  • Wall shear stress measured with 4D flow CMR correlates with biomarkers of inflammation and collagen synthesis in mild-to-moderate ascending aortic dilation and tricuspid aortic valves

    • Aims Understanding the mechanisms underlying ascending aortic dilation is imperative for refined risk stratification of these patients, particularly among incidentally identified patients, most commonly presenting with tricuspid valves. The aim of this study was to explore associations between ascending aortic haemodynamics, assessed using four-dimensional flow cardiovascular magnetic resonance imaging (4D flow CMR), and circulating biomarkers in aortic dilation. Methods and results Forty-seven cases with aortic dilation (diameter >= 40 mm) and 50 sex-and age-matched controls (diameter < 40 mm), all with tricuspid aortic valves, underwent 4D flow CMR and venous blood sampling. Associations between flow displacement, wall shear stress (WSS), and oscillatory shear index in the ascending aorta derived from 4D flow CMR, and biomarkers including interleukin-6, collagen type I alpha 1 chain, metalloproteinases (MMPs), and inhibitors of MMPs derived from blood plasma, were investigated. Cases with dilation exhibited lower peak systolic WSS, higher flow displacement, and higher mean oscillatory shear index compared with controls without dilation. No significant differences in biomarkers were observed between the groups. Correlations between haemodynamics and biomarkers were observed, particularly between maximum time-averaged WSS and interleukin-6 (r = 0.539, P < 0.001), and maximum oscillatory shear index and collagen type I alpha 1 chain (r = -0.575, P < 0.001 in cases). Conclusion Significant associations were discovered between 4D flow CMR derived whole-cardiac cycle WSS and circulating biomarkers representing inflammation and collagen synthesis, suggesting an intricate interplay between haemodynamics and the processes of inflammation and collagen synthesis in patients with early aortic dilation and tricuspid aortic valves.

  • Evaluation of ((4-Hydroxyphenyl)ethyl)maleimide for Site-Specific Radiobromination of Anti-HER2 Affibody

    • Affibody molecules are a new class of small phage-display selected proteins using a scaffold domain of the bacterial receptor protein A. They can be selected for specific binding to a large variety of protein targets. An affibody molecule binidng with high affinity to a tumor antigen HER2 was recently developed for radionuclide diagnostics and therapy in vivo. The use of hte positron-emitting nuclide 76Br(T½ = 16.2 h) could imporve the sensitivity of detection of HER2-expressing tumors. A site-specific radiobromination o fa cysteine-containing variant of the anti-HER2 affibody, (ZHER2:4)2-Cys, using ((4-hydroxpyphenyl)ethyl)maleimide (HPEM), was evaluated in this study. It was found that HPEM can be radiobrominated with an efficiency of 83+0.4% and thereafter coupled to freshly reduced conjugate to exceed 97%. The label was stable against challenge with large excess of nonlabeled bromide and in a high molar strengt solution. In vitro cell tests demonstraded that radiobrominated affibody binds specifically to the HER2-expressing cel-line, SK-OV-3. Biodistribution studies in nude mice bearing SK-OV-3 xenografts have shown tumor accumulation of 4.8 ? 2.2% IA/g and good tumor-to-normal tissue ratios.

  • Tumor imaging using a picomolar affinity HER2 binding affibody molecule

    • The detection of cell-bound proteins that are produced due to aberrant gene expression in malignant tumors can provide important diagnostic information influencing patient management. The use of small radiolabeled targeting proteins would enable high-contrast radionuclide imaging of cancers expressing such antigens if adequate binding affinity and specificity could be provided. Here, we describe a HER2-specific 6 kDa Affibody molecule (hereinafter denoted Affibody molecule) with 22 pmol/L affinity that can be used for the visualization of HER2 expression in tumors in vivo using gamma camera. A library for affinity maturation was constructed by re-randomization of relevant positions identified after the alignment of first-generation variants of nanomolar affinity (50 nmol/L). One selected Affibody molecule, Z(HER2:342) showed a >2,200-fold increase in affinity achieved through a single-library affinity maturation step. When radioiodinated, the affinity-matured Affibody molecule showed clear, high-contrast visualization of HER2-expressing xenografts in mice as early as 6 hours post-injection. The tumor uptake at 4 hours post-injection was improved 4-fold (due to increased affinity) with 9% of the injected dose per gram of tissue in the tumor. Affibody molecules represent a new class of affinity molecules that can provide small sized, high affinity cancer-specific ligands, which may be well suited for tumor imaging.

    • For the third year, the AHA festival investigates the meeting between art and science in a three-day event at the Chalmers University of Technology hosted by the Department of Architecture and the Department of Physics. An international festival intended to provide enlightening experiences, staging surprises, new thoughts and displaced perspectives that lead to alternative modes of thinking about exploring the world through art and science. We invite scientists (physicists, historians, astronomers, engineers), artists (dancers, musicians, painters, poets, acrobats) who reside in these borderlands and wish to share their vision and work. The key intention is to celebrate both art and science as key knowledge building devices.The first year’s theme ’Embodiment’ (2014) explored the body as our anchor in the world, followed by the 2015 theme on ’Numbers’, a delightful net we cast over the world. This year's theme is ’Uni-verse,’ again a natural consequence of our interest in the relation between art and science. The elemental force that drives science as well as art is curiosity. Come be curious with us! During the festival we have chosen to divide the word universe into three: uni and "-" and verse. Uni means that something is combined into a whole. Verse means that we are turned in a direction, the origin of the word tells us that it is the plow that turns at the end of the field. And the dash "-" is all the spaces and cracks where new discoveries can grow. Art and science unfolds in the gap between what we know and what we want to know.

