  • Therapeutic effects of artesunate on lupus-prone MRL/lpr mice are dependent on T follicular helper cell differentiation and activation of JAK2-STAT3 signaling pathway

    • BACKGROUND: Anti-malarial drug artesunate (ART), a semi-synthetic derivative of artemisnin, has immunosuppressive effects on several autoimmune diseases, including Systemic lupus erythematosus (SLE), Rheumatoid arthritis (RA), and Colitis. However, molecular mechanisms of ART, especially on follicular helper T cells (Tfh), central players in SLE pathology, are far from clear.PURPOSE: The object for this work is to investigate the therapeutic effect of ART on lupus-prone MRL/lpr mice and its regulatory function on Tfh cells.STUDY DESIGN AND METHODS: MRL/lpr mice were used to explore therapeutic effects of ART on lupus-prone MRL/lpr mice and its regulatory functions on Tfh cells. Then, experiments of renal function were accomplished using the biochemical kits. Effects of ART on histopathology of kidneys, inflammatory factors and autoantibodies were examined using H&E staining, ELISA and real-time PCR. Flow cytometry and western blot analysis were used to examine effects of ART on Tfh differentiation and Jak2-Stat3 signaling pathway.RESULTS: Upon oral administration, ART significantly prolonged the survival of MRL/lpr mice, ameliorated the lupus nephritis symptoms, decreased the levels of anti-dsDNA antibodies deposited in the kidney, and the levels of pathogenic cytokines (IL-6, IFN-γ and IL-21). After ART treatment, T-cell compartment in the spleen of MRL/lpr mice was restored in terms of reduction in the number of Tfh cells and in the maintenance of the ratio of Tfr to follicular regulatory T cells (Tfh). In addition, ART has significantly inhibited the phosphorylation levels of Jak2 and Stat3 in the MRL/lpr mice.CONCLUSION: ART showed therapeutic effects on lupus-prone MRL/lpr mice by inhibiting the differentiation of Tfh cells as well as altering the activation status of Jak2-Stat3 signaling cascade.   • A metabolite from commensal Candida albicans enhances the bactericidal activity of macrophages and protects against sepsis

    • The gut microbiome is recognized as a key modulator of sepsis development. However, the contribution of the gut mycobiome to sepsis development is still not fully understood. Here, we demonstrated that the level of Candida albicans was markedly decreased in patients with bacterial sepsis, and the supernatant of Candida albicans culture significantly decreased the bacterial load and improved sepsis symptoms in both cecum ligation and puncture (CLP)-challenged mice and Escherichia coli-challenged pigs. Integrative metabolomics and the genetic engineering of fungi revealed that Candida albicans-derived phenylpyruvate (PPA) enhanced the bactericidal activity of macrophages and reduced organ damage during sepsis. Mechanistically, PPA directly binds to sirtuin 2 (SIRT2) and increases reactive oxygen species (ROS) production for eventual bacterial clearance. Importantly, PPA enhanced the bacterial clearance capacity of macrophages in sepsis patients and was inversely correlated with the severity of sepsis in patients. Our findings highlight the crucial contribution of commensal fungi to bacterial disease modulation and expand our understanding of the host-mycobiome interaction during sepsis development. © 2023, The Author(s), under exclusive licence to CSI and USTC.

    • Re-identification (ReID) of occluded persons is a challenging task due to the loss of information in scenes with occlusions. Most existing methods for occluded ReID use 2D-based network structures to directly extract representations from 2D RGB (red, green, and blue) images, which can result in reduced performance in occluded scenes. However, since a person is a 3D non-grid object, learning semantic representations in a 2D space can limit the ability to accurately profile an occluded person. Therefore, it is crucial to explore alternative approaches that can effectively handle occlusions and leverage the full 3D nature of a person. To tackle these challenges, in this study, we employ a 3D view-based approach that fully utilizes the geometric information of 3D objects while leveraging advancements in 2D-based networks for feature extraction. Our study is the first to introduce a 3D view-based method in the areas of holistic and occluded ReID. To implement this approach, we propose a random rendering strategy that converts 2D RGB images into 3D multi-view images. We then use a 3D Multi-View Transformation Network for ReID (MV-ReID) to group and aggregate these images into a unified feature space. Compared to 2D RGB images, multi-view images can reconstruct occluded portions of a person in 3D space, enabling a more comprehensive understanding of occluded individuals. The experiments on benchmark datasets demonstrate that the proposed method achieves state-of-the-art results on occluded ReID tasks and exhibits competitive performance on holistic ReID tasks. These results also suggest that our approach has the potential to solve occlusion problems and contribute to the field of ReID.     • Next generation cellular network is expected to provide the simultaneous high-accuracy localization and ultra-reliable communication services, even in high mobility scenarios. To that end, the novel orthogonal time frequency space (OTFS) modulation has been developed as a promising physical-layer transmission technique, evident by the outstanding performance in terms of robustness against time-frequency selective fading over the orthogonal frequency division multiplexing (OFDM) counterpart. However, when OTFS meets massive multiple-input multiple-output (MIMO), the specific conditions, under which the delay-Doppler (DD) domain channel model holds, are not identified. In addition, the channel estimation and localization performance in such system is rarely studied. In this work, we target at these new challenges, and conduct comprehensive modelling, performance analysis, and algorithm design for massive MIMO-OTFS based simultaneous localization and communications. Specifically, we derive new channel models for the massive MIMO-OTFS system, which captures both time-frequency dispersion and spatial wideband effects. The specific conditions, under which the new models hold has been unveiled as well. Based on the new models, we establish the theoretical foundations for channel estimation and localization, by deriving the Cramer-Rao lower bounds of channel parameter and location estimation errors. Such bounds have been achieved with the newly designed low-complexity channel estimation and localization algorithms. Numerical simulations of the proposed framework with prevailing pulse functions are also conducted and the results validate the proposed designs and analysis. © 1983-2012 IEEE.

