  • Deficiency of SARS-CoV-2 T-cell responses after vaccination in long-term allo-HSCT survivors translates into abated humoral immunity.

    • Recipients of allogeneic hematopoietic stem cell transplantation (allo-HSCT) for hematological diseases are at risk of severe disease and death from COVID-19. To determine the safety and immunogenicity of BNT162b2 and mRNA-1273 COVID-19 vaccines, samples from 50 infection-naive allo-HSCT recipients (median, 92 months from transplantation, range, 7-340 months) and 39 healthy controls were analyzed for serum immunoglobulin G (IgG) against the receptor binding domain (RBD) within spike 1 (S1) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2; anti-RBD-S1 IgG) and for SARS-CoV-2-specific T-cell immunity, reflected by induction of T-cell-derived interferon-γ in whole blood stimulated ex vivo with 15-mer SI-spanning peptides with 11 amino acid overlapS1-spanning peptides. The rate of seroconversion was not significantly lower in allo-transplanted patients than in controls with 24% (12/50) and 6% (3/50) of patients remaining seronegative after the first and second vaccination, respectively. However, 58% of transplanted patients lacked T-cell responses against S1 peptides after 1 vaccination compared with 19% of controls (odds ratio [OR] 0.17; P = .009, Fisher's exact test) with a similar trend after the second vaccination where 28% of patients were devoid of detectable specific T-cell immunity, compared with 6% of controls (OR 0.18; P = .02, Fisher's exact test). Importantly, lack of T-cell reactivity to S1 peptides after vaccination heralded substandard levels (<100 BAU/mL) of anti-RBD-S1 IgG 5 to 6 months after the second vaccine dose (OR 8.2; P = .007, Fisher's exact test). We conclude that although allo-HSCT recipients achieve serum anti-RBD-S1 IgG against SARS-CoV-2 after 2 vaccinations, a deficiency of SARS-CoV-2-specific T-cell immunity may subsequently translate into insufficient humoral responses.

The objectives of the project revolved around technological developments in nucleic acid analysis. The project partners have discovered, created and developed a huge body of insights into nucleic acid analysis, ranging from improvements and implementation of current technologies to the most promising sequencing technologies that constitute a 3(rd) and 4(th) generation of sequencing methods with nanopores and in situ sequencing, respectively.

    • Recent molecular studies have identified substantial fungal diversity in indoor environments. Fungi and fungal particles have been linked to a range of potentially unwanted effects in the built environment, including asthma, decay of building materials, and food spoilage. The study of the built mycobiome is hampered by a number of constraints, one of which is the poor state of the metadata annotation of fungal DNA sequences from the built environment in public databases. In order to enable precise interrogation of such data – for example, “retrieve all fungal sequences recovered from bathrooms” – a workshop was organized at the University of Gothenburg (May 23-24, 2016) to annotate public fungal barcode (ITS) sequences according to the MIxS-Built Environment annotation standard (http://gensc.org/mixs/). The 36 participants assembled a total of 45,488 data points from the published literature, including the addition of 8,430 instances of countries of collection from a total of 83 countries, 5,801 instances of building types, and 3,876 instances of surface-air contaminants. The results were implemented in the UNITE database for molecular identification of fungi (http://unite.ut.ee) and were shared with other online resources. Data obtained from human/animal pathogenic fungi will furthermore be verified on culture based metadata for subsequent inclusion in the ISHAM-ITS database (http://its.mycologylab.org).

  • Sensitive magnetic biodetection using magnetic multi-core nanoparticles and RCA coils

    • We use functionalized iron oxide magnetic multi-core particles of 100 nm in size (hydrodynamic particle diameter) and AC susceptometry (ACS) methods to measure the binding reactions between the magnetic nanoparticles (MNPs) and bio-analyte products produced from DNA segments using the rolling circle amplification (RCA) method. We use sensitive induction detection techniques in order to measure the ACS response. The DNA is amplified via RCA to generate RCA coils with a specific size that is dependent on the amplification time. After about 75 min of amplification we obtain an average RCA coil diameter of about 1 mu m. We determine a theoretical limit of detection (LOD) in the range of 11 attomole (corresponding to an analyte concentration of 55 fM for a sample volume of 200 mu L) from the ACS dynamic response after the MNPs have bound to the RCA coils and the measured ACS readout noise. We also discuss further possible improvements of the LOD.

This article analyzes automotive radar interference and proposes several new approaches that combine industrial and academic expertise toward the goal of achieving interference-free autonomous driving (AD). © IEEE.

    • Intelligent transportation is heavily reliant on radar, which have unique robustness under heavy rain/fog/snow and poor light conditions. With the rapid increase of the number of radars used on modern vehicles, most operating in the same frequency band, the risk of radar interference becomes an important issue. As in radio communication, interference can be mitigated through coordination. We present and evaluate two approaches for radar interference coordination, one for FMCW and one for OFDM, and highlight their challenges and opportunities.

  • Building a generic voxel phantom ofIRINA for Monte Carlo simulations

    • The human phantom IRINA, which is widely used for whole body counting calibrations, has been modelled using MATLAB. This document summa-rizes and explains the procedure that was applied for building voxel ver-sions of IRINA in standing position. All 6 sizes in standing positions were successfully modelled with the help of MATLAB and the files are ready to use for any MC simulation. The MC code GATE was used to verify the geometry of the IRINA phantom by comparing the placement of source tubes and scatterers in the Monte Carlo model to the original IRINA docu-mentation. The methodology can easily be used for building voxel phan-toms of IRINA in sitting and bending position, as well as any other geome-try that may be needed.

    • We have made a comparison between two computational phantoms, modeled from the unified phantom UPh-08T, for whole-body counting applications. One of these was further compared with the ICRP reference adult male computational phantom. The simulations that were performed for the comparison of all three voxel phantoms use various distributions of 60Co. The two voxel phantoms of the UPh-08T showed good agreement, despite different methods of phantom modeling. Also, effects on efficiency of the inhomogeneous distribution of a radionuclide in the computational UPh-08T phantom were studied, using the realistic biodistribution of 140La. Our results show that the activity estimation of radionuclides, which are inhomogeneously distributed in the human body, will be in error if a homogeneous distribution is assumed for the calibration of whole-body counting systems.
