    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

  • Comparing 18F-AV-1451 with CSF t-tau and p-tau for diagnosis of Alzheimer disease

    • To compare PET imaging of tau pathology with CSF measurements (total tau [t-tau] and phosphorylated tau [p-tau]) in terms of diagnostic performance for Alzheimer disease (AD).We compared t-tau and p-tau and 18F-AV-1451 in 30 controls, 14 patients with prodromal AD, and 39 patients with Alzheimer dementia, recruited from the Swedish BioFINDER study. All patients with AD (prodromal and dementia) were screened for amyloid positivity using CSF β-amyloid 42. Retention of 18F-AV-1451 was measured in a priori specified regions, selected for known associations with tau pathology in AD.Retention of 18F-AV-1451 was markedly elevated in Alzheimer dementia and moderately elevated in prodromal AD. CSF t-tau and p-tau was increased to similar levels in both AD dementia and prodromal AD. 18F-AV-1451 had very good diagnostic performance for Alzheimer dementia (area under the receiver operating characteristic curve [AUROC] ∼1.000), and was significantly better than t-tau (0.876), p-tau (0.890), hippocampal volume (0.824), and temporal cortical thickness (0.860). For prodromal AD, there were no significant AUROC differences between CSF tau and 18F-AV-1451 measures (0.836-0.939), but MRI measures had lower AUROCs (0.652-0.769).CSF tau and 18F-AV-1451 have equal performance in early clinical stages of AD, but 18F-AV-1451 is superior in the dementia stage, and exhibits close to perfect diagnostic performance for mild to moderate AD.This study provides Class III evidence that CSF tau and 18F-AV-1451 PET have similar performance in identifying early AD, and that 18F-AV-1451 PET is superior to CSF tau in identifying mild to moderate AD.

  • A matter of cultural distinction: News consumption in the online media landscape

    • The world of online news is a world where news consumers must make choices among a plethora of different news sources. Previous research points towards a fragmentation of news consumption across the citizenry. However, not enough attention has been paid to class, in particular cultural capital, and how it shapes how groups in society develop preferences for different categories of online news. Drawing upon a representative national survey in Sweden (N=11,108), a country historically known for its egalitarian news consumption, we show that cultural capital engenders patterns of taste and distaste for different online national news providers. This is manifested in that those rich in cultural capital are more inclined to consume ‘quality’ news and to neglect ‘popular’ news. A relative lack of cultural capital is associated with a somewhat reverse pattern. News consumption in the online media landscape is a matter of cultural distinction.

  • Boardroom empires? A study of ownership influence in the Swedish press

    • The question of how ownership impacts on the performance of the press is a perennial one. Despite the extensive attention devoted to the perceived consequences of ownership, information about what newspaper owners actually do remains limited. This article discusses an alternative path for research on media ownership. It involves the board of directors, the main agency that is expected to exercise ownership power. The article presents a study that focuses specifically on the decision-making in and around the boardrooms of three Swedish newspaper firms between 1955 and 2005. Building on analyses of board meeting minutes and interviews with board members, the study shows that the impact of ownership is established, and develops, in an intricate web of interacting forces, internal and external to the firm. It thus underlines the need for both a multi-theoretical foundation and a multi-method design in research on ownership influence in the media.

    • En av de viktigaste strukturella förändringarna på den svenska dagspressmarknaden under 2000-talet är den tilltagande domi- nansen av stiftelseägda tidningskoncerner. Tillsammans med Danmark, där flera stora tidningar också ägs av stiftelser (fonder), hör Sverige därmed till de länder där stiftelseägandet inom dags- pressen är mest utbrett. I den här artikeln analyseras konsekven- serna av denna internationellt sett ovanliga ägarform. Artikeln presenterar en konceptuell ram för hur stiftelseägandet i svensk press ska förstås. I fokus står frågan om ägarinflytande – över den ekonomiska verksamheten i tidningsföretagen och den redaktio- nella. Analysen baseras på en historisk komparativ fallstudie av tre medelstora svenska stiftelseägda dagstidningar.

  • Fading support for the Swedish press support

    • The current recession of the printed press highlights the role of the state in relations to the media. This article analyses the public opinion on the Swedish press support – one of the oldest and most extensive press subsidy systems. Building on theories concerning the factors shaping public opinion in political issues, the article shows that the press support has increasingly evolved into a non-issue with the Swedish populace. Two explanations appear paramount. Firstly, the recent transfer of several supported newspapers from party to private ownership has been accompanied by a demobilization of the public opinion on the support on the basis of party preference. Secondly, the decreasing consumption of newspapers in general has been followed by increasing levels of indifference concerning the support to struggling newspapers. The findings provide yet another indicator that the printed press is gradually losing its long-time position as one of the keystones of civil society.
