    • As part of the REgional Carbon Cycle Assessment and Processes Phase 2 (RECCAP2) project, we developed a comprehensive African Greenhouse gases (GHG) budget covering 2000 to 2019 (RECCAP1 and RECCAP2 time periods), and assessed uncertainties and trends over time. We compared bottom-up process-based models, data-driven remotely sensed products, and national GHG inventories with top-down atmospheric inversions, accounting also for lateral fluxes. We incorporated emission estimates derived from novel methodologies for termites, herbivores, and fire, which are particularly important in Africa. We further constrained global woody biomass change products with high-quality regional observations. During the RECCAP2 period, Africa's carbon sink capacity is decreasing, with net ecosystem exchange switching from a small sink of −0.61 ± 0.58 PgC yr−1 in RECCAP1 to a small source in RECCAP2 at 0.16 (−0.52/1.36) PgC yr−1. Net CO2 emissions estimated from bottom-up approaches were 1.6 (−0.9/5.8) PgCO2 yr−1, net CH4 were 77 (56.4/93.9) TgCH4 yr−1 and net N2O were 2.9 (1.4/4.9) TgN2O yr−1. Top-down atmospheric inversions showed similar trends. Land Use Change emissions increased, representing one of the largest contributions at 1.7 (0.8/2.7) PgCO2eq yr−1 to the African GHG budget and almost similar to emissions from fossil fuels at 1.74 (1.53/1.96) PgCO2eq yr−1, which also increased from RECCAP1. Additionally, wildfire emissions decreased, while fuelwood burning increased. For most component fluxes, uncertainty is large, highlighting the need for increased efforts to address Africa-specific data gaps. However, for RECCAP2, we improved our overall understanding of many of the important components of the African GHG budget that will assist to inform climate policy and action.

ImportanceThe association of tumor-infiltrating lymphocyte (TIL) abundance in breast cancer tissue with cancer recurrence and death in patients with early-stage triple-negative breast cancer (TNBC) who are not treated with adjuvant or neoadjuvant chemotherapy is unclear. ObjectiveTo study the association of TIL abundance in breast cancer tissue with survival among patients with early-stage TNBC who were treated with locoregional therapy but no chemotherapy. Design, Setting, and ParticipantsRetrospective pooled analysis of individual patient-level data from 13 participating centers in North America (Rochester, Minnesota; Vancouver, British Columbia, Canada), Europe (Paris, Lyon, and Villejuif, France; Amsterdam and Rotterdam, the Netherlands; Milan, Padova, and Genova, Italy; Gothenburg, Sweden), and Asia (Tokyo, Japan; Seoul, Korea), including 1966 participants diagnosed with TNBC between 1979 and 2017 (with follow-up until September 27, 2021) who received treatment with surgery with or without radiotherapy but no adjuvant or neoadjuvant chemotherapy. ExposureTIL abundance in breast tissue from resected primary tumors. Main Outcomes and MeasuresThe primary outcome was invasive disease-free survival [iDFS]. Secondary outcomes were recurrence-free survival [RFS], survival free of distant recurrence [distant RFS, DRFS], and overall survival. Associations were assessed using a multivariable Cox model stratified by participating center. ResultsThis study included 1966 patients with TNBC (median age, 56 years [IQR, 39-71]; 55% had stage I TNBC). The median TIL level was 15% (IQR, 5%-40%). Four-hundred seventeen (21%) had a TIL level of 50% or more (median age, 41 years [IQR, 36-63]), and 1300 (66%) had a TIL level of less than 30% (median age, 59 years [IQR, 41-72]). Five-year DRFS for stage I TNBC was 94% (95% CI, 91%-96%) for patients with a TIL level of 50% or more, compared with 78% (95% CI, 75%-80%) for those with a TIL level of less than 30%; 5-year overall survival was 95% (95% CI, 92%-97%) for patients with a TIL level of 50% or more, compared with 82% (95% CI, 79%-84%) for those with a TIL level of less than 30%. At a median follow-up of 18 years, and after adjusting for age, tumor size, nodal status, histological grade, and receipt of radiotherapy, each 10% higher TIL increment was associated independently with improved iDFS (hazard ratio [HR], 0.92 [0.89-0.94]), RFS (HR, 0.90 [0.87-0.92]), DRFS (HR, 0.87 [0.84-0.90]), and overall survival (0.88 [0.85-0.91]) (likelihood ratio test, P<10e-6). Conclusions and RelevanceIn patients with early-stage TNBC who did not undergo adjuvant or neoadjuvant chemotherapy, breast cancer tissue with a higher abundance of TIL levels was associated with significantly better survival. These results suggest that breast tissue TIL abundance is a prognostic factor for patients with early-stage TNBC. This study of patients with early-stage triple-negative breast cancer not treated with adjuvant or neoadjuvant chemotherapy analyzes the association between tumor-infiltrating lymphocyte levels, cancer recurrence, and survival.

