    • Digital microfluidic biochips (DMFBs) facilitate modern healthcare applications. One of the most important applications is point-of-care (POC) clinical diagnosis which is directly related to human health. However, biochemical experiments are usually error-prone, which makes monitoring intermediate results during bioassay execution be required to ensure the correctness of POC clinical diagnosis. To tackle this problem, cyber-physical digital microfluidic biochips with integrated sensors have been proposed and have attracted attention recently. In order to fully utilize the sensors for detection, the reusability of sensors should be taken into consideration during the module placement stage of cyber-physical DMFBs synthesis flow. Moreover, excessive actuation of electrodes may cause reliability degradation and this issuse should also be taken care of during module placement stage. In order to deal with the aforementioned problems, this paper presents the first co-optimization method for both module and sensor placement. Experimental results show that the proposed method can effectively minimize bioassay completion time while meeting all constraints from sensors and electrodes.

    • OBJECTIVE: Capillary network formation represents a specialized endothelial cell function and is a prerequisite to establish a continuous vessel lumen. Formation of endothelial cell connections that form the vascular structure is regulated, at least in part, at the transcriptional level. We report here that related transcription enhancer factor-1 (RTEF-1) plays an important role in vascular structure formation.METHODS AND RESULTS: Knockdown of RTEF-1 by small interfering RNA or blockage of RTEF-1 function by the transcription enhancer activators domain decreased endothelial connections in a Matrigel assay, whereas overexpression of RTEF-1 in endothelial cells resulted in a significant increase in cell connections and aggregation. In a model of oxygen-induced retinopathy, endothelial-specific RTEF-1 overexpressing mice had enhanced angiogenic sprouting and vascular structure remodeling, resulting in the formation of a denser and more highly interconnected superficial capillary plexus. Mechanistic studies revealed that RTEF-1 induced the expression of functional gap junction proteins including connexin 43, connexin 40, and connexin 37. Blocking connexin 43 function inhibited RTEF-1-induced endothelial cell connections and aggregation.CONCLUSIONS: These findings provide novel insights into the transcriptional control of endothelial function in the coordination of cell-cell connections.

    • COPI (coat protein I) and the clathrin-AP-2 (adaptor protein 2) complex are well-characterized coat proteins, but a component that is common to these two coats has not been identified. The GTPase-activating protein (GAP) for ADP-ribosylation factor 1 (ARF1), ARFGAP1, is a known component of the COPI complex. Here, we show that distinct regions of ARFGAP1 interact with AP-2 and coatomer (components of the COPI complex). Selectively disrupting the interaction of ARFGAP1 with either of these two coat proteins leads to selective inhibition in the corresponding transport pathway. The role of ARFGAP1 in AP-2-regulated endocytosis has mechanistic parallels with its roles in COPI transport, as both its GAP activity and coat function contribute to promoting AP-2 transport.

    • A simple hydrothermal route to the preparation of the boehmite-doped AgCl nanocubes using AgNO3,AlCl3 center dot 6H(2)O and NaOH at 200 degrees C for 24 h is reported. The products were characterized by X-ray powder diffraction (XRD), Fourier transform infrared spectrometry (FT-IR), field-emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), selected area electron diffraction (SAED), UV-vis, thermogravimetric analysis (TGA), and differential thermal analysis (DTA). FE-SEM and TEM micrographs showed that the obtained boehmite-doped AgCl had nanocube-like morphology. The influence of heating temperature on the phase, microstructure, morphology, and thermal stability of the products were also investigated. UV-visible results indicated that the absorption edge moved to higher wavelength with the increasing heating temperature. These materials would be a promising material for photocatalyst applications.

