  • RNAi delivery by exosome-mimetic nanovesicles - Implications for targeting c-Myc in cancer

    • To develop RNA-based therapeutics, it is crucial to create delivery vectors that transport the RNA molecule into the cell cytoplasm. Naturally released exosomes vesicles (also called "Extracellular Vesicles") have been proposed as possible RNAi carriers, but their yield is relatively small in any cell culture system. We have previously generated exosome-mimetic nanovesicles (NV) by serial extrusions of cells through nano-sized filters, which results in 100-times higher yield of extracellular vesicles. We here test 1) whether NV can be loaded with siRNA exogenously and endogenously, 2) whether the siRNA-loaded NV are taken up by recipient cells, and 3) whether the siRNA can induce functional knock-down responses in recipient cells. A siRNA against GFP was first loaded into NV by electroporation, or a c-Myc shRNA was expressed inside of the cells. The NV were efficiently loaded with siRNA with both techniques, were taken up by recipient cells, which resulted in attenuation of target gene expression. In conclusion, our study suggests that exosome-mimetic nanovesicles can be a platform for RNAi delivery to cell cytoplasm.

    • With the increasing availability of collections of antibodies, their evaluation in terms of binding selectivity becomes an important but challenging task. Planar antigen microarrays are very suitable tools to address this task and provide a powerful proteomics platform for the characterization of the binding selectivity of antibodies toward thousands of antigens in parallel. In this chapter, we describe our in-house developed procedures for the generation of high-density planar antigen microarrays with over 21,000 features. We also provide the details of the assay protocol, which we routinely use for the assessment of binding selectivity of the polyclonal antibodies generated within the Human Protein Atlas.

  • Investigation of Humidity Sensor Effect in Silver Nanoparticle Ink Sensors Printed on Paper

    • Thin inkjet-printed tracks of silver nanoparticles have previously been observed to show a non-reversible decrease in resistance when exposed to a high degree of relative humidity and thus providing sensor functionality with a memory effect. This paper provides a more in-depth explanation of the observed humidity sensor effect that originates from inkjet-printed silver nanoparticle sensors on a paper substrate. It is shown that the geometry of the sensor has a large effect on the sensor's initial resistance, and therefore also on the sensor's resistive dynamic range. The importance of the sensor geometry is believed to be due to the amount of solvent from the ink interacting with the coating of the paper substrate, which in turn enables the diffusion of salts from the paper coating into the ink and thus affecting the silver ink.

    • Colour and light have largely been considered as belonging to two different fields of knowledge, having disparate theoretical, terminological and methodological traditions. This creates a ground for misunderstandings and obstructs a fruitful interdisciplinary and interprofessional collaboration. A survey over international research literature from 2006 -2011 shows that there has been only little research on the spatial interaction between colour and light, but that the interest for this area has recently increased. The interdisciplinary Nordic research project SYN-TES: Human colour and light synthesis. Towards a coherent field of knowledge was carried out during 2010-11. Colour and light experts from Nordic universities and companies investigated different aspects of the spatial interaction between colour and light and its importance for human beings. This paper deals with the general learnings from the process. Specific results are presented in other papers at this conference.

    • Powder Bed Fusion-Laser Beam (PBF-LB) processing of magnesium (Mg) alloys is gaining increasing attention due to the possibility of producing complex biodegradable implants for improved healing of large bone defects. However, the understanding of the correlation between the PBF-LB process parameters and the microstructure formed in Mg alloys remains limited. Thus, the purpose of this study was to enhance the understanding of the effect of the PBF-LB process parameters on the microstructure of Mg alloys by investigating the applicability of computational thermodynamic modelling and verifying the results experimentally. Thus, PBF-LB process parameters were optimized for a Mg WE43 alloy (Mg-Y-3.(9wt%)-Nd-3wt%-Zr-0.5wt%) on a commercially available machine. Two sets of process parameters successfully produced sample densities >99.4%. Thermodynamic computations based on the Calphad method were employed to predict the phases present in the processed material. Phases experimentally established for both processing parameters included alpha-Mg, Y2O3, Mg3Nd, Mg24Y5 and hcp-Zr. Phases alpha-Mg, Mg24Y5 and hcp-Zr were also predicted by the calculations. In conclusion, the extent of the applicability of thermodynamic modeling was shown, and the understanding of the correlation between the PBF-LB process parameters and the formed microstructure was enhanced, thus increasing the viability of the PBF-LB process for Mg alloys.

    • Magnesium (Mg) has gained a lot of attention for its biodegradable properties. Additive manufacturing of Mg would further provide an opportunity to manufacture optimized structures for implants. However, much work remains to ensure stable processes and to understand the correlation between processing parameters, microstructure and related properties. The aim of this study was to process gas-atomized WE43 powder by Powder Bed Fusion-Laser Beam (PBF-LB) in a commercially available machine (EOS M290). The built material was characterized in terms of microstructure and composition and compared with the virgin powder. Thermodynamic computations based on the Calphad-method were for the first time employed to an additively manufactured Mg-alloy to predict phases present. Samples with above 99% density were successfully manufactured. The phases predicted using computational thermodynamics partly overlapped with phases observed experimentally. The study confirms the feasibility of using commercial PBF-LB systems for processing Mg-alloys, and the applicability of thermodynamic modelling to this system.

