  • A Combined Measurement and Modelling Approach to Assess the Sustainability of Whole-Tree Harvesting—A Swedish Case Study

    • The demand of renewable energy has increased the interest in whole-tree harvesting. The sustainability of whole-tree harvesting after clear-cutting, from an acidification point of view, depends on two factors: the present acidification status and the further loss of buffering capacity at harvesting. The aims of this study were to investigate the relationship between these two factors at 26 sites along an acidification gradient in Sweden, to divide the sites into risk classes, and to examine the geographical distribution of them in order to provide policy-relevant insights. The present status was represented by the acid neutralizing capacity (ANC) in soil solution, and the loss of buffering capacity was represented by the estimated exceedance of critical biomass harvesting (CBH). The sites were divided into three risk classes combining ANC and exceedance of CBH. ANC and exceedance of CBH were negatively correlated, and most sites had either ANC < 0 and exceedance (high risk) or ANC > 0 and no exceedance (low risk). There was a geographical pattern, with the high risk class concentrated to southern Sweden, which was mainly explained by higher historical sulfur deposition and site productivity in the south. The risk classes can be used in the formulation of policies on whole-tree harvesting and wood ash recycling.

  • Dopamine D(1) receptors and age differences in brain activation during working memory

    • In an fMRI study, 20 younger and 20 healthy older adults were scanned while performing a spatial working-memory task under two levels of load. On a separate occasion, the same subjects underwent PET measurements using the radioligand [(11)C] SCH23390 to determine dopamine D(1) receptor binding potential (BP) in caudate nucleus and dorsolateral prefrontal cortex (DLPFC). The fMRI study revealed a significant load modulation of brain activity (higher load>lower load) in frontal and parietal regions for younger, but not older, adults. The PET measurements showed marked age-related reductions of D(1) BP in caudate and DLPFC. Statistical control of caudate and DLPFC D(1) binding eliminated the age-related reduction in load-dependent BOLD signal in left frontal cortex, and attenuated greatly the reduction in right frontal and left parietal cortex. These findings suggest that age-related alterations in dopaminergic neurotransmission may contribute to underrecruitment of task-relevant brain regions during working-memory performance in old age.

    • Dopamine (DA) availability in both striatal and extrastriatal brain regions has been implicated in cognitive performance. Given that different brain regions are neuroanatomically and functionally different, DA receptor binding in different brain regions may be selectively important to specific cognitive functions. Using PET and the radioligand SCH23390, we measured D1 receptor binding potential (BP(ND)) in dorsolateral prefrontal cortex (DLPFC), hippocampus (HC), as well as in sensorimotor (SMST), associative (AST), and limbic (LST) striatum in 20 healthy younger persons. Subjects completed tasks assessing executive functioning, episodic memory, speed, and general knowledge. Unlike previous reports, we found no linear or curvilinear relationships between D1 receptor binding in DLPFC and performance in any cognitive task. However, BP(ND) in HC was positively linked to executive performance as well as to speed and knowledge. With regard to the striatal subregions, D1 BP(ND) in SMST was more strongly related to speed compared to the other striatal subregions, whereas D1 BP(ND) in AST was more strongly linked to general knowledge. These findings provide support for the notion that D1 receptors in separate brain regions are differentially related to performance in tasks tapping various cognitive domains.

  • Dopamine D1 Receptor Associations within and between Dopaminergic Pathways in Younger and Elderly Adults : Links to Cognitive Performance

    • Age-related dopamine (DA) losses have been extensively demonstrated for the D2 receptor subtype. Comparatively little is known about adult age changes regarding D1 receptors. In this study, we demonstrate marked age-related D1 receptor losses in striatal, limbic, and cortical areas using positron emission tomography and the radioligand [11C]SCH23390 in humans. Interregional correlations of binding potential (BP) values were high for areas within DA pathways in younger and elderly adults alike. Furthermore, interregional correlations in D1 BP between DA pathways were uniformly high in younger adults, indicating that D1 receptor densities in striatal, limbic, and cortical areas are not regulated independently, despite dopaminergic innervation from different midbrain areas. For elderly adults, between-pathway correlations of D1 receptor densities were preserved only between mesolimbic and mesocortical areas, whereas striatal BPs were weakly related to those in limbic and neocortical regions. Importantly, weak between-pathway correlations in elderly adults were found only for the slower half of the sample when BP was estimated during a cognitive interference task. These results suggest that D1 receptor densities in different pathways are not regulated independently in younger adults, but segregate in older age, and that this segregation of D1 receptor systems may be related to age-related cognitive slowing.

  • Atrophin controls developmental signaling pathways via interactions with Trithorax-like

    • Mutations in human Atrophin1, a transcriptional corepressor, cause dentatorubral-pallidoluysian atrophy, a neurodegenerative disease. Drosophila Atrophin (Atro) mutants display many phenotypes, including neurodegeneration, segmentation, patterning and planar polarity defects. Despite Atros critical role in development and disease, relatively little is known about Atros binding partners and downstream targets. We present the first genomic analysis of Atro using ChIP-seq against endogenous Atro. ChIP-seq identified 1300 potential direct targets of Atro including engrailed, and components of the Dpp and Notch signaling pathways. We show that Atro regulates Dpp and Notch signaling in larval imaginal discs, at least partially via regulation of thickveins and fringe. In addition, bioinformatics analyses, sequential ChIP and coimmunoprecipitation experiments reveal that Atro interacts with the Drosophila GAGA Factor, Trithorax-like (Trl), and they bind to the same loci simultaneously. Phenotypic analyses of Trl and Atro clones suggest that Atro is required to modulate the transcription activation by Trl in larval imaginal discs. Taken together, these data indicate that Atro is a major Trl cofactor that functions to moderate developmental gene transcription.

