  • Two genome-wide association studies of aggressive prostate cancer implicate putative prostate tumor suppressor gene DAB2IP

    • Background: The consistent finding of a genetic susceptibility to prostate cancer suggests that there are germline sequence variants predisposing individuals to this disease. These variants could be useful in screening and treatment. Methods: We performed an exploratory genome-wide association scan in 498 men with aggressive prostate cancer and 494 control subjects selected from a population-based case-control study in Sweden. We combined the results of this scan with those for aggressive prostate cancer from the publicly available Cancer Genetic Markers of Susceptibility (CGEMS) Study. Single-nucleotide polymorphisms (SNPs) that showed statistically significant associations with the risk of aggressive prostate cancer based on two-sided allele tests were tested for their association with aggressive prostate cancer in two independent study populations composed of individuals of European or African American descent using one-sided tests and the genetic model (dominant or additive) associated with the lowest value in the exploratory study. Results: Among the approximately 60000 SNPs that were common to our study and CGEMS, we identified seven that had a similar (positive or negative) and statistically significant (P<.01) association with the risk of aggressive prostate cancer in both studies. Analysis of the distribution of these SNPs among 1032 prostate cancer patients and 571 control subjects of European descent indicated that one, rs1571801, located in the DAB2IP gene, which encodes a novel Ras GTPase-activating protein and putative prostate tumor suppressor, was associated with aggressive prostate cancer (one-sided P value =. 004). The association was also statistically significant in an African American study population that included 210 prostate cancer patients and 346 control subjects (one-sided P value =. 02). Conclusion: A genetic variant in DAB2IP may be associated with the risk of aggressive prostate cancer and should be evaluated further.

  • Anger and disgust shape judgments of social sanctions across cultures, especially in high individual autonomy societies

    • When someone violates a social norm, others may think that some sanction would be appropriate. We examine how the experience of emotions like anger and disgust relate to the judged appropriateness of sanctions, in a pre-registered analysis of data from a large-scale study in 56 societies. Across the world, we find that individuals who experience anger and disgust over a norm violation are more likely to endorse confrontation, ostracism and, to a smaller extent, gossip. Moreover, we find that the experience of anger is consistently the strongest predictor of judgments of confrontation, compared to other emotions. Although the link between state-based emotions and judgments may seem universal, its strength varies across countries. Aligned with theoretical predictions, this link is stronger in societies, and among individuals, that place higher value on individual autonomy. Thus, autonomy values may increase the role that emotions play in guiding judgments of social sanctions.

    • Chronic or recurrent inflammation has been suggested as a causal factor in several human malignancies, including prostate cancer. Genetic predisposition is also a strong risk factor in the development of prostate cancer. In particular, Toll-like receptors (TLR), especially the TLR6-1-10 gene cluster, are involved in prostate cancer development. Interleukin-1 receptor-associated kinases (IRAK) 1 and 4 are critical components in the TLR signaling pathway. In this large case-control study, we tested two hypotheses: (a) sequence variants in IRAK1 and IRAK4 are associated with prostate cancer risk and (b) sequence variants in IRAK1/4 and TLR1-6-10 interacts and confers a stronger risk to prostate cancer. We analyzed 11 single nucleotide polymorphisms (four in IRAK1 and seven in IRAK4) among 1,383 newly diagnosed prostate cancer patients and 780 population controls in Sweden. Although the single-nucleotide polymorphisms in IRAK1 and IRAK4 alone were not significantly associated with prostate cancer risk, one single-nucleotide polymorphism in IRAK4, when combined with the high-risk genotype at TLR6-1-10, conferred a significant excess risk of prostate cancer. In particular, men with the risk genotype at TLR6-1-10 and IRAK4-7987 CG/CC had an odds ratio of 9.68 (P = 0.03) when compared with men who had wildtype genotypes. Our findings suggest synergistic effects between sequence variants in IRAK4 and the TLR 6-1-10 gene cluster. Although this study was based on a priori hypothesis and was designed to address many common issues facing this type of study, our results need confirmation in even larger studies.

    • Throughout its history, Operational Research has evolved to include methods, models and algorithms that have been applied to a wide range of contexts. This encyclopedic article consists of two main sections: methods and applications. The first summarises the up-to-date knowledge and provides an overview of the state-of-the-art methods and key developments in the various subdomains of the field. The second offers a wide-ranging list of areas where Operational Research has been applied. The article is meant to be read in a nonlinear fashion and used as a point of reference by a diverse pool of readers: academics, researchers, students, and practitioners. The entries within the methods and applications sections are presented in alphabetical order. The authors dedicate this paper to the 2023 Turkey/Syria earthquake victims. We sincerely hope that advances in OR will play a role towards minimising the pain and suffering caused by this and future catastrophes.

