  • A high-stringency blueprint of the human proteome

    • The Human Proteome Organization (HUPO) launched the Human Proteome Project (HPP) in 2010, creating an international framework for global collaboration, data sharing, quality assurance and enhancing accurate annotation of the genome-encoded proteome. During the subsequent decade, the HPP established collaborations, developed guidelines and metrics, and undertook reanalysis of previously deposited community data, continuously increasing the coverage of the human proteome. On the occasion of the HPP’s tenth anniversary, we here report a 90.4% complete high-stringency human proteome blueprint. This knowledge is essential for discerning molecular processes in health and disease, as we demonstrate by highlighting potential roles the human proteome plays in our understanding, diagnosis and treatment of cancers, cardiovascular and infectious diseases.

  • Risk factors for knee osteoarthritis after traumatic knee injury : A systematic review and meta-analysis of randomised controlled trials and cohort studies for the OPTIKNEE Consensus

    • Objective: To identify and quantify potential risk factors for osteoarthritis (OA) following traumatic knee injury. Design: Systematic review and meta-analyses that estimated the odds of OA for individual risk factors assessed in more than four studies using random-effects models. Remaining risk factors underwent semiquantitative synthesis. The modified GRADE (Grading of Recommendations Assessment, Development and Evaluation) approach for prognostic factors guided the assessment. Data sources: MEDLINE, EMBASE, CENTRAL, SPORTDiscus, CINAHL searched from inception to 2009-2021. Eligibility: Randomised controlled trials and cohort studies assessing risk factors for symptomatic or structural OA in persons with a traumatic knee injury, mean injury age ≤30 years and minimum 2-year follow-up. Results: Across 66 included studies, 81 unique potential risk factors were identified. High risk of bias due to attrition or confounding was present in 64% and 49% of studies, respectively. Ten risk factors for structural OA underwent meta-analysis (sex, rehabilitation for anterior cruciate ligament (ACL) tear, ACL reconstruction (ACLR), ACLR age, ACLR body mass index, ACLR graft source, ACLR graft augmentation, ACLR+cartilage injury, ACLR+partial meniscectomy, ACLR+total medial meniscectomy). Very-low certainty evidence suggests increased odds of structural OA related to ACLR+cartilage injury (OR=2.31; 95% CI 1.35 to 3.94), ACLR+partial meniscectomy (OR=1.87; 1.45 to 2.42) and ACLR+total medial meniscectomy (OR=3.14; 2.20 to 4.48). Semiquantitative syntheses identified moderate-certainty evidence that cruciate ligament, collateral ligament, meniscal, chondral, patellar/tibiofemoral dislocation, fracture and multistructure injuries increase the odds of symptomatic OA. Conclusion: Moderate-certainty evidence suggests that various single and multistructure knee injuries (beyond ACL tears) increase the odds of symptomatic OA. Risk factor heterogeneity, high risk of bias, and inconsistency in risk factors and OA definition make identifying treatment targets for preventing post-traumatic knee OA challenging.

  • Tungsten spectroscopy at the Livermore electron beam ion trap facility

    • The utilization of tungsten spectroscopy for diagnostics of magnetically confined fusion plasmas requires the radiative properties of tungsten ions to be accurately known. At the Lawrence Livermore National Laboratory, a program to gather spectroscopic data on tungsten ions has been initiated with the purpose to study spectral signatures and identify candidate fusion plasma diagnostics. In this paper, an overview of recent results from the Livermore WOLFRAM spectroscopy project is presented, which includes experimental investigations at the EBIT-I and SuperEBIT electron beam ion traps. In particular, the spectra of highly charged M-and L-shell tungsten ions have been studied. These investigations cover energy measurements of n = 2 to n = 2, 3 transitions in Ne-like W64+ through Li-like W71+ ions and soft X-ray measurements of n = 3 to n = 3, 4 transitions in M-shell ions with emphasis on the Ni-like W46+ and Si-like W60+ through Na-like W63+ ions. The measurements are complemented by atomic-structure calculations and spectral modeling using the Flexible Atomic Code (FAC).

  • Unmet challenges in septoplasty-nordic studies from a uniform healthcare and geographical area

    • PurposeNasal septoplasty is one of the most common surgical procedures in otorhinolaryngology and optimising both patient selection and the surgery is a challenge. The Nordic countries have similar public healthcare systems and comparable populations in terms of size. MethodsThis is a review of studies of outcome and predictors related to septoplasty from Denmark, Finland, Norway and Sweden, published during the last decade. The aim of this review was to identify areas in need of further research to meet the challenges of septoplasty in the Nordic countries with reference to international data. ResultsPostoperative patient satisfaction at 6-12 months was reported in around 2/3 of the patients and well in line with international data. Patients with more severe symptoms had a higher chance of improvement. Lack of standardisation in patient selection, surgical methods and skills, and follow up procedures, still makes it difficult to explain the 25% failure rate in septoplasty surgery. ConclusionThis review of the Nordic studies from the last decade shows that septoplasty in general is effective in relieving nasal obstruction. There is a need for studies addressing the standardisation of diagnostic tools and algorithms and the systematic and continuous implementation of follow-up of the surgical results at both departmental and personal level. This includes an awareness of how surgical skills in septoplasty are obtained and maintained.

    • ObjectivesThe use of magnetic resonance imaging (MRI) is safe from a long-term perspective since there are no known cumulative risks for patients or personnel. However, the technique comes with several acute risks associated with the powerful electromagnetic fields that are necessary to produce medical images. These risks include, among other things, a projectile hazard, loud noise, and the risk of heating. Safe use of MRI requires knowledge about the different hazards related to MRI and organizational structured work including the implementation of routines describing a safe workflow from the referral of a patient to the signed report. In this article, the risks associated with MRI are described along with suggestions for how each risk can be minimized or eliminated.ConclusionThe aim of this article is to provide support for the development of, and compliance with, MRI safety routines, and to work with the technique in a safe way. The scope of this treatise does not cover specific details of implant safety, however, the physical principles described can be applied to the risk assessment of implants.Key Points.