  • Core-valence double photoionization of the CS2 molecule

    • Double photoionization spectra of the CS2 molecule have been recorded using the TOF-PEPECO technique in combination with synchrotron radiation at the photon energies h nu=220, 230, 240, 243, and 362.7 eV. The spectra were recorded in the S 2p and C 1s inner-shell ionization regions and reflect dicationic states formed out of one inner-shell vacancy and one vacancy in the valence region. MCSCF calculations were performed to model the energies of the dicationic states. The spectra associated with a S 2p vacancy are well structured and have been interpreted in some detail by comparison to conventional S 2p and valence photoelectron spectra. The lowest inner-shell-valence dicationic state is observed at the vertical double ionization energy 188.45 eV and is associated with a (2p(3/2))(-1)(2 pi(g))(-1) double vacancy. The spectrum connected to the C 1s vacancy shows a distinct line at 310.8 eV, accompanied by additional broad features at higher double ionization energies. This line is associated with a (C 1s)(-1)(2 pi(g))(-1) double vacancy. (C-) 2010 American Institute of Physics. [doi: 10.1063/1.3469812]

  • Formation of Kr3+ via core-valence doubly ionized intermediate states

    • The time-of-flight photoelectron-photoion coincidence technique has been used to study single-photon 3d(9)4p(5) core-valence double ionization of Kr and subsequent Auger decay to triply charged states associated with the 4s(2)4p(3) and 4s(1)4p(4) configurations. The photon energy used was h nu = 150 eV. Multiconfiguration Dirac-Fock calculations were performed both for the doubly ionized intermediate states and the triply ionized final states. The intermediate states of Kr2+ are observed between 120 and 125 eV, whereas the final states of Kr3+ are observed between 74- and 120-eV ionization energy. Assignments of all structures are made based on the present numerical results. The calculated Auger rates give a detailed explanation of the relative line strengths observed.

    • Multielectron coincidence data for triple ionization of krypton have been recorded above the 3d ionization threshold at two photon energies (140 and 150 eV). Three principal transition pathways have been observed, two involving double Auger transitions from Kr+, and one involving single Auger transitions from Kr2+ created by direct single-photon double ionization. The decay of the 3d(9) D-2(5/2,3/2) states in Kr+ has been analyzed in some detail and is found to be strongly dominated by cascade processes where two electrons with well-defined energies are emitted. The decay paths leading to the 4s(2)4p(3) S-4, D-2, and P-2 states of Kr3+ are analyzed and energies of seven intermediate states in Kr2+ are given. A preliminary investigation of the decay paths from Kr+ 3d (9)4p(5)nl shake-up states has also been carried out.

  • Widespread episodic thiamine deficiency in Northern Hemisphere wildlife

    • Many wildlife populations are declining at rates higher than can be explained by known threats to biodiversity. Recently, thiamine (vitamin B-1) deficiency has emerged as a possible contributing cause. Here, thiamine status was systematically investigated in three animal classes: bivalves, ray-finned fishes, and birds. Thiamine diphosphate is required as a cofactor in at least five life-sustaining enzymes that are required for basic cellular metabolism. Analysis of different phosphorylated forms of thiamine, as well as of activities and amount of holoenzyme and apoenzyme forms of thiaminedependent enzymes, revealed episodically occurring thiamine deficiency in all three animal classes. These biochemical effects were also linked to secondary effects on growth, condition, liver size, blood chemistry and composition, histopathology, swimming behaviour and endurance, parasite infestation, and reproduction. It is unlikely that the thiamine deficiency is caused by impaired phosphorylation within the cells. Rather, the results point towards insufficient amounts of thiamine in the food. By investigating a large geographic area, by extending the focus from lethal to sublethal thiamine deficiency, and by linking biochemical alterations to secondary effects, we demonstrate that the problem of thiamine deficiency is considerably more widespread and severe than previously reported.

  • Chloride retention in forest soil by microbial uptake and by natural chlorination of organic matter

    • Inorganic chlorine (i.e. chloride; Cl-in) is generally considered inert in soil and is often used as a tracer of soil and ground water movements. However, recent studies indicate that substantial retention or release of Cl-in can occur in soil, but the rates and processes responsible under different environmental conditions are largely unknown. We performed Cl-36 tracer experiments which indicated that short-term microbial uptake and release of Cl-in, in combination with more long-term natural formation of chlorinated organic matter (Cl-org), caused Cl-in imbalances in coniferous forest soil. Extensive microbial uptake and release of Cl-in occurred over short time scales, and were probably associated with changes in environmental conditions. Up to 24% of the initially available Clin within pore water was retained by microbial uptake within a week in our experiments, but most of this Cl-in, was released to the pore water again within a month, probably associated with decreasing microbial populations. The natural formation of Clorg resulted in a net immobilization of 4% of the initial pore water Clin over four months. If this rate is representative for the area where soil was collected, Clorg formation would correspond to a conversion of 25% of the yearly wet deposition of Cl-in. The study illustrates the potential of two Clin retaining processes in addition to those previously addressed elsewhere (e.g. uptake of chloride by vegetation). Hence, several processes operating at different time scales and with different regulation mechanisms can cause Clin imbalances in soil. Altogether, the results of the present study (1) provide evidence that Cl-in cannot be assumed to be inert in soil, (2) show that microbial exchange can regulate pore water Cl-in, concentrations and (3) confirm the controversial idea of substantial natural chlorination of soil organic matter.
