    • This paper presents the potential for the Swedish Laqua system to be used as a sustainable method for on-site landfill leachate management in Northern Ireland, specifically the potential to use locally sourced filter materials from Northern Ireland as part of the filter system. Four carbon containing ashes and four types of peat were tested over a 24 hours period by a shaking test with untreated landfill leachate. Considering the results of this screening test, and the economical and sustainable supply of filter materials, one combination of ash and peat was selected to be column tested. Column testing with artificial leachate containing 7 organic pollutants (3 PAHs and 4 PCBs) and 9 inorganic pollutants showed that locally sourced filter materials effectively removed both organic and inorganic pollutants. A subsequent column test with landfill leachate for 13 weeks demonstrated it was feasible to apply the Laqua system with economical locally sourced filter materials.

  • Gene-Wide Analysis Detects Two New Susceptibility Genes for Alzheimer's Disease

    • Background: Alzheimer's disease is a common debilitating dementia with known heritability, for which 20 late onset susceptibility loci have been identified, but more remain to be discovered. This study sought to identify new susceptibility genes, using an alternative gene-wide analytical approach which tests for patterns of association within genes, in the powerful genome-wide association dataset of the International Genomics of Alzheimer's Project Consortium, comprising over 7 m genotypes from 25,580 Alzheimer's cases and 48,466 controls. Principal Findings: In addition to earlier reported genes, we detected genome-wide significant loci on chromosomes 8 (TP53INP1, p = 1.4x10(-6)) and 14 (IGHV1-67 p = 7.9x10(-8)) which indexed novel susceptibility loci. Significance: The additional genes identified in this study, have an array of functions previously implicated in Alzheimer's disease, including aspects of energy metabolism, protein degradation and the immune system and add further weight to these pathways as potential therapeutic targets in Alzheimer's disease.

    • It is estimated that 350 million individuals worldwide suffer from rare diseases, which are predominantly caused by mutation in a single gene(1). The current molecular diagnostic rate is estimated at 50%, with whole-exome sequencing (WES) among the most successful approaches(2-5). For patients in whom WES is uninformative, RNA sequencing (RNA-seq) has shown diagnostic utility in specific tissues and diseases(6-8). This includes muscle biopsies from patients with undiagnosed rare muscle disorders(6,9), and cultured fibroblasts from patients with mitochondrial disorders(7). However, for many individuals, biopsies are not performed for clinical care, and tissues are difficult to access. We sought to assess the utility of RNA-seq from blood as a diagnostic tool for rare diseases of different pathophysiologies. We generated whole-blood RNA-seq from 94 individuals with undiagnosed rare diseases spanning 16 diverse disease categories. We developed a robust approach to compare data from these individuals with large sets of RNA-seq data for controls (n = 1,594 unrelated controls and n = 49 family members) and demonstrated the impacts of expression, splicing, gene and variant filtering strategies on disease gene identification. Across our cohort, we observed that RNA-seq yields a 7.5% diagnostic rate, and an additional 16.7% with improved candidate gene resolution.

  • Convergent genetic and expression data implicate immunity in Alzheimer's disease

    • Background: Late-onset Alzheimer's disease (AD) is heritable with 20 genes showing genome-wide association in the International Genomics of Alzheimer's Project (IGAP). To identify the biology underlying the disease, we extended these genetic data in a pathway analysis. Methods: The ALIGATOR and GSEA algorithms were used in the IGAP data to identify associated functional pathways and correlated gene expression networks in human brain. Results: ALIGATOR identified an excess of curated biological pathways showing enrichment of association. Enriched areas of biology included the immune response (P = 3.27 X 10(-12) after multiple testing correction for pathways), regulation of endocytosis (P = 1.31 X 10(-11)), cholesterol transport (P = 2.96 X 10(-9)), and proteasome-ubiquitin activity (P = 1.34 X 10(-6)). Correlated gene expression analysis identified four significant network modules, all related to the immune response (corrected P = .002-.05). Conclusions: The immime response, regulation of endocytosis, cholesterol transport, and protein ubiquitination represent prime targets for AD therapeutics.

  • Wintering bird communities are tracking climate change faster than breeding communities

    • Global climate change is driving species' distributions towards the poles and mountain tops during both non-breeding and breeding seasons, leading to changes in the composition of natural communities. However, the degree of season differences in climate-driven community shifts has not been thoroughly investigated at large spatial scales. We compared the rates of change in the community composition during both winter (non-breeding season) and summer (breeding) and their relation to temperature changes. Based on continental-scale data from Europe and North America, we examined changes in bird community composition using the community temperature index (CTI) approach and compared the changes with observed regional temperature changes during 1980–2016. CTI increased faster in winter than in summer. This seasonal discrepancy is probably because individuals are less site-faithful in winter, and can more readily shift their wintering sites in response to weather in comparison to the breeding season. Regional long-term changes in community composition were positively associated with regional temperature changes during both seasons, but the pattern was only significant during summer due to high annual variability in winter communities. Annual changes in community composition were positively associated with the annual temperature changes during both seasons. Our results were broadly consistent across continents, suggesting some climate-driven restructuring in both European and North American avian communities. Because community composition has changed much faster during the winter than during the breeding season, it is important to increase our knowledge about climate-driven impacts during the less-studied non-breeding season.

  • Kinetics and kinematics of the horse comparing left and right rising trot

    • Reasons for performing study: At rising trot the rider sits alternately down on one diagonal pair of limbs and rises up on the other. The possible effects on asymmetry of locomotion induced by rising trot have rarely been studied.Objectives: To demonstrate whether, and if so to what extent, rising trot causes asymmetrical loading in the vertical ground reaction force (VGRF) and/or asymmetrical effects on the locomotion pattern, comparing left and right side.Methods: Seven elite horses were ridden in left and right rising trot on a treadmill, while VGRF and kinematics were measured, with the horses' neck raised, the poll high and the bridge of the nose slightly in front of the vertical.Results: Force loading was generally increased in the limbs of the sitting diagonal. The lumbar back was lower between mid-stances of the sitting and nonsitting stance, pelvic roll was limited and the tuber coxae heights were lower on the sitting side. Maximal hindlimb protraction was decreased. Forelimb retraction was increased and the T6 height decreased.Conclusion: The rider movement induces an uneven biphasic load that affects the back, pelvis and limb kinematics and VGRF.Potential relevance: The generally advocated technique of alternating limbs when riding in rising trot is supported. The VGRF changes between rising on the left or right diagonal were distinct, but minor in absolute terms and therefore unlikely to have direct impact on the occurrence of locomotor injuries. Knowledge of an increase of asymmetry in rising trot is potentially useful for riders/trainers.
