  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Background: Progress in cancer biomarker discovery is dependent on access to high-quality biological materials and high-resolution clinical data from the same cases. To overcome current limitations, a systematic prospective longitudinal sampling of multidisciplinary clinical data, blood and tissue from cancer patients was therefore initiated in 2010 by Uppsala and Umeå Universities and involving their corresponding University Hospitals, which are referral centers for one third of the Swedish population.Material and Methods: Patients with cancer of selected types who are treated at one of the participating hospitals are eligible for inclusion. The healthcare-integrated sampling scheme encompasses clinical data, questionnaires, blood, fresh frozen and formalin-fixed paraffin-embedded tissue specimens, diagnostic slides and radiology bioimaging data.Results: In this ongoing effort, 12,265 patients with brain tumors, breast cancers, colorectal cancers, gynecological cancers, hematological malignancies, lung cancers, neuroendocrine tumors or prostate cancers have been included until the end of 2016. From the 6914 patients included during the first five years, 98% were sampled for blood at diagnosis, 83% had paraffin-embedded and 58% had fresh frozen tissues collected. For Uppsala County, 55% of all cancer patients were included in the cohort.Conclusions: Close collaboration between participating hospitals and universities enabled prospective, longitudinal biobanking of blood and tissues and collection of multidisciplinary clinical data from cancer patients in the U-CAN cohort. Here, we summarize the first five years of operations, present U-CAN as a highly valuable cohort that will contribute to enhanced cancer research and describe the procedures to access samples and data.

  • Insufficient antibody validation challenges oestrogen receptor beta research

    • The discovery of oestrogen receptor beta (ER beta/ESR2) was a landmark discovery. Its reported expression and homology with breast cancer pharmacological target ER alpha (ESR1) raised hopes for improved endocrine therapies. After 20 years of intense research, this has not materialized. We here perform a rigorous validation of 13 anti-ER beta antibodies, using well-characterized controls and a panel of validation methods. We conclude that only one antibody, the rarely used monoclonal PPZ0506, specifically targets ER beta in immunohistochemistry. Applying this antibody for protein expression profiling in 44 normal and 21 malignant human tissues, we detect ER beta protein in testis, ovary, lymphoid cells, granulosa cell tumours, and a subset of malignant melanoma and thyroid cancers. We do not find evidence of expression in normal or cancerous human breast. This expression pattern aligns well with RNA-seq data, but contradicts a multitude of studies. Our study highlights how inadequately validated antibodies can lead an exciting field astray.

  • Photoresponsive and Polarization-Sensitive Structural Colors from Cellulose/Liquid Crystal Nanophotonic Structures

    • Cellulose nanocrystals (CNCs) possess the ability to form helical periodic structures that generate structural colors. Due to the helicity, such self-assembled cellulose structures preferentially reflect left-handed circularly polarized light of certain colors, while they remain transparent to right-handed circularly polarized light. This study shows that combination with a liquid crystal enables modulation of the optical response to obtain light reflection of both handedness but with reversed spectral profiles. As a result, the nanophotonic systems provide vibrant structural colors that are tunable via the incident light polarization. The results are attributed to the liquid crystal aligning on the CNC/glucose film, to form a birefringent layer that twists the incident light polarization before interaction with the chiral cellulose nanocomposite. Using a photoresponsive liquid crystal, this effect can further be turned off by exposure to UV light, which switches the nematic liquid crystal into a nonbirefringent isotropic phase. The study highlights the potential of hybrid cellulose systems to create self-assembled yet advanced photoresponsive and polarization-tunable nanophotonics.