    • Poly(methyl methacrylate) (PMMA) is the most commonly used material for the treatment of osteoporosis-induced vertebral compression fractures. However, its high stiffness may introduce an increased risk of adjacent vertebral fractures post-surgery. One alternative in overcoming this concern is the use of additives. This presents its own challenge in maintaining an adequate biocompatibility when modifying the base cement. The aim of this study was to evaluate the in vivobiocompatibility of linoleic acid (LA)-modified acrylic bone cement using a large animal model for the first time, in order to further advance towards clinical use. A worst-case approach was used, choosing a slow-setting base cement. The in vitro monomer release from the cements was also assessed. Additional material characterization, including mechanical tests, are summarized in Appendix A. Unmodified and LA-modified cements were injected into a total of 56 bone defects created in the femur and humerus of sheep. Histopathologic and histomorphometric analysis indicated that LA-modified cement showed a harmless tissue response similar to that of the unmodified cement. Adjacent bone remodeling was observed microscopically 4 weeks after implantation, suggesting a normal healing process of the bone tissues surrounding the implant. LA-modified cement exhibited lower mechanical properties, with a reduction in the elastic modulus of up to 65%. The handling properties were slightly modified without negatively affecting the injectability of the base cement. LA-modified bone cement showed good biocompatibility as well as bone compliant mechanical properties and may therefore be a promising material for the treatment of osteoporotic vertebral fractures.

  • Evaluation of coatings applied to flexible substrates to enhance quality of ink jet printed silver nano-particle structures

    • Different types of the commercial surface treatment InkAid have been evaluated as a surface treatment to enhance print quality of silver nano-particle ink structures printed on polyimide and polyethene substrate. Originally these coatings have been specified to be applied on substrates for graphical ink jet printing. On the coated polyimide and polyethene substrates lines of different widths have been printed using a Dimatix materials printer together with silver nano-particle ink manufactured by Advanced Nano Products. The prints have then been evaluated in terms of print quality and resistivity before and after sintering. The results show that the application of these coatings can improve the print quality considerably, making it possible to print lines with a good definition, which is not  otherwise possible with this type of ink on this substrate types. It has been found that the coating Semi Gloss provides the best results, both in terms of print quality as well as the lowest resistivity. The resistivity on polyethene is 3.5*10-7Ωm at best when sintered at 150°C and for polyimide  8.9*10-8Ωm sintered at 200°C. This corresponds to a conductivity of about  4.5% and  18%of bulk silver, respectively. It can be concluded that applying such PVP based coatings to polyethene and polyimide will increase the print quality quite substantially, making it possible to print patterns with requirements of smaller line widths and more details than what is possible without coating.

  • The influence of paper coating content on room temperature sintering of silver nanoparticle ink

    • The resistance of inkjet printed lines using a silver nanoparticle based ink can be very dependent on the substrate. A very large difference in resistivity was observed for tracks printed on paper substrates with aluminum oxide based coatings compared to silica based coatings. Silica based coatings are often cationized with polymers using chloride as a counter ion. It is suggested that the precipitation of silver salts is the cause of the high resistivity, since papers pretreated with salt solutions containing ions that precipitate silver salts gave a high resistance. Silver nitrate has a high solubility and paper pretreated with nitrate ions gave a low resistivity without sintering. The results obtained show that, by choosing the correct type of paper substrate, it is possible to manufacture printed structures, such as interconnects on paper, without the need for, or at least to reduce the need for, post-print sintering. This phenomenon is, of course, ink specific. Inks without or with a low silver ion content are not expected to behave in this manner. In some sensor applications, a high resistivity is desired and, by using the correct combination of ink and paper, these types of sensors can be facilitated.

  • Microfluidic analysis of antibody specificity in a compact disk format

    • A new and flexible technology for high throughput analysis of antibody specificity and affinity is presented. The method is based on microfluidics and takes advantage of compact disks (CDs) in which the centrifugal force moves fluids through microstructures containing immobilized metal affinity chromatography columns. Analyses are performed as a sandwich assay, where antigen is captured to the column via a genetically attached His(6)-tag. The antibodies to be analyzed are applied onto the columns. Thereafter, fluorescently labeled secondary antibodies recognize the bound primary antibodies, and detection is carried out by laser-induced fluorescence. The CDs contain 104 microstructures enabling analysis of antibodies against more than 100 different proteins using a single CD. Importantly, through the three- dimensional visualization of the binding patterns in a column it is possible to separate high affinity from low affinity binding. The method presented here is shown to be very sensitive, flexible and reproducible.