  • Smart AUV-Assisted Localization and Time Synchronization of Underwater Acoustic Devices

    • The localization and time synchronization of Underwater Acoustic Devices (UADs) have many potential applications in industry and scientific research. This can be done by deploying location-aware fixed anchors on the sea surface or bottom. However, this solution has poor accuracy and induces huge costs. In recent years, with the surge of Autonomous Underwater Vehicles (AUVs), the possibility of employing smart AUVs as mobile anchors have been extensively discussed. In this chapter, three smart AUV-assisted underwater localization and time synchronization techniques will be introduced, based on ToA (Time of Arrival), Doppler shift, and linear frequency modulated signals. The system model will be presented in detail, and the corresponding algorithms will be briefly discussed. For each of them, some preliminary numerical evaluations are conducted to directly show their performance.

    • 5G positioning is a very promising area that presents many opportunities and challenges. Many existing techniques rely on multiple anchor nodes and line-of-sight (LOS) paths, or single reference node and single-bounce non-LOS (NLOS) paths. However, in dense multipath environments, identifying the LOS or single-bounce assumptions is challenging. The multi-bounce paths will make the positioning accuracy deteriorate significantly. We propose a robust 5G positioning algorithm in NLOS multipath environments to mitigate the effects of multi-bounce paths. © 2023 IEEE.

    • Mobile edge computing provides the radio access networks with cloud computing capabilities to fulfill the requirements of the Internet of Things services such as high reliability and low latency. Offloading services to edge servers can alleviate the storage and computing limitations and prolong the lifetimes of the IoT devices. However, offloading in MEC faces scalability problems due to the massive number of IoT devices. In this article, we present a new integration architecture of the cloud, MEC, and IoT, and propose a lightweight request and admission framework to resolve the scalability problem. Without coordination among devices, the proposed framework can be operated at the IoT devices and computing servers separately, by encapsulating latency requirements in offloading requests. Then a selective offloading scheme is designed to minimize the energy consumption of devices, where the signaling overhead can be further reduced by enabling the devices to be self-nominated or self-denied for offloading. Simulation results show that our proposed selective offloading scheme can satisfy the latency requirements of different services and reduce the energy consumption of IoT devices. © 2018 IEEE

    • The future sixth generation (6G) networks will feature great importance on the integration of communications and localization, to realize the Internet of Everything (IoE). In this paper, we investigate the hybrid precoding design for the integrated communications and localization (ICAL) in the massive multiple-input multiple-output (MIMO) low Earth orbit (LEO) systems. In particular, we first derive an upper bound of the communication spectral efficiency (SE) and the squared position error bound (SPEB) of localization. Then, we formulate a multi-objective optimization problem to simultaneously operate communications and localization. Simulation results demonstrate the satisfactory performance of the proposed massive MIMO LEO ICAL system for typical setups. © 2023 IEEE.

    • Vehicle-to-Grid (V2G) is a promising paradigm to alleviate energy supply and demand imbalance of the grid. To further improve the power transmission efficiency, in this paper, we propose a cloudlet-based Vehicle-to-Vehicle (V2V) energy exchange framework. In the framework, the Energy Switch Center (ESC) serves as a trading broker, which purchases electricity from discharging vehicles and then resells it to the charging ones without energy transmission on the grid. The energy trading process is modeled in a contract theoretic approach. We derive the optimal feasible contracts which maximize the profit of the ESC. Furthermore, we systematically study the practical scenario where both the charging demand and renewable energy supplement are random variables, and propose a practical optimal contract-based electricity purchase scheme. Simulation results show that the proposed scheme can efficiently increase the profit of the ESC than the other mechanisms. © 2016 IEEE.