  • Synthetic Plant Cuticle Coating as a Biomimetic Moisture Barrier Membrane for Structurally Colored Cellulose Films

    • Photonic films based on cellulose nanocrystals (CNCs) are sustainable candidates for sensors, structurally colored radiative cooling, and iridescent coatings. Such CNC-based films possess a helicoidal nanoarchitecture, which gives selective reflection with the polarization of the incident light. However, due to the hygroscopic nature of CNCs, the structural colored material changes and may be irreversibly damaged at high relative humidity. Thus, moisture protection is essential in such settings. In this work, hygroscopic CNC-based films are protected with a bioinspired synthetic plant cuticle; a strategy already adopted by real plants. The protective cuticle layers altered the reflected colors to some extent, but more importantly, they significantly reduced the water vapor permeance by more than two orders of magnitude, from 2.1 × 107 (pristine CNC/GLU film) to 12.3 × 104 g µm m−2 day−1 atm−1 (protected CNC/GLU film). This expands significantly the time window of operation for CNC/GLU films at high relative humidity.

  • Industrialization of Services : Technology and Routinization in the 21st Century

    • Sweden has a long tradition of process improvements, productivity increments, waste reduction in manufacturing, continuing a mode of industrialization that helped achieve prosperity. With growing urban centers, mechanization of agriculture, outsourcings by manufacturing and the public sector with subsequent acquisitions, large service firms have appeared in an ever-growing service sector. Some firms with a previously simple operation have become employers of hundreds of thousands of people in what can be described as a Nordic phenomenon in its origin when looking at the roots of the largest firms.With this development in mind, the aim of the thesis is to explore the factors that can be found in the case study of industrialization of services in Nordic based firms. Using an abductive approach and mixed methods, the theoretical focus has been on the organization, routines, and knowledge. The data used in this thesis originates from three embedded contexts ranging from power and telecom infrastructural services, facility management, and a payment service provider. Summarily, the research questions touch upon (1) tacit knowledge and its transfer; (2) technology adoption; and (3) the grander implications of technology and industrialization in services.The thesis has four main contributions. First, the first paper provides a unique empirical insight into firms that rarely see any academic attention, a domain dominated by craftmanship to some degree, and with that an environment saturated with tacit knowledge. Secondly, the results indicate that while tacit knowledge is a significant part of service firms, recent technological advancement can overcome difficulties of transferring certain somatic aspects it. Capturing tacit knowledge digitally and transforming it into rich data, turns it into a crystallized artifact that is separated from time and space. High bandwidth enables the transfer of these crystallized artifacts of rich data, to any place, at any time. Thirdly, looking at the adoption of automation technology in various firms, it becomes clear that firm leadership has the pivotal role when it comes to technology adoption in contrast to the user. Depending on firm characteristics the readiness varies. Furthermore, technology itself may provide more opportunities than it originally intended. However, there are further dimensions that come into play such as network effects. Finally, the thesis can summate that technology has an incredible importance in industrializing services, but also posits that it may change the concept of what services truly are.For practitioners it is important to appreciate that technology has an increasingly important role in Services. Service firms that are geographically dispersed, or plan to become so via growth, do not necessarily need to be constrained in terms of knowledge transfer. However, adoption of technology requires a deliberate strategy from an enabling and proactive senior leadership.

  • Lifetime of racetrack skyrmions

    • The skyrmion racetrack is a promising concept for future information technology. There, binary bits are carried by nanoscale spin swirls-skyrmions-driven along magnetic strips. Stability of the skyrmions is a critical issue for realising this technology. Here we demonstrate that the racetrack skyrmion lifetime can be calculated from first principles as a function of temperature, magnetic field and track width. Our method combines harmonic transition state theory extended to include Goldstone modes, with an atomistic spin Hamiltonian parametrized from density functional theory calculations. We demonstrate that two annihilation mechanisms contribute to the skyrmion stability: At low external magnetic field, escape through the track boundary prevails, but a crossover field exists, above which the collapse in the interior becomes dominant. Considering a Pd/Fe bilayer on an Ir(111) substrate as a well-established model system, the calculated skyrmion lifetime is found to be consistent with reported experimental measurements. Our simulations also show that the Arrhenius pre-exponential factor of escape depends only weakly on the external magnetic field, whereas the pre-exponential factor for collapse is strongly field dependent. Our results open the door for predictive simulations, free from empirical parameters, to aid the design of skyrmion-based information technology.

