  • Designing an Autonomous Integrated Downstream Sequence From a Batch Separation Process − An Industrial Case Study

    • This work is a proof of concept of how a sequence of industrial batch separation steps together are used to form an integrated autonomous downstream process. The sequence in this case study consisted of an anion chromatography step, virus inactivation and finally a hydrophobic chromatography step. Moving from batch to integrated separation minimizes hold‐up times, storage tanks, and required equipment. The conversion from batch to integrated mode is achieved by extracting operating points and separation data from batch chromatograms. The integrated separation process is realized on an ÄKTA Pure controlled by an open research software called Orbit, making it possible to operate complex process configurations including multiple steps. The results from this case study is the principle and method of the steps taken to automation, achieving a more continuous and efficient downstream process.

  • Grundskoleelevers design i lärande : En studie om lärprocesser i programmering

    • The aim of this study is to contribute to the knowledge about how pupils design their learning in programming. It is mainly the learning process, how pupils deal with problems in programming and how they become, are and remain engaged in the tasks, which is of interest. Programming can be understood in many ways; coding, a digital competence, creativity, or ways to solve problems. The digitalisation of society has also evoked a need to learn programming from an early age in compulsory school. In this study, programming is seen as a part of the digital competence that all pupils should have the opportunity to develop, which is a common thread that runs throughout compulsory school.  The study was conducted during programming lessons at three science centres in Sweden. The centres had previous experience in teaching programming. This was used at the time of the study by schools that in this way could offer pupils to try programming even before it became part of the compulsory teaching. The lessons were adapted for novices in programming and were conducted as part of the regular school day for pupils in grades 1-8.  The theoretical framework is based on design-oriented theory with a focus on how settings and design for learning includes both opportunities and dilemmas for learning. It provides a basis for the analysis of pupils’ approaches when learning programming as well as how different types of engagement relates to their design in learning. The results are presented in two articles, which contribute with different aspects of learning. Together the articles provide a picture of pupils’ learning design within programming in compulsory school.   The first article highlights knowledge of five different approaches that pupils used to solve assignments using programming. The qualitatively different ways that pupils used during the observed lessons were mathematically, trial and error, step-by-step, routine as well as aesthetically.  Each of these approaches allows pupils to use and practice different abilities that are important for programming. The abilities are compared to, but not equal to computational thinking (CT), which (in its turn) is linked to competencies that are important for an active participation in a digital society.   The second article contributes to the understanding of how behavioural, emotional, and cognitive engagement can be identified when pupils are learning programming. To understand how the different types of engagement are individually important, yet intertwined and influencing each other, is keen knowledge. The results show how different types of engagement become visible during programming lessons. Furthermore, it is discussed how the pupils’ identified engagement can be related to how their learning process is designed.  In the study, taken as a whole, the results of the two articles show how pupils become designers in their programming learning process. The pupils designed their learning throughout their learning process regarding to the settings, to the approach they used and in the way they became engaged.

  • Seasonal Variations of Redox State in Hemiboreal Soils Indicated by Changes of δ56Fe, Sulfate, and Nitrate in Headwater Streams

    • During recent decades, much focus has been put on the iron (Fe) isotope ratios in soils, rivers, and oceans, while studies on the variation in headwater streams are scarce. Here we assess seasonal water chemical data from 104 hemiboreal headwater streams. Between summer and late autumn, decreasing Fe concentrations and simultaneously increasing sulfate and nitrate concentrations suggest a shift from reduced to oxidized conditions in the soils along the main groundwater flow paths. Fe isotope data, obtained from a subpopulation of 16 streams, show low δ56Fe ratios during summer drought, indicating an important influx of reduced groundwater to the streams with primarily Fe(II) as an important Fe source. In total, the δ56Fe data ranged between −0.8 ± 0.1 and 1.8 ± 0.1‰ with the lowest values in summer and maximum δ56Fe ratios in late autumn or spring, indicating an influx of more oxidized, less Fe(II) rich groundwater during those seasons. Local differences in δ56Fe ratios between the headwater streams, seemed to be driven by the different soil redox status of the catchments. The streams with the lowest δ56Fe ratios during summer are characterized by a small share (4.4 ± 6.6%) of wetlands, indicating discharge of reduced groundwater from mainly anoxic, moist, organic-rich mineral soils during drought. Relatively high total organic carbon (TOC) concentrations (2.4 ± 1.1 mM) and low pH (5.2 ± 0.8) may have restricted efficient Fe(II) oxidation in streamwater especially during the late autumn survey. Our results from hemiboreal headwater streams reveal the importance of climatic, pedogenic, and land cover-derived controls on the provenance of stream Fe loads that is likely broadly applicable to similar streams elsewhere.

