  • Genome-wide association study of prostate-specific antigen levels in 392,522 men identifies new loci and improves cross-ancestry prediction

    • We conducted a multi-ancestry genome-wide association study of prostate-specific antigen (PSA) levels in 296,754 men (211,342 European ancestry; 58,236 African ancestry; 23,546 Hispanic/Latino; 3,630 Asian ancestry; 96.5% of participants were from the Million Veteran Program). We identified 318 independent genome-wide significant (p≤5e-8) variants, 184 of which were novel. Most demonstrated evidence of replication in an independent cohort (n=95,768). Meta-analyzing discovery and replication (n=392,522) identified 447 variants, of which a further 111 were novel. Out-of-sample variance in PSA explained by our new polygenic risk score reached 16.9% (95% CI=16.1%-17.8%) in European ancestry, 9.5% (95% CI=7.0%-12.2%) in African ancestry, 18.6% (95% CI=15.8%-21.4%) in Hispanic/Latino, and 15.3% (95% CI=12.7%-18.1%) in Asian ancestry, and lower for higher age. Our study highlights how including proportionally more participants from underrepresented populations improves genetic prediction of PSA levels, with potential to personalize prostate cancer screening.

  • On the evolution of the size of Lyman alpha halos across cosmic time: no change in the circumgalactic gas distribution when probed by line emission

    • Lyman ? (Ly?) is now routinely used as a tool for detecting high redshift galaxies and its resonant nature means it can trace neutral hydrogen around star-forming galaxies. Integral field spectrograph measurements of high-? Ly? emitters indicate that significant extended Ly? halo emission is ubiquitous around such objects. We present a sample of redshift 0.23 to 0.32 galaxies observed with the Hubble Space Telescope selected to match the star formation properties of high-? samples while optimizing the observations for detection of low surface brightness Ly? emission. All seven targets show Ly? emission, with total escape fractions between 0.7% and 37%, and extended Ly? emission. We find Ly? halo to UV scale length ratios around 10:1, and halo flux fractions between 60% and 85% —consistent with high redshift observations—when using comparable methods. However, our targets show additional extended stellar UV emission which we parametrize with a new three exponential component model for Ly? emission. Halo fractions become much smaller with this methodology and some become consistent with zero. Comparing UV and H? emission reveals that H? is also extended and can partially explain the origin of diffuse Ly?. We interpret this as evidence that the creation of Ly? halos cannot be attributed to one single process but rather a combination of in-situ recombination and spatial scattering or possibly collisional excitation. We discuss our results in the context of high-redshift galaxies, evolutionary studies of the circumgalactic medium in emission, and inferences concerning the emission of ionizing radiation.

  • Micromechanics and microstructure evolution during in situ uniaxial tensile loading of TRIP-assisted duplex stainless steels

    • Two transformation-induced plasticity (TRIP) assisted duplex stainless steels, with three different stabilities of the austenite phase, were investigated by synchrotron X-ray diffraction characterization during in situ uniaxial tensile loading. The micromechanics and the deformation-induced martensitic transformation (DIMT) in the bulk of the steels were investigated in situ. Furthermore, scanning electron microscopy supplemented the in situ analysis by providing information about the microstructure of annealed and deformed specimens. The dependence of deformation structure on austenite stability is similar to that of single-phase austenitic steels where shear bands and bcc-martensite (α’) are generally observed, and blocky α’ is only frequent when the austenite stability is low. These microstructural features, i.e. defect structure and deformation-induced martensite, are correlated with the micro- and macro-mechanics of the steels with elastoplastic load transfer from the weaker phases to the stronger α’, in particular this occurs close to the point of maximum rate of α’ formation. A clear strain-hardening effect from α’ is seen in the most unstable austenite leading to a pronounced TRIP effect.

  • Comparison of histological delineations of medial temporal lobe cortices by four independent neuroanatomy laboratories

    • The medial temporal lobe (MTL) cortex, located adjacent to the hippocampus, is crucial for memory and prone to the accumulation of certain neuropathologies such as Alzheimer's disease neurofibrillary tau tangles. The MTL cortex is composed of several subregions which differ in their functional and cytoarchitectonic features. As neuroanatomical schools rely on different cytoarchitectonic definitions of these subregions, it is unclear to what extent their delineations of MTL cortex subregions overlap. Here, we provide an overview of cytoarchitectonic definitions of the cortices that make up the parahippocampal gyrus (entorhinal and parahippocampal cortices) and the adjacent Brodmann areas (BA) 35 and 36, as provided by four neuroanatomists from different laboratories, aiming to identify the rationale for overlapping and diverging delineations. Nissl-stained series were acquired from the temporal lobes of three human specimens (two right and one left hemisphere). Slices (50 µm thick) were prepared perpendicular to the long axis of the hippocampus spanning the entire longitudinal extent of the MTL cortex. Four neuroanatomists annotated MTL cortex subregions on digitized (20X resolution) slices with 5 mm spacing. Parcellations, terminology, and border placement were compared among neuroanatomists. Cytoarchitectonic features of each subregion are described in detail. Qualitative analysis of the annotations showed higher agreement in the definitions of the entorhinal cortex and BA35, while definitions of BA36 and the parahippocampal cortex exhibited less overlap among neuroanatomists. The degree of overlap of cytoarchitectonic definitions was partially reflected in the neuroanatomists' agreement on the respective delineations. Lower agreement in annotations was observed in transitional zones between structures where seminal cytoarchitectonic features are expressed more gradually. The results highlight that definitions and parcellations of the MTL cortex differ among neuroanatomical schools and thereby increase understanding of why these differences may arise. This work sets a crucial foundation to further advance anatomically-informed human neuroimaging research on the MTL cortex.
