporins.

  • Novel Method for Controlled Wetting of Materials in the Environmental Scanning Electron Microscope

    • Environmental scanning electron microscopy has been extensively used for studying the wetting properties of different materials. For some types of investigation, however, the traditional ways of conducting in situ dynamic wetting experiments do not offer sufficient control over the wetting process. Here, we present a novel method for controlled wetting of materials in the environmental scanning electron microscope (ESEM). It offers improved control of the point of interaction between the water and the specimen and renders it more accessible for imaging. It also enables the study of water transport through a material by direct imaging. The method is based on the use of a piezo-driven nanomanipulator to bring a specimen in contact with a water reservoir in the ESEM chamber. The water reservoir is established by local condensation on a Peltier-cooled surface. A fixture was designed to make the experimental setup compatible with the standard Peltier cooling stage of the microscope. The developed technique was successfully applied to individual cellulose fibers, and the absorption and transport of water by individual cellulose fibers were imaged.

    • Drug release from oral pharmaceutical formulations can be modified by applying a polymeric coating film with controlled mass transport properties. Interaction of the coating film with water may crucially influence its composition and permeability to both water and drug. Understanding this interaction between film microstructure, wetting, and mass transport is important for the development of new coatings. We present a novel method for controlled wetting of polymer coating films in an environmental scanning electron microscope, providing direct visual information about the processes occurring as the film goes from dry to wet. Free films made of phase-separated blends of water-insoluble ethyl cellulose (EC) and water-soluble hydroxypropyl cellulose (HPC) were used as a model system, and the blend ratio was varied to study the effect on the water transport properties. Local variations in water transport through the EC/HPC films were directly observed, enabling the immediate analysis of the structure-mass transport relationships. The leaching of HPC could be studied by evaporating water from the films in situ. Significant differences were observed between films of varying composition. The method provides a valuable complement to the current approach of making distinct diffusion and microscopy experiments for studying the dynamic interaction of polymer films with water.

    • This literature survey focuses on studies of the train driver system and its connections to other parts of the larger train traffic system. The main part of the studies concern Swedish conditions, but other references are included as well.In the first part, different ways of analysing accidents are discussed, including organisational aspects. In the second part, research results from different areas, all assumed to be relevant to the train driver task, are aggregated into a body of knowledge about the train driver system. Comparisons are made between the train driver task and other operator tasks. Further, the effects of a higher degree of automation are discussed.The task of driving a train is modelled as a dynamic decision task, where the driver's mental representation of the technical system and the optic flow of information are assumed to be important parts of the train driver task.In the last part of the literature survey, the content of 40 accident reports is discussed. Further, an analysis and a classification of Swedish train accidents are made by using CREAM (Hollnagel, 1998). The content of the reports, as well as the CREAM-method are evaluated, and different classification criteria are discussed.
