  • Final report of the CCQM-K145 : Toxic and essential elements in bovine liver

    • Liver plays a major role in metabolism and acts as a source of energy for the body by storing glycogen. With the growing interest and investigation in the biological effects in recent years, it is important and necessary to develop accurate and comparable analytical methods for elements in bio-samples. It has, however, been 10 years since the tissue sample (bovine liver) of CCQM-K49 key comparison. The purpose of CCQM-K145 is to ensure the comparable and traceable measurement results for essential and toxic elements such as P, S, Zn, Mn, Ni, Mo, Sr, Cr, Co, Pb, As and Hg in bovine liver among NMIs and other designated measurement bodies worldwide. The comparison was agreed by IAWG as 6th IAWG Benchmarking Exercise with Zn and Ni as exemplary elements at the meeting in Korea in the early October 2016. The results of CCQM-K145 are expected to cover the measurement capability and support CMCs claiming for inorganic elements in the similar biological tissue materials and food samples. 30 NMIs and DIs registered in CCQM-K145. With respect to the methodology, a variety of techniques such as IDMS, ICP-OES, ICP-MS(non-ID), AAS and NAA were adopted by the participants. For Zn, Ni, Sr, Pb and Hg measurements, most participants chose ID-ICP-MS method, which showed the better performance in terms of consistency and reliability of the measurement results. In aspect of the traceability for the measurement results in CCQM-K145, most participants used their own (in house) CRMs or other NMI's CRMs to guarantee trace to SI unit. Most participants used similar matrix CRMs for quality control or method validation. Base on different statistic way to calculate the reference mass fraction values and associated uncertainties for each measurand, removal of the suspected extreme values, and discussion at the IAWG meetings, the median values are proposed as the KCRV for Zn, Ni, Mn, Mo, Cr, Pb and Hg; the arithmetic mean values are proposed as the KCRV for P, S, Sr, Co and As. In general, the performances of the majority of CCQM-K145 participants are very good, illustrating their measurement capabilities for Zn, Ni, P, S, Mn, Mo, Sr, Cr, As, Co, Pb and Hg in a complex biological tissue matrix. Bovine liver contains many kinds of nutrients and microelements, it can be regarded as a typical representative material of biological tissue and food. In CCQM-K145, the analytes involved alkali metals and transition elements, metalloids/semi-metals and non metals with a range of mass fraction from mg/g to μg/kg. CCQM-K145 also tested the ability of NMIs/DIs to determine elements that were easy to be lost and polluted, and interfered significantly. The chemical pretreatment methods of samples used in the comparison is suitable for general food and biological matrix samples. A variety of measurement methods used in the comparison represent the main instrumental technology for elemental analysis. Therefore, for supporting CMC claim, CCQM-K145 is readily applicable to measurement of more elements in a wide range of biological materials (including liquids and solids) and meat products.     • In this study, a comparative study on motion response and added resistance of a Supramax bulk carrier (K-Supramax Original) in regular waves was carried out to evaluate the reliabilities and accuracies of experimental and numerical simulation techniques. Two kinds of experiments were performed; one is the towing tank model testing for head wave conditions conducted at Seoul National University, and the other is the free-running model test for head and oblique wave conditions conducted at SSPA Sweden AB. Also, nine numerical computation results submitted by seven institutions were compared with the experimental data. The computation results were obtained by various seakeeping analysis methods such as the 2D strip theory, 3D Rankine panel method, and Computational Fluid Dynamics (CFD) based analysis. Based on the comparison, the characteristics of each numerical technique and resultant accuracies of seakeeping analyses were investigated. It was also confirmed that different results were obtained although the same program was used because of the user dependencies; setting for computation parameters, numerical schemes, and mesh generations, etc. Furthermore, the sensitivities of seakeeping quantities with respect to wave amplitudes were examined by conducting both model tests and nonlinear numerical simulations for different wave slopes. Lastly, the tendencies of ship motion and added resistance depending on the heading angle were identified, and the reliabilities of experiments and numerical computations for oblique waves were discussed.

    • This paper presents a comparative study on the motion responses and added resistance of a container ship. Eight institutions participated in the comparative study, and ten numerical results were compared with two experimental results. Two experimental results were obtained from Seoul National University towing tank and Sweden SSPA seakeeping basin. The results of two experimental institutions in head sea condition were compared and showed good agreement with each other. The difference in motion responses and added resistance according to the numerical analysis method were compared. Even though the same program was used, it was observed that different results were obtained depending on the users. The comparison of the motion response and the added resistance according to the change of wave slope showed that the added resistance greatly changed according to the wave slope. This tendency was the same in experimental results and CFD analysis results. From the comparative study, the influence of the experiment method on the added resistance, and the characteristics of numerical each code were identified.

