  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Investigation of DC-Biased Optical OFDM With Precoding Matrix for Visible Light Communications : Theory, Simulations, and Experiments

    • Orthogonal frequency-division-multiplexing (OFDM) technology is widely used in visible light communication (VLC) to achieve high data rate transmission. However, the traditional direct-current (DC)-biased optical OFDM (DCO-OFDM) VLC systems suffer from the high peak-to-average power ratio (PAPR) which causes signal clipping distortion, and, thus, performance degradation. Furthermore, severe high-frequency fading due to the limited system bandwidth results in poor bit error rate (BER) performance. Precoding matrix (PM) techniques have been proposed to enhance the performance of VLC OFDM transmission, but a little or no work has been carried out in investigating the theory of PM used in OFDM VLC systems. In this paper, we aim to reveal the theory of PM-DCO-OFDM for a VLC system. To figure out the intrinsic laws of a PM method, we investigate the principles of PAPR reduction, clipping distortion optimization, and signal-to-noise ratio (SNR) distribution equalization. Based on the analysis of PAPR, we theoretically proved the simplicity of PM as a method to reduce the possibility of high PAPR by improving the autocorrelation performance of input symbols. The clipping distortion could be improved due to the reduction of high PAPR. Moreover, the relatively uniform SNR distribution can be achieved by PM through equalizing the clipping and channel noise, which is beneficial to improve the BER performance in high-frequency constrained systems. However, the PM method used in a DCO-OFDM VLC system should consider the transmitting power, modulation format, and transmission distance as a whole to achieve the transmission performance improvement. The simulation results demonstrate the complementary cumulative distribution function of PAPR can be reduced similar to 3 dB, while the performance of clipping distortion power and clipping error probability are significantly improved. Furthermore, experiment is carried out with results showing that the PM method can improve the BER performance in the case that VLC OFDM transmission has enough transmitting power, but with the low transmitting power, the PM also can damage the BER performance. The simulation and experiment results are consistent with our theoretical analysis.

  • High throughput decoding approach for luminescencekinetics-based optical encoding of lanthanide upconversion nanoparticles

    • Lanthanide upconversion nanoparticles (UCNPs) are increasingly explored to develop high-security-level anti-counterfeiting and multiplexing applications, due to the availability of ample encoding dimensions. Among other applications, upconversion luminescence(UCL) kinetics-based optical encoding is particularly attractive as it provides an almost unlimited encoding capacity due to the ease of manipulating the UCL kinetics by chemical engineering. However, current decoding methods limit its applications because of a typically low throughput and high cost of the system. In this Letter, we propose a novel decoding approach for UCL kinetics-based optical encoding, which utilizes a pulsed excitation source with adjustable pulse duration and a low time-resolution and large-area detector. We develop a theoretical fitting model and show how fingerprint time constants of the UCNPs (the encoding identities) can be extracted from the correlation between the averaged UCL intensity and the pulse duration. Our new approach provides a high-throughput and cost-effective solution to decode UCL kinetics.

  • Overview of physics studies on ASDEX Upgrade

    • The ASDEX Upgrade (AUG) programme, jointly run with the EUROfusion MST1 task force, continues to significantly enhance the physics base of ITER and DEMO. Here, the full tungsten wall is a key asset for extrapolating to future devices. The high overall heating power, flexible heating mix and comprehensive diagnostic set allows studies ranging from mimicking the scrape-off-layer and divertor conditions of ITER and DEMO at high density to fully non-inductive operation (q 95 = 5.5, ) at low density. Higher installed electron cyclotron resonance heating power 6 MW, new diagnostics and improved analysis techniques have further enhanced the capabilities of AUG. Stable high-density H-modes with MW m-1 with fully detached strike-points have been demonstrated. The ballooning instability close to the separatrix has been identified as a potential cause leading to the H-mode density limit and is also found to play an important role for the access to small edge-localized modes (ELMs). Density limit disruptions have been successfully avoided using a path-oriented approach to disruption handling and progress has been made in understanding the dissipation and avoidance of runaway electron beams. ELM suppression with resonant magnetic perturbations is now routinely achieved reaching transiently . This gives new insight into the field penetration physics, in particular with respect to plasma flows. Modelling agrees well with plasma response measurements and a helically localised ballooning structure observed prior to the ELM is evidence for the changed edge stability due to the magnetic perturbations. The impact of 3D perturbations on heat load patterns and fast-ion losses have been further elaborated. Progress has also been made in understanding the ELM cycle itself. Here, new fast measurements of and E r allow for inter ELM transport analysis confirming that E r is dominated by the diamagnetic term even for fast timescales. New analysis techniques allow detailed comparison of the ELM crash and are in good agreement with nonlinear MHD modelling. The observation of accelerated ions during the ELM crash can be seen as evidence for the reconnection during the ELM. As type-I ELMs (even mitigated) are likely not a viable operational regime in DEMO studies of 'natural' no ELM regimes have been extended. Stable I-modes up to have been characterised using -feedback. Core physics has been advanced by more detailed characterisation of the turbulence with new measurements such as the eddy tilt angle - measured for the first time - or the cross-phase angle of and fluctuations. These new data put strong constraints on gyro-kinetic turbulence modelling. In addition, carefully executed studies in different main species (H, D and He) and with different heating mixes highlight the importance of the collisional energy exchange for interpreting energy confinement. A new regime with a hollow profile now gives access to regimes mimicking aspects of burning plasma conditions and lead to nonlinear interactions of energetic particle modes despite the sub-Alfvénic beam energy. This will help to validate the fast-ion codes for predicting ITER and DEMO.

    •  Based on the structural characteristics and preliminary design of submerged floating tunnel (SFT) prototype in Qiandao Lake, this paper gives the risk index system of public safety of SFT and risk assessment methods by analyzing different impact factors of SFT public safety. Moreover, the public safety risk is evaluated during construction and operation of SFT prototype in Qiandao Lake by the presented analytic hierarchy process method. The results show that in spite of facing many technical problems and potential risks, these potential risks of SFT can be controlled or reduced to a minimum level with the help of reasonable design and certain measures

    • An unprecedented silver-promoted regioselective (4 + 1) annulation of isocyanoacetates with pyridinium salts is reported. The established protocol provides controlled, facile, and modular access to a range of synthetically useful N-fused heterocyclic scaffolds containing indolizines, pyrrolo[1,2-a]quinolines, pyrrolo[2,1-a]isoquinolines, and 1H-imidazo[4,5-a]indolizin-2(3H)-ones. A mechanistic pathway involving nucleophilic addition/protonation/elimination/cydoisomerization is proposed.

    • Conflict monitoring and processing are crucial components of the human cognitive system, with significant implications for daily life and the diagnosis of cognitive disorders. The Stroop task, combined with brain function detection technology, has been widely employed as a classical paradigm for investigating conflict processing. However, there remains a lack of public datasets that integrate Electroencephalogram (EEG) and functional Near-infrared Spectroscopy (fNIRS) to simultaneously record brain activity during a Stroop task. We introduce a dual-modality Stroop task dataset incorporating 34-channel EEG (sampling frequency is 1000 Hz) and 20-channel high temporal resolution fNIRS (sampling frequency is 100 Hz) measurements covering the whole frontal cerebral cortex from 21 participants (9 females/12 males, aged 23.0 ± 2.3 years). Event-related potential analysis of EEG recordings and activation analysis of fNIRS recordings were performed to show the significant Stroop effect. We expected that the data provided would be utilized to investigate multimodal data processing algorithms during cognitive processing.