    • Background: Chronic inflammation plays an important role in several human cancers and may be involved in the etiology of prostate cancer. Toll-like receptors (TLRs) are important in the innate immune response to pathogens and in cross-talk between innate immunity and adaptive immunity. Our previous finding of an association of TLR4 gene sequence variants and prostate cancer risk provides evidence for a role of TLRs in prostate cancer. In this study, we investigated whether sequence variants in the TLR6-TLR1-TLR10 gene cluster, residing within a 54-kb region on 4p14, were associated with prostate cancer risk. Methods: We selected 32 single-nucleotide polymorphisms (SNPs) covering these three genes and genotyped these SNPs in 96 control subjects from the Cancer Prostate in Sweden (CAPS) population-based prostate cancer case-control study. Five distinct haplotype blocks were inferred at this region, and we identified 17 haplotype-tagging SNPs (htSNPs) that could uniquely describe < 95% of the haplotypes. These 17 htSNPs were then genotyped in the entire CAPS study population (1383 case subjects and 780 control subjects). Odds ratios of prostate cancer for the carriers of a variant allele versus those with the wild-type allele were estimated using unconditional logistic regression. Results: The allele frequencies of 11 of the 17 SNPs were statistically significantly different between case and control subjects (P = .04-.001), with odds ratios for variant allele carriers (homozygous or heterozygous) compared with wild-type allele carriers ranging from 1.20 (95% confidence interval [CI] = 1.00 to 1.43) to 1.38 (95% CI = 1.12 to 1.70). Phylogenetic tree analyses of common haplotypes identified a clade of two evolutionarily related haplotypes that are statistically significantly associated with prostate cancer risk. These two haplotypes contain all the risk alleles of these 11 associated SNPs. Conclusion: The observed multiple associated SNPs at the TLR6-TLR1-TLR10 gene cluster were dependent and suggest the presence of a founder prostate cancer risk variant on this haplotype background. The TLR6-TLR1-TLR10 gene cluster may play a role in prostate cancer risk, although further functional studies are needed to pinpoint the disease-associated variants in this gene cluster.

    • Comprehensive molecular simulations have been carried out to compute local structures and transport properties of different components of binary NaCl-KCl over a wide operating temperature range. The partial radial distribution functions, coordination number curves and angular distribution functions were calculated to analyze the influence of temperature and component on local structures of molten Alkali Chlorides. Transport properties were calculated by using reverse non-equilibrium molecular dynamics (RNEMD) simulations including densities, shear viscosity and thermal conductivity. The results show that ion clusters are considered to be formed and the distance of ion clusters become larger with increasing temperature which has great influence on macro-properties. The calculated properties have a good agreement with the experimental data, and similar method could be used to computationally calculate the properties of various molten salts and their mixtures.

    • The water-energy nexus has been introduced into urban water supply systems (WSSs) to improve the current plight of scarce resources and greenhouse effect in recent years. Urban water-energy integrated management is limited by the characteristics and reality of each city. In this paper, we characterize the comprehensive influence factors of energy use in China urban WSSs including geographic differences and city forms. The results indicate that the pressure of pipeline network and plain area ratio restricted by geomorphology would significantly impact the energy consumption during conveyance and distribution stage. For the city form aspect, the total volume of urban water supply and the leakage rate of pipeline networks play important roles in energy consumption of urban WSSs in China. In this study, the specific electricity consumption in WSSs was quantified, and several factors affected by city features which show strong correlation with energy use were determined. The results are of great significance to the energy saving in water supply systems in urban areas.

  • Multi-stage supply restoration of active distribution networks with SOP integration

    • Supply restoration from outages is essential for improving the reliability of active distribution networks (ADNs) after fault isolation. Soft open point (SOP) can adjust the power flow among feeders and provide voltage support for power outage areas. Considering the sequential coordination of switching operation and SOP control mode selection, a multi-stage supply restoration method with SOPs is proposed for ADNs. First, the sequential energization is formulated, in which the impact of SOP control mode on restoration sequence is analyzed. By providing voltage support, the coordination of SOPs will rapidly energize the outage area and improve the voltage profile. Then, a multi-stage restoration model with SOPs is proposed, in which reconfiguration of switches and control mode selection of SOPs are coordinated in sequence to maximize the load recovery level of ADNs. Through the switching action-time mapping, secure operation is ensured during the entire supply restoration process. Finally, the effectiveness of the proposed method is validated on a modified IEEE 33-node distribution system and practical distribution networks with four-terminal SOP. Results show that the proposed method can fully exploit the potential benefits of SOPs and effectively enhance the load recovery level of ADNs.

    • Energy Policy; Energy Resources; Energy Systems; Energy Management © 2020 This study develops a new concept involving using the existing infrastructure for photovoltaic (PV) generation to reduce the costs associated with increased land use and to avoid curtailment due to the mismatch between power supply and demand. We establish a method to estimate the technological potential and economic performance of the PV systems deployed in coal-fired power plants in China. The potential capacity of the examined 1,082 units in China reaches 4 GWe, which is equivalent to 32% of China's newly installed distributed PV capacity in 2019. A total of 87% of PV systems achieve plant-side grid parity compared with desulfurized coal benchmark electricity prices. To the best of our knowledge, this is the first study that investigates the use of rooftops and coal storage sheds in power plants to facilitate low-cost, flexible PV power generation, thus opening a new channel for future PV generation development.

  • Effects of organic composition on mesophilic anaerobic digestion of food waste

    • Anaerobic digestion of food waste (FW) has been widely investigated, however, little is known about the influence of organic composition on the FW digestion process. This study aims to identify the optimum composition ratios of carbohydrate (CA), protein (CP) and lipid (EE) for maintaining high methane yield and process stability. The results show that the CA–CP–EE ratio was significantly correlated with performance and degradability parameters. Controlling the CA–CP–EE ratio higher than 1.89 (CA higher than 8.3%, CP lower than 5.0%, and EE lower than 5.6%) could be an effective way to maintain stable digestion and achieve higher methane production (385–627 mL/g VS) and shorter digestion retention (196–409 h). The CA-CP-EE ratio could be used as an important indicator for digestion performance. To effectively evaluate organic reduction, the concentration and removal efficiency of organic compositions in both solid phases and total FW should be considered.