  • Biogasproduktion inom svensk pappers- och massaporduktion : Syntes av möjligheter och begränsningar samt teknisk utvärdering : Bilaga 2 Etablering/effektivisering av biogasproduktion inom svensk pappers- och massaindustri

    • Linköpings Universitet har tillsammans med Pöyry och Scandinavian Biogas Fuels drivit projektet ”Etablering/effektivisering av  biogasproduktion inom svensk pappers- och massaproduktion”. Potentialen hos det organiska materialet i avloppsvatten från svensk pappers- och massaindustri (PMI) till biogasproduktion skattades vid projektstart till 100 milj. Nm3 metan per år (1 TWh). Denna rapport är en syntes av resultaten från projektet med syfte att ge visa hur de genererade resultaten kan omsättas i teknisk praktik med tillhörande ekonomiska insatser. Syftet är att ge underlag och stöd till PMI-branschen och externa intressenter, som överväger att implementera biogasproduktion inom PMI.Substraten för biogasproduktion som återfinns i pappers- och massaindustrins avloppsvatten och slam kännetecknas av stora volymer med låga COD-halter. Detta kräver rötningstekniker, som tillåter mycket korta uppehållstider jämfört med mer traditionellt utformade biogasanläggningar för att inte tankstorleken ska bli för stor. Två tekniker, som utvecklats inom projektet, klarar detta: EGSB (expanded granular sludge bed) och CSTR (completely stirred tank reactor) med slamåterföring. Dessa tekniker har därför utvärderats för tre olika typbruk, ett CTMP-bruk, ett TMP-bruk och ett sulfatmassabruk. Resultaten från dessa experimentella studier är utgångspunkten för i utvärderingen i föreliggande rapport. För varje processkoncept har en grov kostnadsuppskattning (±20 %) gjorts för den investering som krävs för biogasproduktion.En EGSB på ett TMP-bruk med ett totalavlopp på 1500 m3/h, där hela blekeriavloppet från peroxidblekningen och en del av det övriga avloppet behandlas i en 4000 m3 reaktor förväntas ge 2,5 milj Nm3 metan/år. Investeringskostnaden för anläggningen uppskattas till 75 milj. SEK (±20 %).En EGSB på ett CTMP-bruk med ett totalavlopp på 170 m3/h där hela avloppet behandlas i en 3000 m3 reaktor förväntas ge 1,8 milj Nm3 metan/år. Investeringskostnaden för anläggningen uppskattas till 64 milj. SEK (±20%).En CSTR med slamåterföring som körs på bioslam från ett sulfatmassabruk där ett bioslamflöde på 46 m3/h behandlas i en 4000 m3 reaktor förväntas ge 1,0 milj Nm3 metan/år. I denna design är strategin för den aeroba bioreningen ändrad för att producera ett bioslam optimerat för att ge högsta möjliga biogaspotential. Detta innebär produktion av större mängd slam, som i största mån kan rötas till metan, dvs mängd metan per mängd rötat organiskt material samtidigt som COD-reduktionen i vattenreningen bibehålls. Investeringskostnaden för anläggningen uppskattas till 32 milj. SEK (±20%).Baserat på de COD-kvantiteter som når de luftade dammarna inom PMIs vattenreningssystem förbrukas årligen ca 0,8 TWh el. Införande av biogasproduktion i massaindustrins spillvattenrening skulle reducera mängden COD med mellan 30-50%, vilket får till följd att den årliga elförbrukningen i samband med den aeroba reningen går ner med ca 0,2-0,4 TWh. Detta innebär alltså ett energitillskott av 0,9 – 1,1 TWh givet att hela den tillgängliga biogaspotentialen skulle byggas ut. Till detta kommer eventuella vinster relaterade till slamhanteringen.

  • Drawing Lines in the Sand : Organizational Responses to Evaluations in a Swedish University

    • As organizations are increasingly evaluated, evaluative practices are becoming central to organization theory. While highlighting how organizations may respond to evaluations, earlier work on organizations and evaluation has been limited to inter-organizational evaluations, such as third-party rankings or ratings. As a consequence, the prevailing perspective on organizational responses has been developed almost entirely based on an idea that what is being evaluated in the first place – the evaluation entities – is unproblematic. Acknowledging that the definition of the evaluation entity may be ambiguous prior to the evaluation, and that this definition might vary over time, some interesting areas of inquiry emerge in relation to current understandings of organizational responses to evaluations.First, it raises questions of how entities are defined in evaluations, and how this can be seen as an organizational response? Second, it raises questions about our current understanding of the dynamics of organizational responses and how organizational responses shape organizational adaptation and change. Third, it raises question about how entity definitions shape how organizations become evaluated. Despite recent interest in understanding organizational responses to evaluations, organizational scholars have largely refrained from entertaining these questions.The objective of this dissertation is to revisit the assumption of the unambiguously defined evaluation entity in the current theorizing on organizational responses to evaluations. An opportunity to investigate this question emerged when the oldest university in Sweden, Uppsala University, decided to carry out two large evaluations of the research activities within the university – in 2007 and in 2011. The evaluation was made public and aimed to identify and evaluate (partly through a rating) the research achievements of the departments of the university. This presented an opportunity to study organizational responses to evaluations in a setting that is different from what has been studied previously. Based on a study of 44 of the departments within the university my main finding show how entities are defined and how this can be seen as an organizational response to evaluations. My findings also show how entity definitions interact with the more traditional responses taken after an evaluation, and that entity definitions shape the evaluation of the department. This dissertation thus lends support for the idea that apart from responding by changing activities or their organizational units, organizations can also respond to evaluations by the way they shape what will be evaluated – the entities being subject for the evaluation. The findings in this dissertation holds consequences for organizational theory and the evaluation of organizations.