    • The amount of available hypoxia-inducible factor (HIF)-1α has been considered to be largely a consequence of post-translational modification by multiple ubiquitin-proteasome pathways. However, the role of transcriptional regulation of HIF-1α is less certain, and the mechanisms of transcriptional regulation of HIF-1α require further investigation. Here we report that related transcriptional enhancer factor-1 (RTEF-1), a member of the TEF transcriptional factor family, transcriptionally regulates the HIF-1α gene under normoxic and hypoxic conditions. The expression of HIF-1α mRNA was decreased in endothelial cells in which RTEF-1 was knocked down with siRNA. Sequential deletional analysis of the HIF-1α promoter revealed that the MCAT-like element in the HIF-1α promoter was essential for HIF-1α transcription. Binding of RTEF-1 to the MCAT-like element was confirmed by ChIP. Treatment of endothelial cells with a HIF-1 inhibitor resulted in retardation of RTEF-1-induced proliferation and tube formation. Moreover, increased HIF-1α expression was observed in transgenic mice expressing RTEF-1 under the VE-cadherin promoter (VE-Cad/RTEF-1). VE-Cad/RTEF-1 mice subjected to hindlimb ischemia demonstrated increased levels of HIF-1α, accelerated recovery of blood flow, and increased capillary density compared with littermate controls. These results identify RTEF-1 as a regulator of HIF-1α transcription, which results in up-regulation of HIF-1α and acceleration of recovery from ischemia.

  • Assembly and succession of the phyllosphere microbiome and nutrient-cycling genes during plant community development in a glacier foreland

    • The phyllosphere, particularly the leaf surface of plants, harbors a diverse range of microbiomes that play a vital role in the functioning of terrestrial ecosystems. However, our understanding of microbial successions and their impact on functional genes during plant community development is limited. In this study, considering core and satellite microbial taxa, we characterized the phyllosphere microbiome and functional genes in various microhabitats (i.e., leaf litter, moss and plant leaves) across the succession of a plant community in a low-altitude glacier foreland. Our findings indicate that phyllosphere microbiomes and associated ecosystem stability increase during the succession of the plant community. The abundance of core taxa increased with plant community succession and was primarily governed by deterministic processes. In contrast, satellite taxa abundance decreased during plant community succession and was mainly governed by stochastic processes. The abundance of microbial functional genes (such as C, N, and P hydrolysis and fixation) in plant leaves generally increased during the plant community succession. However, in leaf litter and moss leaves, only a subset of functional genes (e.g., C fixation and degradation, and P mineralization) showed a tendency to increase with plant community succession. Ultimately, the community of both core and satellite taxa collaboratively influenced the characteristics of phyllosphere nutrient-cycling genes, leading to the diverse profiles and fluctuating abundance of various functional genes during plant community succession. These findings offer valuable insights into the phyllosphere microbiome and plant–microbe interactions during plant community development, advancing our understanding of the succession and functional significance of the phyllosphere microbial community.

    • The so-called merit factor approach (MFA) to radar binary sequence design has led to several theoretical contributions in fairly diverse research areas including information theory, computer science, combinatorial optimization, and analytical number theory. However, the MFA-which basically aims at minimizing the clutter effect on radar performance-implicitly assumes the use of a least squares (LS) receiver that is optimal only when there is no clutter. This problem can be eliminated by using a more general optimal instrumental-variables (IV) receiver in lieu of the LS receiver. The IV receiver can reject clutter more efficiently than the LS receiver. Additionally, the binary sequence design problem associated with the IV approach has an interesting form.

However, the pragmatic motivation of this approach is shown here to be questionable. Specifically, the MFA - which basically aims at minimizing the clutter effect on radar performance - implicitly assumes the use of a least-squares (LS) receiver that is optimal only when there is no clutter. This apparent inconsistency in the problem formulation can be eliminated by using a more general optimal instrumental-variables (M receiver in lieu of the LS receiver. The IV receiver proposed here is shown to reject clutter much more efficiently than the LS receiver. Additionally, the binary sequence design problem associated with the IV receiver is shown to have an interesting form that is likely to attract the attention of the readers previously interested in the MFA.

    • Pulse compression radar systems make use of transmit code sequences and receive filters that are specially designed to achieve good range resolution and target detection capability at practically acceptable transmit peak power levels. The present paper is a contribution to the literature on the problem of designing transmit codes and receive filters for radar. In a nutshell: the main goal of this paper, which considers the cases of both negligible and non-negligible Doppler shifts, is to show how to design the receive filter (including its length) and the transmit code sequence via. the optimization of a number of relevant metrics considered separately or in combination. The paper also contains several numerical studies whose aim is to illustrate the performance of the proposed designs.
