  • Mast cells express functional CD30 ligand and are the predominant CD30L-positive cells in Hodgkin's disease

    • Hodgkin's disease (HD) tumours are characterized by the presence of few tumour cells, the Hodgkin and Reed-Sternberg (HRS) cells, surrounded by a large amount of non-neoplastic cells. The role of this cell infiltrate for the development of HD is not known. CD30, belonging to the tumour necrosis factor receptor superfamily, is highly expressed on HRS cells and believed to be involved in tumourigenesis and tumour progression. Tumour samples from 42 patients were immunohistochemically double-stained for tryptase, a mast cell-specific proteinase and CD30 ligand (CD30L). Tryptase-positive mast cells were present in all tumours. Of these cells, 50% expressed CD30L and 66% of the CD30L-positive cells were mast cells. CD30L mRNA in in vitro developed normal mast cells and malignant human and murine mast cell lines was detected using reverse transcription polymerase chain reaction. CD30L protein expressed on human mast cells was detected using flow cytometry. In a co-culture assay, the human mast cell line HMC-1 stimulated thymidine uptake in HRS cell lines, and the stimulation could be blocked using CD30L-specific monoclonal antibodies. In conclusion, mast cells are present in HD tumours and are the predominant CD30L-expressing cells. CD30L-CD30 interaction is a pathway by which mast cells may stimulate DNA synthesis in HRS cells.

  • Selective determination of organophosphate flame-retardants and plasticizers in indoor air by gas chromatography, positive-ion chemical ionization and collision-induced dissosiation mass spectrometry

    • Gas chromatography/ion trap mass spectrometry with in-source ionization and dissociation was used in positive-ion chemical ionization (PICI) mode for the determination of organophosphate triesters in indoor air. These compounds are widely used as additive flame retardants and plasticizers in different types of materials and have become ubiquitous pollutants in indoor environments. When using collision-induced dissociation in PICI mode the fragmentation of the organophosphate triesters can be performed in a more controllable way than in electron ionization (EI) mode. The developed selected-reaction monitoring method provided high selectivity for the investigated compounds. For 8-h air measurements (corresponding to 1.5 m3 of sampled air) the limit of detection of the method was determined to be in the range 0.1–1.4 ng m−3, which is comparable with nitrogen-phosphorus detection and about 50-fold lower than when using EI in selected-ion monitoring mode. The presented method was applied to samples from three common indoor environments, in which a number of organophosphate triesters were identified and quantified. The dominating compound was found to be tris(2-chloropropyl) phosphate, which occurred at levels up to 0.8 μg m−3.

  • Neoadjuvant, adjuvant and palliative treatment of gastrointestinal stromal tumours (GIST) with imatinib: a centre-based study of 17 patients.

    • Malignant gastrointestinal stromal tumours (GIST) have a poor prognosis. Since these tumours are resistant to conventional radiation and chemotherapy, surgery has been the mainstay of treatment. However, surgery is usually inadequate for the treatment of malignant GIST. Imatinib, a KIT tyrosine kinase inhibitor, has recently been found to have a dramatic antitumour effect on GIST. In this centre-based study of 17 consecutive patients with high-risk or overtly malignant GIST, imatinib was used in three different settings - palliatively, adjuvantly, and neoadjuvantly. The treatment was found to be safe and particularly effective in tumours with activating mutations of exon 11 of the KIT gene. Clinical response to imatinib treatment correlated morphologically to tumour necrosis, hyalinisation, and reduced proliferative activity. The value of neoadjuvant imatinib treatment was illustrated in one case.

  • Speciation of heavy metals in road runoff and roadside total deposition

    • The concentrations of Cd, Co, Cu, Pb, W and Zn were measured in road runoff and total deposition at two Swedish field sites during one year. It was found that the concentrations of most elements increased significantly during the winter, up to one order of magnitude. For cobalt and tungsten, it was found that around 90% of the total mass transport occurred during the winter, whereas for Cu, Pb, Cd, Zn and Na, the corresponding figures were 70–90, 40–80, 60–90, 50–70 and >99% depending on site specific conditions. The deicing salts (rock salts) did not significantly contribute to the increase in trace element concentrations. Instead, the increased concentrations were due to more intense wearing of the pavement during the winter because of the use of studded tires in combination with the chemical effects caused by the use of deicing salts. New potential elemental markers for roads and traffic are suggested.

  • Influence of ethyl acetate production and ploidy on the anti-mould activity of Pichia anomala

    • A diploid and a haploid strain of Pichia anomala were tested for their biocontrol ability against the spoilage mould Penicillium roqueforti in glass tubes filled with grain at two water activities (a(w)). At a(w) 0.98, the two yeast strains grew and inhibited mould growth equally well and showed similar patterns of ethyl acetate production, reaching maximum values of 10-14 mug ml(-1) headspace. At a(w) 0.95, both growth and biocontrol performance of the haploid strain were reduced. Ethyl acetate formation was also substantially reduced, with maximum headspace concentrations of 4 mug ml(-1). We conclude that ethyl acetate is a major component of the anti-mould activity. The inhibitory effect of ethyl acetate was confirmed in a bioassay where the pure compound reduced biomass production of P. roqueforti.

    • As an inexpensive, simple, and low-solvent consuming extraction technique, the suitability of solid-phase microextraction (SPME) with polydimethylsiloxane (PDMS) sorbent was investigated as a quantitative method for sampling gaseous organophosphate triesters in air. These compounds have become ubiquitous in indoor air, because of their widespread use as additive flame retardants/plasticizers in various indoor materials. Results obtained by sampling these compounds at controlled air concentrations using SPME and active sampling on glass fibre filters were compared to evaluate the method. A constant linear airflow of 10 cm s–1 over the fibres was applied to increase the extraction rate. For extraction of triethyl phosphate with a 100-m PDMS fibre, equilibrium was achieved after 8 h. The limit of detection was determined to be less than 10 pg m–3. The PDMS–air partition coefficients, Kfs, for the individual organophosphate triesters were determined to be in the range 5–60×106 at room temperature (22–23°C). Air measurements were performed utilising the determined coefficients for quantification. In samples taken from a lecture room four different airborne organophosphate esters were identified, the most abundant of which was tris(chloropropyl) phosphate, at the comparatively high level of 1.1 g m–3. The results from SPME and active sampling had comparable repeatability (RSD less than 17%), and the determined concentrations were also similar. The results suggest that the investigated compounds were almost entirely associated with the gaseous phase at the time and place sampled.

  • Inflammatory mediators expressed in human islets of Langerhans : implications for islet transplantation

    •   Expression of immune modulating mediators in human Islets of Langerhans could have important implications for development of autoimmunity in type 1 diabetes and influence the outcome of clinical islet transplantation. Islets obtained from five donors were analyzed at various times after isolation using cDNA array technology. The Atlas Human Cytokine/Receptor and Hematology/Immunology nylon membranes representing 268 genes and 406, respectively, were used and the relative expression of each gene analyzed. Of the 51 gene products identified, high mRNA expression of MCP-1, MIF, VEGF, and thymosin beta-10 was detected in all islet samples. IL-8, IL-1-beta, IL-5R, and INF-gamma antagonist were expressed in islets cultured for 2 days. IL-2R was expressed in islets cultured for more than 6 days. In conclusion, several inflammatory mediators were expressed in isolated islets, particularly at an early stage after isolation, indicating that a few days of culture could be beneficial for the outcome of islet transplantation.