  • Anticipatory cues can mitigate car sickness on the road

    • Car passengers experience much more car sickness than car drivers. We assume that this is because drivers can better anticipate the car's motions. Does helping passengers to anticipate the car's motions then mitigate car sickness? Indeed, laboratory studies have shown that anticipatory cues which announce one-dimensional motions of a linear sled mitigate sickness to a small extent. Does this mitigation generalize to real car driving? We tested this in a car ride on a test track along a trajectory involving lane changes, accelerations, and decelerations. We show that vibrotactile cues mitigated car sickness in passengers. Auditory cues were less effective. The mitigating effect of the vibrotactile cue was considerable: a 40% decrease in car sickness symptoms, a larger effect than we found in the laboratory. Automated vehicles can predict their own motion very well. They could thus provide vibrotactile cues to mitigate car sickness in their passengers.

    • Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its ‘Minimal Information for Studies of Extracellular Vesicles’, which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly.

  • Search for Na-22 in novae supported by a novel method for measuring femtosecond nuclear lifetimes

    • Classical novae are thermonuclear explosions in stellar binary systems, and important sources of Al-26 and Na-22. While ? rays from the decay of the former radioisotope have been observed throughout the Galaxy, Na-22 remains untraceable. Its half-life (2.6 yr) would allow the observation of its 1.275 MeV ?-ray line from a cosmic source. However, the prediction of such an observation requires good knowledge of its nucleosynthesis. The Na-22(p, ?)Mg-23 reaction remains the only source of large uncertainty about the amount of Na-22 ejected. Its rate is dominated by a single resonance on the short-lived state at 7785.0(7) keV in Mg-23. Here, we propose a combined analysis of particle-particle correlations and velocity-difference profiles to measure femtosecond nuclear lifetimes. The application of this method to the study of the Mg-23 states, places strong limits on the amount of Na-22 produced in novae and constrains its detectability with future space-borne observatories. The authors report a particle-particle correlation and velocity-difference profile method to measure nuclear lifetime. The results obtained for excited states of 23Mg are used to constrain the production of 22Na in the astrophysical novae explosions.

  • Examination of how properties of a fissioning system impact isomeric yield ratios of the fragments

    • The population of isomeric states in the prompt decay of fission fragments-so-called isomeric yield ratios (IYRs)-is known to be sensitive to the angular momentum J that the fragment emerged with, and may therefore contain valuable information on the mechanism behind the fission process. In this work, we investigate how changes in the fissioning system impact the measured IYRs of fission fragments to learn more about what parameters affect angular momentum generation. To enable this, a new technique for measuring IYRs is first demonstrated. It is based on the time of arrival of discrete gamma rays, and has the advantage that it enables the study of the IYR as a function of properties of the partner nucleus. This technique is used to extract the IYR of 134Te, strongly populated in actinide fission, from the three different fissioning systems: 232Th(n, f), 238U(n, f), at two different neutron energies, as well as 252Cf(sf). The impacts of changing the fissioning system, the compound nuclear excitation energy, the minimum J of the binary partner, and the number of neutrons emitted on the IYR of 134Te are determined. The decay code TALYS is used in combination with the fission simulation code FREYA to calculate the primary fragment angular momentum from the IYR. We find that the IYR of 134Te has a slope of 0.004 +/- 0.002 with increase in compound nucleus (CN) mass. When investigating the impact on the IYR of increased CN excitation energy, we find no change with an energy increase similar to the difference between thermal and fast fission. By varying the mass of the partner fragment emerging with 134Te, it is revealed that the IYR of 134Te is independent of the total amount of prompt neutrons emitted from the fragment pair. This indicates that neutrons carry minimal angular momentum away from the fission fragments. Comparisons with the FREYA+TALYS simulations reveal that the average angular momentum in 134Te following 238U(n, f) is 6.0 h over bar . This is not consistent with the value deduced from recent CGMF calculations. Finally, the IYR sensitivity to the angular momentum of the primary fragment is discussed. These results are not only important to help understanding the underlying mechanism in nuclear fission, but can also be used to constrain and benchmark fission models, and are relevant to the gamma -ray heating problem of reactors.