  • Interprofessional education in a student-led emergency department : A realist evaluation

    • This article reports a realist evaluation undertaken to identify factors that facilitated or hindered the successful implementation of interprofessional clinical training for undergraduate students in an emergency department. A realist evaluation provides a framework for understanding how the context and underlying mechanisms affect the outcome patterns of an intervention. The researchers gathered both qualitative and quantitative data from internal documents, semi-structured interviews, observations, and questionnaires to study what worked, for whom, and under what circumstances in this specific interprofessional setting. The study participants were medical, nursing, and physiotherapy students, their supervisors, and two members of the emergency department's management staff. The data analysis indicated that the emergency ward provided an excellent environment for interprofessional education (IPE), as attested by the students, supervisors, and the clinical managers. An essential prerequisite is that the students have obtained adequate skills to work independently. Exemplary conditions for IPE to work well in an emergency department demand the continuity of effective and encouraging supervision throughout the training period and supervisors who are knowledgeable about developing a team.

    • Generation P? is based on a unique and comprehensive study giving voice to the words and opinions of the young people themselves. Never before have pornography and sexualized material been so readily available and pervasive in young people's everyday life. TV programmes, advertising, and the music industry exploit and play with pornographic codes and scenarios. The sex industry launches and promotes its products via youth channels and websites. How do young people navigate through this pornographic landscape? Does the omnipresence of pornography breed curiosity or resistance? How does pornography challenge the role of parents and teachers? Generation P? provides answers to these questions and presents a unique body of new research on youth, gender and pornography. The study shows that the vast majority of young people in the Nordic countries have seen porn. But young people do not swallow the pornographic messages without resistance, and some are very critical of or actively opposed to them. It is very seldom that the voices of young people are heard in the public debate on pornography. In Generation P? focus is set on the voices, definitions and experiences with pornography of young people themselves. The book contains contributions from leading researchers from different academic fields: sociology, psychology, media research, social work and public health.

  • Integration of a complete downstream process for the automated lab-scale production of a recombinant protein

    • In this work, an automated downstream process for the purification and formulation of a recombinant protein was integrated at lab scale in a single chromatography unit. The purification chain consists of three bind-and-elute chromatography columns, a flow-through membrane chromatography step, and a final ultrafiltration-diafiltration (UFDF) step to concentrate and formulate the sample. An integrated downstream process increases productivity and decreases process time and hold-up volume. In addition, the automation of the process allows reducing the manual work and increases reproducibility. To integrate the downstream steps, all the intermediate tanks are removed, and the eluate of a column is loaded directly onto the next one. This makes it necessary to design the process in order to minimize the column volumes and the process time. A research software called Orbit was used to automate the purification process and implement a UFDF step in the chromatography unit. The whole downstream sequence was successfully implemented at lab scale, getting a pure concentrated and formulated product with a productivity of 1.09 mg mL−1 h−1, achieving a time reduction from almost two to one working day, while getting a similar yield and purity. Regarding the UFDF operation, the sample was concentrated 10 times, and 97% of the old buffer was exchanged by the formulation buffer with a sequential diafiltration.

  • Abdominal adhesions in gynaecologic surgery after caesarean section : a longitudinal population-based register study.

    • OBJECTIVE: The aim of the study was to evaluate the association between abdominal adhesions at the time of gynaecologic surgery and a history of caesarean delivery, and to investigate obstetric factors contributing to adhesion formation after caesarean section (CS).DESIGN: Longitudinal population-based register study.SETTING: Sweden.POPULATION: Women undergoing benign hysterectomy and/or adnexal surgery in Sweden, 2000-2014, with a previous delivery during 1973-2013 (n = 15 479).METHODS: Information about abdominal adhesions during gynaecological surgery, prior medical history, pregnancies and deliveries were retrieved from Swedish National Health and Quality registers.MAIN OUTCOME MEASURES: Adhesions.RESULTS: In women with previous CS, adhesions were present in 37%, compared with 10% of women with no previous CS [odds ratio (OR): 5.18, 95% confidence interval (CI): 4.70-5.71]. Adhesions increased with the number of caesarean sections: 32% after one CS; 42% after two CS and 59% after three or more CS (P < 0.001). Regardless of the number of CS, factors at CS such as age ≥35 years (aOR: 1.28, 95% CI: 1.05-1.55), body mass index (BMI) ≥30 [adjusted OR (aOR): 1.91, 95% CI: 1.49-2.45] and postpartum infection (aOR: 1.55, 95% CI: 1.05-2.30) increased the risk of adhesions.CONCLUSIONS: Presence of adhesions in abdominal gynaecological surgery is associated with women's personal history of caesarean delivery. The number of caesarean sections was the important predictor of adhesions; advanced age, obesity and postpartum infection further increased the incidence.TWEETABLE ABSTRACT: Repeat caesarean, age, obesity and infection increased the risk of pelvic adhesions after caesarean section.
