  • Comprehensive analysis to explain reduced or increased SOD1 enzymatic activity in erythrocytes in ALS patients and their relatives

    • Our objective was to in blood samples from 3723 individuals including ALS patients without a coding SOD1 mutation and 372 control individuals characterize stabilities of mutant SOD1s, compare SOD1 enzymatic activities between patients with different genetic causes of ALS, and search for underlying causes of deviant SOD1 activities in individuals lacking SOD1 mutations. Erythrocyte SOD1 enzymatic activities normalized to hemoglobin content were determined. Coding SOD1 sequences were analyzed by Sanger sequencing, copy number variations by fragment length analysis and by TaqMan Assay. Hemoglobin disorders were searched for. Of the 46 SOD1 mutations found, ¾ caused severe destabilization of the mutant protein but in ¼ SOD1 was essentially physically stable. Mutations producing structural changes all caused halved SOD activities. There were no differences in SOD1 activities between controls and patients without any detected SOD1 mutations or patients with C9ORF72HRE or TBK1 mutations. In the low and high SOD1 activity groups no deviations were found in exon copy numbers and intron gross structures. Also, no uncommon variants in exon-flanking sequences were detected. Thalassemias and iron deficiency anemia were associated with increased SOD1 activity/hemoglobin ratios. In conclusion, adjunct erythrocyte SOD activity analysis is of value to signal the presence of exon and splice-site-intron mutations that influence the SOD1 structure.

  • Siliac-acid and incidence of hospitalization for diabetes and its complications during 40-years of follow-up in a large cohort : the Värmland survey

    • Aim To examine the association of sialic acid (SA) with first recorded diabetes mellitus-related hospitalization.Methods From a population-based study in Värmland, Sweden, between 1962 and 1965, 87,035 men and women were selected and followed for first recorded diabetesrelated hospitalization until 2005. The association of SA was calculated and stratified for gender by Cox´s proportional hazards models. Adjustments were made for conventional risk factors and socioeconomic status. Association analyses were made for comparisons between SA-levels above and below median.Results The mean age was 47.2 (SD 13.0) years and the total numbers of incident diabetes-related hospitalizations in men and women were 3445 and 3273, respectively. Hazard ratios per one standard deviation of SA were 1.12 (95% CI: 1.08 to 1.17, p<0.0001) in men and 1.17 (95% CI: 1.13 to 1.22, p<0.0001) in women. Interaction analyses indicated a relatively higher SA-associated risk in women than in men with above median SA levels.Conclusions In this large population-based cohort followed for more than 40 years, elevated SA, as a marker of systemic inflammation, was independently associated with risk of diabetes and diabetes-related hospitalizations.

  • Operando XPS and SXRD study of the Fischer-Tropsch reaction on flat and stepped Co at up to around 1 bar

    • The Fischer-Tropsch catalytic reaction on Co has still several unknows regarding the surface chemical state of the catalyst and presence of various adsorbed intermediates appearing under operando conditions. Here, we report an operando X-ray photoelectron spectroscopy study of the flat Co(0001) and stepped Co(10-14) model catalyst surfaces during Fischer-Tropsch synthesis conditions at 150 mbar – 975 mbar pressure and 406 K – 548 K temperature in a H2/CO gas mixture using X-ray photoelectron spectroscopy as well surface X-ray diffraction. We observe that the Co surface remains metallic at all conditions with only adsorbed species. There are a number of different intermediates on the surface such as adsorbed CO in only on-top position, C/-CH intermediates, -CH2- species and various longer hydrocarbon molecules. The coverage of the hydrocarbon species decreases with increasing temperature but increase strongly with pressure. The stepped surface shows a significant increase in adsorbed hydrocarbon species which is a clear indication of higher reactivity. There is no observation of any CHO or CH2O species indicating a direct CO dissociation pathway and that to our observation only hydrocarbon species participate in the chain growth. The relative high coverage of CO that is almost constant for the various conditions points to that CO dissociation is one of the rate-limiting steps. The rapid increase in the coverage of hydrocarbon species at high pressure conditions and on the stepped surface further indicates that the final hydrogen induced termination step is also rate-limiting. By following the time dependence of the C 1s region with rapid change in the flow of reactants, we demonstrate that the surface species are highly dynamic resulting in delays in reaching steady-state conditions.

  • Validating the selectivity of antibodies used in multiplexed serum profiling via parallel immunocapture analysis

    • The increasing availability of antibodies towards human proteins drives the generation of new and exploratory data, which can be generated by profiling e.g. plasma samples using multiplexed arrays. However, antibody assays can lead to erroneous results due to cross-­‐reactivity to off-­‐targets. Here, we describe an approach in which an antibody-­‐based suspension bead array is combined with subsequent validation of on-­‐ target binding using a coupled affinity purification assay. Based on differential heat treatment of samples, antibody performance was investigated and the results for antibodies directed towards several complement factors provide insights on the detection of proteins in sera from patients with complement deficiency. In conclusion, the combined parallel flow and blot based immunocapture analysis serves an important, first line tool for resolving differently detected serum or plasma protein profiles proposed by antibody arrays.

  • Plasma levels of carnosine dipeptidase 1 decrease in prostate cancer patients with lymph node metastasis

    • There is a need for a better differentiation of aggressive tumors in prostate cancer to design a tailored treatment for each patient, preferably by a minimally invasive analysis of blood samples. In a previous study, we discovered a decrease of plasma levels of carnosine dipeptidase 1 (CNDP1) in association with aggressive prostate cancer. Now this relation has been investigated and characterized further by generating several new antibodies for extended analysis of CNDP1 in plasma. Multi-antibody sandwich assays were developed and applied to 1,214 samples from two Swedish cohorts that confirmed decreased levels of CNDP1 in plasma for patients with advanced disease. Therein, CNDP1 assays revealed superior differentiation for tumor N stages than clinical tPSA. Further investigations can now elucidate mechanisms behind decreasing levels of CNDP1 in plasma and primary in regards to lymph node metastasis.