  • Surface Modification of Nanocellulose towards Composite Applications

    • Nanocelluloses have attracted great interest during recent decades owing to their renewability, abundancy and remarkable physical and mechanical properties. The aim of this work was to investigate new strategies for surface modification and functionalization of nanocelluloses and their subsequent incorporation in polymer-host matrices.Nanocomposites of cellulose nanofibrils (CNF) and polycaprolactone (PCL) were produced by employing CNF nanopaper (NP) as a template and surface-initiated ring-opening polymerization (SI-ROP) of ε-caprolactone (ε-CL). SI-ROP of ε-CL from filter paper (FP) was also carried out for comparison. A larger amount of PCL was grafted from NP than from FP. The grafted NP had stronger mechanical properties than neat PCL.Cellulose nanocrystal (CNC)-reinforced polyvinyl acetate (PVAc) nanocomposites were also investigated. CNC were modified via “SI-reversible addition-fragmentation chain transfer and macromolecular design via the interchange of xanthate” (SI-RAFT/MADIX) polymerization of vinyl acetate (VAc). The resulting nanocomposites exhibited improved mechanical performance than the unmodified CNC.It is generally agreed that covalent grafting is superior to physical adsorption for the modification of a reinforcing agent. However, this hypothesis has never been thoroughly investigated. CNC was modified either through covalent grafting or through physical adsorption of poly(butyl methacrylate) (PBMA). Both methods resulted in improved mechanical performance than that of pure PCL or PCL containing unmodified CNC. However, covalent grafting gave the best mechanical performance even at high relative humidity.Functionalized CNC (F-CNC) were obtained through a versatile methodology employing organic acids bearing a functional group were employed for the simultaneous acid hydrolysis and esterification of cellulose fibers. This provided a facile route for the preparation of F-CNC.

    • Applied studies with Large Eddy Simulation (LES) of hazardous gas dispersion around buildings in cities have become increasingly feasible due to rapid advancements in computing technology. However, there is little extant literature investigating how each model's results compare with others, as well as their ability to predict near-field dispersion in a real city. In this study, three typical LES sub-grid-scale models are used to simulate gas dispersion, utilizing alternatively constant values and synthetic turbulence at inflow boundaries. The results are compared with data from the Joint Urban 2003 Atmospheric Dispersion Study in Oklahoma City. Flow and turbulence statistics of the simulation is presented at two probe locations, one inside the city-core and one outside. In addition, comparisons with the measured mean concentration and maximum concentration values are conducted. It was found that in the core of the city, simulated turbulence is mainly determined by buildings and their configurations, and is only weakly affected by model type and assumed turbulence at the inflow boundaries. On the other hand, outside and upwind the city center the turbulence set at the inflow boundaries is very important if realistic turbulence statistics is to be achieved. Downstream of the source, all tested models produce similar predictions of maximum concentration values, which in turn are similar to the experimental data. Thus, the results indicate that it could be better to use the LES calculated maximum-concentration instead of the calculated mean-concentration when developing methods for hazard area estimation.

    • A multifunctional initiator for ATRP has been synthesized by reacting a hyperbranched polyether, based on 3-ethyl-3-(hydroxymethyl)oxetane, with 2-bromo-isobutyrylbromide. The macroinitiator contained approximately 25 initiating sites per molecule. It was used for the atom transfer radical polymerization of methyl acrylate mediated by Cu(I)Br and tris(2-(dimethylamino)ethyl)amine (Me-6-TREN) in ethyl acetate at room temperature. This yielded a co-polymer with a dendritic-linear architecture. The large number of growing chains from each macromolecule increases the probability of inter-and intramolecular reactions. In order to control these kinds of polymerizing systems and prevent them from forming a gel, the concentration of propagating radicals must be kept low. The polymerizations under these conditions were well controlled. When a ratio of initiating sites-to-catalyst of 1:0.05 was used, the polymers from all of the reactions had a low polydispersity, ranging from 1.1 to 1.4. None of the polymerizations under these conditions gave gelation. Monomer conversions as high as 65% were reached while maintaining control over the polymerization. (C) 2002 Elsevier Science Ltd.
