  • Genetic Studies of Leptin Concentrations Implicate Leptin in the Regulation of Early Adiposity

    • Leptin influences food intake by informing the brain about the status of body fat stores. Rare LEP mutations associated with congenital leptin deficiency cause severe early-onset obesity that can be mitigated by administering leptin. However, the role of genetic regulation of leptin in polygenic obesity remains poorly understood. We performed an exome-based analysis in up to 57,232 individuals of diverse ancestries to identify genetic variants that influence adiposity-adjusted leptin concentrations. We identify five novel variants, including four missense variants, in LEP, ZNF800, KLHL31, and ACTL9, and one intergenic variant near KLF14. The missense variant Val94Met (rs17151919) in LEP was common in individuals of African ancestry only, and its association with lower leptin concentrations was specific to this ancestry (P = 2 × 10-16, n = 3,901). Using in vitro analyses, we show that the Met94 allele decreases leptin secretion. We also show that the Met94 allele is associated with higher BMI in young African-ancestry children but not in adults, suggesting that leptin regulates early adiposity.

  • EISCAT_3D - a next-generation European radar system for upper atmosphere and geospace research

    • The EISCAT Scientifi c Association, together with a number of collaborating institutions, has recently completed a feasibility and design study for an enhanced performance research radar facility to replace the existing EISCAT UHF and VHF systems. This study was supported by EU Sixth-Framework funding. The new radar retains the powerful multi-static geometry of the EISCAT UHF, but will employ phased arrays, direct-sampling receivers, and digital beamforming and beam steering. Design goals include, inter alia, a tenfold improvement in temporal and spatial resolution, an extension of the instantaneous measurement of full-vector ionospheric drift velocities from a single point to the entire altitude range of the radar, and an imaging capability to resolve small-scale structures. Prototype receivers and beamformers are currently being tested on a 48-element, 224 MHz array (the "Demonstrator") erected at the Kiruna EISCAT site, using the EISCAT VHF transmitter as an illuminator.

  • Molecular motion of the Morpholin-1-yl radical in CF2 ClCFCl2 as studied by ESR : use of residual anisotrophy of powder spectra to extract dynamics

    • The dynamics of the deprotonated neutral morpholin-1-yl radical, trapped in a halocarbon matrix, CF2ClCFCl2, has been studied utilizing electron spin resonance (ESR) spectroscopy. The experimental lineshapes of the radical exhibit an alterating line-width effect in the temperature range 105-144 K. The major changes of the ESR lineshape were governed by the averaging of the nitrogen hyperfine anisotropy while no exchange of the isotropic hyperfine coupling constants was observed. Geometrical parameters specifying the restricted anisotropic rotation of the whole radical trapped in the matrix could be extracted. Two methods for simulating anisotropic exchange broadened ESR spectra, the secular and non-perturbative, were utilized to investigate the dynamics of the radical. A surprisingly simple ''three-site'' jump model with a barrier of almost-equal-to 3.6 kcal/mol can be applied in the simulation of the experimental spectra. It has been shown that the secular method cannot reproduce the exchange broadened ESR spectra of systems with large hyperfine anisotropy undergoing large internal reorganisation.

    • Trans-azobenzene dissolved in different liquid hydrocarbons absorbs fluorescence arising from all acceptors previously used in Fluorescence Detected Magnetic Resonance (FDMR) and Optically Detected ESR (OD ESR) spectroscopy making optical detection impossible. In this report a new acceptor, rubrene, having sufficient quantum yield of fluorescence in the red band 550-620 nm, has been proven successful. OD ESR spectra of the radical-ion pair trans-azobenzene+/rubrene- were detected in liquid squalane (2,6,10,15,19,23-hexamethyl-tetracosane) solution in the temperature range 294-243 K. The experimental isotropic hyperfine splittings of the radical cation of trans-azobenzene (a(N) = 1.4 mT) have been compared with those from MNDO/INDO calculations and with those of earlier work using freon matrix studies.

    • The current study presents the effects of strain and temperature on the mechanical response and microstructure evolution in AA7075-T651 at high strain rates. Compression tests have been performed at room temperature (RT), 200, 300 and 400 °C using a Split-Hopkinson pressure bar (SHPB) setup with strain rates ranging between 1400 and 5300 s−1. For deformation at RT, the flow stress increases with increase in strain rate. Whereas deformation at elevated temperatures show a non-monotonous behavior of the flow stress with respect to the strain rate. This trait is attributed to the pronounced effects from the adiabatic shear bands (ASBs); namely, distorted shear bands (DSBs) and transformed shear bands (TSBs); and cracks resulting from the plastic deformation instability during hot deformation. The sequence of microstructure evolution is: inhomogeneity in the initial microstructure – DSB – TSB – crack –fracture. The feasibility of formation and growth of ASBs and cracks increases with increase in strain and temperature, neglecting any significant effect from the strain rate. During the compression tests, temperature of the material rises due to adiabatic heating. Considering a certain strain developed in the material, this adiabatic temperature rise decreases as the deformation temperature is increased. Furthermore, during individual deformation processes, the temperature rise increases with increasing strain. The adiabatic temperature leading to the formation of TSB is approximated to be 0.7 times of the melting temperature of the alloy. These results from the current study are to be used in developing a physics-based material model for the alloy.

  • Transient bending waves in anisotropic plates studied by hologram interferometry

    • Propagating bending waves are studied in plates made of glass-fiber reinforced polyester. The waves are generated by the impact of a ballistic pendulum. Hologram interferometry, with a double pulsed ruby laser as light source, is used to record the out of plane motion of the waves. The interferograms have an elliptic-like symmetry for an orthotropic plate, while the wave pattern for a symmetric angle-ply reinforced plate has a symmetry about the axes of reinforcements. Experimental data are compared on one hand to analytical results obtained by assuming that the orthotropic plate can be described as if isotropic along the main axes, and on the other hand to numerical results from calculations using the finite-element method. The effective Young's modulus raised to power 1/4 is shown to be an important parameter for the description of the dispersive wave pattern. A defect in the plate alters the wave pattern in the interferograms significantly. This may have technical use.

The methodology used is capable of allowing for interactions between thermal, mechanical, and transformation processes. Experimental values for a 2.25Cr-1Mo ferritic stainless steel were used in the simulation. It seems possible to simulate most welding situations given sufficient computer resources

    • An experimental and simulation study of residual stresses was made in the vicinity of a gas tungsten arc weld, used to join a hemispherical end cap to a cylinder. The capped cylinder is used in a satellite application and was fabricated from a Co-based Haynes® 25 alloy. The cylinder was 34.7 mm in outer diameter and 3.3 mm in thickness. The experimental measurements were made by neutron diffraction and the simulation used the implicit Marc finite element code. The experimental resolution was limited to approximately 3 mm parallel to the axis of the cylinder (the weld was 6 mm in the same direction) and comparison over the same volume of the finite element prediction showed general agreement. Subject to the limited spatial resolution, the largest experimentally measured tensile residual stress was 180 MPa, located at the middle of the weld. However, the predictions suggest that there are regions in the weld where average tensile residual stresses as much as 400 MPa exist. One qualitative disparity between the model and the experiments was that the measurement included a larger degree of asymmetry on either side of the weld than predicted by the model.