    • In this paper, the added resistance of a large tanker is estimated experimentally and numerically in oblique sea. Experiments on ship motion response and added resistance in oblique sea are performed in the SSPA seakeeping basin. The experiments are conducted using the self-propulsion test for seven wave directions between 180° and 0°. In the self-propulsion test, the added resistance is estimated from the difference between the thrust of the propeller in calm water and waves. In the case of the head sea, the results are compared with those obtained from the captive test at the towing test of Seoul National University. As numerical method, two methods are selected: the strip method and the 3D Rankine panel method. The maximum value of the added resistance is observed between the incident wave directions of 180° and 150°. From 120°, the added resistance tends to decrease and the peak of the added resistance shifted to the short waves. Through the two numerical analysis methods, the tendency of added resistance and the cause of the change of the added resistance in the oblique sea are investigated.

  • Validation and development of MTH1 inhibitors for treatment of cancer

    • Background: Previously, we showed cancer cells rely on the MTH1 protein to prevent incorporation of otherwise deadly oxidised nucleotides into DNA and we developed MTH1 inhibitors which selectively kill cancer cells. Recently, several new and potent inhibitors of MTH1 were demonstrated to be non-toxic to cancer cells, challenging the utility of MTH1 inhibition as a target for cancer treatment. Material and methods: Human cancer cell lines were exposed in vitro to MTH1 inhibitors or depleted of MTH1 by siRNA or shRNA. 8-oxodG was measured by immunostaining and modified comet assay. Thermal Proteome profiling, proteomics, cellular thermal shift assays, kinase and CEREP panel were used for target engagement, mode of action and selectivity investigations of MTH1 inhibitors. Effect of MTH1 inhibition on tumour growth was explored in BRAF V600E-mutated malignant melanoma patient derived xenograft and human colon cancer SW480 and HCT116 xenograft models. Results: Here, we demonstrate that recently described MTH1 inhibitors, which fail to kill cancer cells, also fail to introduce the toxic oxidized nucleotides into DNA. We also describe a new MTH1 inhibitor TH1579, (Karonudib), an analogue of TH588, which is a potent, selective MTH1 inhibitor with good oral availability and demonstrates excellent pharmacokinetic and anti-cancer properties in vivo. Conclusion: We demonstrate that in order to kill cancer cells MTH1 inhibitors must also introduce oxidized nucleotides into DNA. Furthermore, we describe TH1579 as a best-in-class MTH1 inhibitor, which we expect to be useful in order to further validate the MTH1 inhibitor concept.

    • Assessment of skin irritation is an essential component of the safety evaluation of medical devices. OECD Test Guideline 439 describes the use of reconstructed human epidermis (RhE) as an in vitro test system for classification of skin irritation by neat chemicals. An international round robin study was conducted to evaluate the RhE method for determination of skin irritant potential of medical device extracts. Four irritant polymers and three non-irritant controls were obtained or developed that had demonstrated their suitability to act as positive or negative test samples. The RhE tissues (EpiDerm™ and SkinEthic™ RHE) were dosed with 100 μL aliquots of either saline or sesame oil extract. Incubation times were 18 h (EpiDerm™) and 24 h (SkinEthic™ RHE). Cell viability reduction > 50% was indicative of skin irritation. Both the EpiDerm™ and SkinEthic™ RHE tissues were able to correctly identify virtually all of the irritant polymer samples either in the saline, sesame oil or both solvent extracts. Our results indicate that RhE tissue models can detect the presence of strong skin irritants at low levels in dilute medical device polymer extracts. Therefore, these models may be suitable replacements for the rabbit skin irritation test to support the biological evaluation of medical devices.

    • High-temperature operating performance of p-i-p quantum dots-in-awell infrared photodetectors (QDIPs) is successfully demonstrated. The optically active region consists of 10 layers of p-doped selfassembled InAs quantum dots (QDs) asymmetrically positioned in In0.15Ga0.85As quantum wells (QWs). The dark current is suppressed by an incorporated superlattice (SL) structure composed of 10 pairs of AlGaAs/GaAs heterostructure. The very low recorded dark current makes the fabricated p-i-p QDIPs suitable for high-temperature operation. The measured photoresponse reveals broad mid-wave infrared (MWIR) detection up to 200 K.

  • Function selection among popular dive computer models : A review and proposed improvements

    • For optimal safety a dive computer should be easy to use and the displayed information easy to understand. The present study examines the usability of dive computers and potential technologies to enhance safety. It should be noted that even if the ease of use of a dive computer is increased to an extent where it is intuitive to use, this does not release the diver from the recommendation to read the dive computer manual to safely dive with it. For the present work, 47 dive computer models by 14 manufacturers were purchased and the manuals of another three were studied. Function selection was noted for each model. Where selection required a combination of long and short pushes, or more than one button, it was considered necessary to read the instruction manual merely to modify settings in the dive computer. The mean number of buttons, switches or contacts per dive computer was 3.3 (SD 1.1, range 1–7). Twelve models (24%) did not have multiple functions per button, one model (2%) had a single multi-function and 36 models (72%) had multiple multi-functions per button. Accessing these functions required short or long push combinations. In 41 out of 50 (82%) of the dive computer models, the user interface was not intuitive. The majority of popular dive computers employ combinations of long and short pushes to access multiple functions, requiring training and mnemonic effort to operate the device. They are not intuitive, and scope exists to improve the usability and safety of dive computers. Possibilities are described including touch screens, a wheel to replace traditional buttons and near field communications (NFC).
